[Expression of VEGF and tumor angiogenesis in bladder cancer].

OBJECTIVE: To evaluate expression of vascular endothelial growth factor (VEGF) and its relation to angiogenesis in bladder cancer. METHODS: The expression of VEGF by immunohistochemical staining was examined in 68 cases of primary transitional carcinoma (TCC) and 7 subjects with normal urothelium. Microvessel density (MVD) in 40 cases of invasive bladder cancer was determined by Factor VIII immunohistochemical staining. RESULTS: The expression of VEGF was negative or low level in all normal bladder tissue but high in bladder cancer. VEGF and MVD were closely related to tumor cell nuclear grade and clinical stage. The average MVD was higher in the cases with the high level expression of VEGF, than in those with the low level expression of VEGF. CONCLUSIONS: This study indicates that VEGF is positively correlated with the occurrence and progression of bladder cancer. VEGF is one of the primary angiogenic factors for bladder cancer, and is able to induce tumor angiogenesis and accelerate tumor growth. Quantification of VEGF may provide a valuable marker for a poorer prognosis of patients.

Role of p53 gene mutations in human esophageal carcinogenesis: results from immunohistochemical and mutation analyses of carcinomas and nearby non-cancerous lesions.

In order to characterize p53 alterations in esophageal cancer and to study their roles in carcinogenesis, we performed gene mutation and immunohistochemical analysis on 43 surgically resected human esophageal specimens, which contain squamous cell carcinoma (SCC) and adjacent non-cancerous lesions, from a high-incidence area of Linzhou in Henan, China. A newly developed immunohisto-selective sequencing (IHSS) method was used to enrich the p53 immunostain-positive cells for mutation analysis. p53 gene mutations were detected in 30 out of 43 (70%) SCC cases. Among 29 SCC cases that were stained positive for p53 protein, 25 (86%) were found to contain p53 mutations. In five cases of SCC with homogeneous p53 staining, the same mutation was observed in samples taken from four different positions of each tumor. In a well differentiated cancer nest, p53 mutation was detected in only the peripheral p53-positive cells. In tumor areas with heterogeneous p53 staining, either the area stained positive for p53 had an additional mutation to the negatively stained area or both areas lacked any detectable p53 mutation. In the p53-positive non-cancerous lesions adjacent to cancer, p53 mutations were detected in seven out of 16 (47%) samples with basal cell hyperplasia (BCH), eight out of 12 (67%) samples with dysplasia (DYS), and six out of seven (86%) samples with carcinoma in situ (CIS). All mutations found in lesions with DYS and CIS were the same as those in the nearby SCC. In seven cases of BCH containing mutations, only three had the same mutations as the nearby SCC. The results suggest that p53 mutation is an early event in esophageal carcinogenesis occurring in most of the DYS and CIS lesions, and cells with such mutations will progress to carcinoma, whereas the role of p53 mutations in BCH is less clear.

[The effect of combined endocrine therapy on prostate and testis].

OBJECTIVE: To study the pathological changes of the effect of combined endocrine therapy on normal prostate, benign prostatic hyperplasia, prostatic adenocarcinoma and testis. METHODS: 11 radical prostatectomy specimens, 3 core-needle biopsies of prostate and 3 testes obtained after at least 3 months of enatone-flutamide inhibition therapy were studied. Step-section was performed on radical specimens and average 16 sections per case were reviewed. PSA, PSAP and AE1/AE3 were immunostained on 14 cases of prostate cancer and a comparative study pre- and post-treatment was made. RESULTS: No residual tumor was recognized in 2 cases. Such characteristic changes were found in 9 cases as prominent acinar atrophy, decreased ratio of acini to stroma, stromal fibrosis, squamous metaplasia of carcinoma, cytoplasmic vaculation, nuclear shrinkage, and nucleolar shrinkage. No apparent change was discovered in 3 cases. Secretory epithelial atrophy and basal cell hyperplasia were the popular change in BPH and normal prostate. The expressions of both PSA and PSAP were markedly reduced in prostate carcinoma and nonneoplastic glands. Pathological downstaging of the tumor was not found statistically. Epithelial atrophy also existed in seminal vesicles and Leydig cells of the testes after hormone therapy. CONCLUSIONS: Combined endocrine therapy results in histologically distinctive changes that can be found in both nonneoplastic and neoplastic prostate tissue. However, the drugs can not eradicate prostate cancer completely. Testis atrophy is the direct action of the therapy.

[The detection of high risk human papillomaviruses in papillary transitional cell carcinoma of urinary bladder].

