The antibiotics congocidine and distamycin A inhibited the replication of Shope fibroma and vaccinia viruses in BSC(1) cells. Treatment of infected cultures with congocidine for a 12-hr period resulted in an irreversible inhibitory effect on virus development. A longer period of treatment with the antibiotic also resulted in inhibition of the residual infectious virions present in the treated cells. Distamycin A inhibited the replication of poxviruses, but its inhibitory effect was reversed when the antibiotic was removed from the infected cells even as long as 24 hr after treatment. The inhibitory effect of distamycin A and its reversibility resembled the antipoxvirus activity of rifampin.

Establishment of selective immunity, local or systemic, made it possible to evaluate the pathogenesis of Newcastle disease virus (NDV) in the respiratory tract of chickens that were previously immunized with beta-propiolactone-inactivated antigen. NDV was inoculated intranasally or intramuscularly to chickens in different states of immunity (local or systemic). Humoral antibodies protected chickens against intranasal as well as intramuscular infection. Local antibodies, on the other hand, conferred immunity only against intranasal challenge. The respiratory tract supported multiplication of the virus, producing a self-limited subclinical infection. Replication of the virus in this system was negligible, playing only a minor role in the pathogenesis of the disease.
Serial passage of two lentogenic Newcastle disease virus (NDV) strains in kidney cell lines increased virulence and changed viral biological properties. In two cell lines (BHK and MDBK), elevation in virulence was demonstrated by plaque formation under an overlay with no additives, decrease in mean death times, elevated intracerebral pathogenicity index, and cytopathic effect in chicken embryo fibroblasts. Some other markers not directly correlated to virulence, such as heat inactivation of hemagglutinin and neuraminidase, were not influenced by passage in kidney cells. In addition, all strains were slow eluters. This observation emphasizes the importance of preventing the virus from reaching the viscera.