OBJECTIVE: To investigate the histological evidence of high risk human papillomavirus infection in bladder cancer. METHODS: 52 cases of papillary transitional cell carcinoma of the urinary bladder were studied for the incorporation of high risk human papillomavirus type 16 and type 18 DNA by non-isotopic in situ hybridization (NISH). RESULTS: 19 of 52 cases (36.5%) were positive for HPV DNA with punctuate and/or diffuse purple-blue staining in the nucleus of cancer cells, including two cases positive both for HPV16/18. HPV DNA were predominantly expressed in superficial tumors. Among these positive tumors, adjacent normal epithelium and Brunn's nests also showed positive staining. Morphologically, HPV positive cells existed in the whole layer of papillary tumor cells, 3 of them presented koilocytoid change. CONCLUSIONS: HPV infection might be a common event in urinary bladder and a potential factor for carcinogenesis of bladder cells.

[Clinicopathologic study of 11 patients with Paget's disease of scrotum].

OBJECTIVE: To probe the relationship of clinicopathologic presentations and prognosis of Paget's disease primarily involving the scrotum. METHODS: 11 cases of scrotal Paget's disease were studied retrospectively from 1984 to 1997. PAS was stained and EMA, S-100, HMB45, CEA and CA15-3 were immunostained in all cases for the differential diagnosis from Bowen's disease and malignant melanoma. RESULTS: Six of 11 cases involved hair follicle and/or sweat glands, and 3 cases involved dermis (one had metastasis of left superficial and deep inguinal lymph nodes, and left internal iliac lymph nodes). Paget's cells were positive for PAS (11/11), EMA (11/11), CEA (6/11), CA15-3 (11/11), and negative for S-100, HMB45. Wide local excision of the lesions was performed in 10 of 11 patients both for primary disease and recurrent disease. 3 of the 10 patients underwent lymph nodes resection and metastasis was found in 1 case pathologically. One patient accepted chemotherapy and died, from respiratory insufficiency after one month. The other 10 patients were followed up for 6 to 127 months and no patient died of Paget's disease. CONCLUSIONS: Involvement of hair follicles and sweat glands by Paget's cell does not influence the prognosis.However, invasion of dermis by Paget's cells may suggest possible metastasis.

[Polymorphism distribution of ST14(DXS52) VNTR in normal individuals in northeastern region of China and its application in gene diagnosis of hemophilia A].

OBJECTIVE: To find out the polymorphism distribution of St14(DXS52) variable number of tandem repeat (VNTR) in normal individuals in the northeastern region of China and hence provide a proof for the gene diagnosis of hemophilia A. METHODS: 60 unrelated individuals (male 12, female 48) in the northeastern region of China were detected using PCR method. RESULTS: 8 allelic fragments detected were 0.7kb(A), 1. 3kb(B), 1.39kb(C), 1.57kb(D), 1.63kb(E), 1.69kb(F), 2.1kb(G), 2. 4kb(H) long in turn. Their frequencies were A 0.38, B0.046, C0.232, E0.111, F0.130, G0.009, H0.009 respectively. Polymorphism information contents (PIC) was 76.36%. Gene diagnosis of 3 hemophilia A patients was performed using this VNTR polymorphism. In one family a female was determined to be a normal individual, not a carrier; 2 male fetus patients were detected in other 2 families. CONCLUSION: St14 is a valuable polymorphism marker for gene diagnosis of hemophilia A.

Quox 1 homeobox protein is expressed in postmitotic sensory neurons of dorsal root ganglia

The expression of vertebrate homeoproteins has been extensively studied in a variety of normal and cancerous tissues, but little is known on the role of vertebrate homeoproteins in the proliferation and differentiation of cells from these tissues. In the present study, we investigate the relationship between Quox 1 protein (a quail homeodomain containing protein) expression and the proliferation and differentiation of quail dorsal root ganglia (DRG) and neural crest cells. In vivo [3H]TdR labeling experiments demonstrate that the postmitotic sensory neuroblasts appear before the formation of the ganglion, and that more than half of sensory neuroblasts from DRG have already terminated their proliferation in embryos of 2 days of incubation (E2). All DRG neurons have completely ceased to proliferate from E6.5 onwards. By means of immunocytochemistry, we observe that Quox 1 protein is accumulated exclusively in all bipolar neurons in culture of DRG from E9-E11, and in all postmitotic sensory-like neuroblasts during in vitro cell differentiation of the neural crest. The Quox 1 immunoreactive neurons express simultaneously neurofilaments or substance P, and they are never labeled by anti-bromodeoxyuridine. These observations together with the morphology of Quox 1 positive cells, demonstrate that Quox1 protein is expressed in the postmitotic sensory neurons of DRG. Our previous experiments have shown that between E4 and E6, the accumulation of Quox 1 protein increases in DRG in vivo, but decreases in the central nervous system in which cell proliferation decreases (Xue et al., (1993) Mech. Dev. 43, 149-158). Taken together, our results show that the accumulation of Quox 1 protein in DRG is tightly linked to the increase in the number of postmitotic neurons, whereas in the central nervous system the level of expression of Quox 1 seems concomitant with the extent of cell proliferation.

[Primary epididymal tumors].

OBJECTIVE: To improve the diagnosis of primary epididymal tumors. METHOD: In 23 primary epididymal tumors (22 benign and 1 malignant), 14 were adenomatoid tumors, 6 were leiomyomas, 1 was multiple fibrous pseudomonas and 1 was sclerosing hemangioma. Epididymal adenocarcinoma was seen in a 57-year-old man. RESULT: Epididymal tumors might be easily misdiagnosed as epididymal tuberculosis, chronic epididymis and spermatocele. Apart from certain benign clinical characteristics, benign epididymal tumors could be diagnosed by B-ultrasonography and aspiration biopsy. Most malignant tumors of epididymis presented as rapidly growing scrotal masses and cured by the removal of tumors or the whole epididymis of the same side. 19 of 22 cases were free from recurrence. CONCLUSION: Malignant epididymal tumors should be treated as malignant tumors of testis and their prognosis is extremely poor.

[The use of laparoscopy in urology: report of 141 cases].

OBJECTIVE: To modify laparoscopy in urology. METHOD: From May 1992 to Nov. 1996, 141 patients (110 males and 31 females) were subjected to laparoscopy in the treatment of urological diseases. The age of the patients ranged from 9 to 76 years (mean 39 years). Five patients had nephrectomy, 12 adrenalectomy, 48 renal cyst resection, 61 high ligation of internal spermatic vein, 5 pelvic staging lymphadenectomy, 9 orchiectomy, and 1 removal of bladder diverticulum. RESULT: The rate of successful operation was 95.7% and that of complications was 2.22%. The mean hospitalization was 4.9 +/- 4.1 days. CONCLUSION: Laparoscopy can be used widely in clinical practice.

[Alpha1-adrenoceptor subtypes in rabbit penile corpus cavernosum].

OBJECTIVE: To study alpha(1)- adrenoceptor (alpha(1)- AR) subtypes with the functional role and the distribution of its subtypes in rabbit penile corpus cavernosum. METHOD: The rabbit penile corpus cavernosum was studied by using functional experiment and radioligand binding assay. RESULT: In the radioligand binding assay, the maximal binding capacity (Bmax of (125)IBE and alpha(1)-AR) decreased obviously after pretreatment with chloroethylclonidine (CEC), which were 746 +/- 236 fmol/mg protein and 214 +/- 71 fmol/mg protein, respectively. Alpha(1A)-, alpha(1D)-and alpha(1B)-AR took about 20%, 25% and 55%, respectively. In the functional experiments, the maximal contraction induced by norepinephrine (NE) was not decreased significantly after pretreatment with CEC. The pA(2) values of two alpha(1)-AR selective antagonists, 5-MU, BMY7378, for antagonizing NE-induced penile corpus cavernosum contraction had a high correlative association with the pK(I) values for cloned alpha(1A)-AR. CONCLUSION: The functional alpha(1)-AR subtype is alpha(1A)-AR although there are three subtypes in the penile corpus cavernosum.

NNF1 is an essential yeast gene required for proper spindle orientation, nucleolar and nuclear envelope structure and mRNA export.

The nuclear envelope is central to nuclear structure and function. It plays a role in maintaining nuclear shape, allowing the exchange of macromolecules between the nucleus and the cytoplasm (via the nuclear pore complexes), and providing attachment sites for microtubules during chromosome segregation and nuclear migration (via the spindle pole body). We have isolated an essential yeast gene, NNF1 that is required for a number of nuclear functions. Cells depleted of Nnf1p or containing a temperature-sensitive nnf1 mutation have elongated microtubules and become bi- and multinucleate. They also have a fragmented nucleolous and accumulate poly(A)+ RNA inside the nucleus. A similar constellation of phenotypes has been reported in cells carrying mutations in a number of nuclear pore proteins, components of the Ran GTPase cycle, and the nuclear localization sequence receptor protein. Our results suggest that Nnf1p plays a role in a number of nuclear functions.

[Influence of fibrin glue on biliointestinal anastomotic scar formation].

Conventional suture technique in the reconstruction of the bile duct often result in anastomotic stenosis and eventual obstructive jaundice and biliary cirrhosis. To overcome the aforementioned drawbacks, fibrin glue (FG) was used instead in the biliointestinal anastomosis where indwelling stent was inserted to complete the experimental model. The experiment consisted of pathological study and ECVD evaluation of anastomotic specimens in order to follow up the whole process of anastomotic wound healing. The experimental studies dis-closed that FG promotes wound healing, limits scar formation, and accelerates scar softening and maturnation. FG plus 6-month indwelling stent can guarantee scar maturation. It is an effective method in the prevention of bile duct stricture.

[Circadian variations of plasma SOD and MDA in health subjects].

Oxygen derived free radical system has been to be implicated in the pathogenesis of many diseases, such as cardiovascular disease, tumor, trauma and radiation injury. Many studies have suggested that oxygen free radical initiate a series of events that result in cell membrane alterations leading to the development of cell injury. The objective of the present study was to examine the circadian characteristics of some parameters of this system in the blood. One parameter was superoxide dismutase (SOD), a major oxygen free radical scavenger; the other was malondialdehyde (MDA), an important product of lipid peroxidation initiated by oxygen free radicals. Blood samples were collected at 6 h intervals beginning at 08:00 over a 24 h span in nine healthy volunteers (5 females, 4 males, average age 30 +/- 5 years). All participants were synchronized for one week with diurnal activity from 07:00 to 23:00 and nocturnal rest, with three meals served at 07:30, 12:00 and 19:00. Plasma SOD, MDA levels were measured by pyrogallol autoxidation assay, thiobarbituric acid spectrophotometry respectively, and the data were fitted with a 24 h cosine curve and analyzed further by a population-mean cosinor. The results showed that the plasma SOD levels exhibited a highly significant circadian rhythm (P < 0.001) in healthy subjects, with acrophase (phi) at 21:17, mesor (M) of 3975.3 +/- 680.5 u/g Hb, and amplitude (A) of 273.62 u/g Hb. A circadian rhythm of plasma MDA was also detected (P < 0.01), phi = 15:15, M = 4.23 +/- 0.86 mumol/L, A = 2.37 mumol/L). The evaluation of circadian natures of SOD and MDA may be important in the prevention, diagnosis and treatment of associated diseases.

[Motion control system in BJ14 medical electron accelerator].

A dual computer based control mode was used in 14 Mev medical electron accelerator. The special non-linear velocity transformation method makes the positioning operation very fast and convenient. The independent driving mode of four blades are suitable to clinical uses. A multi-interlock function provides a safe environment for user's operation.

[The effect of combining intra-aortic balloon pumping with tianma injection on myocardial ischemic-reperfusional injury in goats].

The objective of this study was to explore the effect of combined Intra-Aortic Balloon Pumping (I-ABP) and Tianma Injection on necrotic area in goat myocardial ischemic-reperfusional models which were established by two hours ischemia and a follow-up reperfusion. The results showed that the combined effect of IABP and Tianma Injection was more significant than that of IABP or Tianma separately. It is suggested that IABP and Tianma Injection have synergetic effect on the limitation of necrotic area, which possibly results from their mechanisms for decreasing the size of infarct.

[Relationship between variations of temperature and humidity with reproduction of medical meloids].

This paper reports the relationship between variations of temperature and humidity with reproduction of two medical meloids (Mylabris phalerata, M. cichorii). The results show: for the copulation of two meloids, the suitable temperature is 22-28 degrees C, humidity 26-90%; for the suitable temperature and humidity of their oviposition, M. phalerata 26-28 degrees C and 60-68%, and M. cichorii 23-26 degrees C and 72-78%, respectively; for incubation, M. phalerata is 26-28 degrees C and 70-75% respectively, and M. cichorii must undergo the high and lower variation of temperature, the efficacious accumulative temperature must get of 1680.85 day. degrees C.

Yeast mutants that produce a novel type of ascus containing asci instead of spores.

Tetraploid yeast cells lacking BFR1 or overexpressing an essential gene BBP1 produce a novel type of ascus that contains asci instead of spores. We show here that the asci within an ascus likely arise because a/alpha spores undergo a second round of meiosis. Cells depleted of Bbp1p or lacking Bfr1p are defective in a number of processes such as nuclear segregation, bud formation, cytokinesis and nuclear spindle formation. Furthermore, deletion of BFR1 or overexpression of BBP1 leads to an increase in cell ploidy, indicating that Bfr1p and Bbp1p play roles in both the mitotic cell cycle and meiosis. Bfr1p and Bbp1p interact with each other in a two hybrid assay, further suggesting that they might form a complex important for cell cycle coordination.
