Increased angiogenesis in cutaneous T-cell lymphomas.

Primary cutaneous T-cell lymphomas (CTCL) represent a heterogeneous group of neoplasms derived from skin-homing T cells. CTCL behave similarly to indolent B-cell lymphomas. There is increasing evidence that angiogenesis may be important in lymphoproliferative disorders. The aim of the study was to evaluate microvessel density (MVD) as a parameter of tumor angiogenesis measured by the expression of CD34 in the skin samples in CTCL patients. Formaldehyde-fixed, paraffin-embedded skin tumor biopsy specimens from 25 patients (16 men, 9 women) with CTCL (mycosis fungoides), and 8 skin samples from healthy volunteers were analysed. The preparations were stained with haematoxylin and eosin, and evaluated histopathologically. Staining for endothelial cells with monoclonal antibody against CD34 revealed a mean number of 134 dots per mm2 for CTCL and 106 dots/mm2 for controls; the difference was statistically significant (p=0.0388). Our study shows a higher number of microvessels in primary CTCL compared with normal skin. Microvascular endothelial cells have become an important target in cancer therapy. Increased MVD in the skin of CTCL patients indicate that angiogenesis may play a role in the growth of CTCL, and raises the possibility of using angiogenesis inhibitors in CTCL therapy.

[Histopathologic studies in experimental uveitis].

UNLABELLED: Experimental uveitis is one of the main models in the diseases of autoimmunological background. The purpose of this paper was to analyze the quantitative histological changes in the experimental uveitis, induced by different types of homogenous endotoxin salts of Havnia alvei. We studied 74 eyes of Lewis rats (males) divided into 4 groups. Each group received a homogenous salt of Havnia alvei in a single subcutaneous injection. In the 1 group-LPS Ca++, in 2 group-LPS Na++, in 3 group-LPS 981, in 4 group physiological salt (control group). The histologic and immunocitochemical examinations were performed after 24, 48 hours, and after 4 and 7 days following the injection. The histologic changes were analyzed (he intensity of inflammatory reaction) using a Highly Optimazed Microscope Enviroment system. The most intensive inflammation was observed in experimental group after 24 hours (n the LPS 981D group in 5 rats out of 6, in LPS Ca++ group in 3 rats out of 6). After 48 hours the intensity of inflammatory reaction visibly decreased. On the fourth day the inflammation revealed a minimal intensity and after 7 days was practically absent. In the control group minimal inflammation was observed only in a few rats. Cilliary body hypermia was present for 48 hours in most of the experimental rats. Only a few of them had hyperemia on the fourth day. In the posterior segment minimal inflammation was noted at the end of the first day (I and II-nd group after 24 hours); this process continued until the fourth day, on the 7th day disappeared. CONCLUSIONS: The most intensive inflammation of the anterior and posterior choroidal segment is caused by homogenous salts of Havnia alvei 981.

The expression of proliferating cell nuclear antigen (PCNA) and of nucleolar organizer regions (AgNORs) correlates with the morphological type of bone marrow plasma cells in multiple myeloma (MM).

UNLABELLED: Proliferating cell nuclear antigen (PCNA) is a DNA polymerase delta accessory protein, and is an indicator of the proliferative state of the cell. Nucleolar organizer regions (NORs) are loops of DNA that carry the ribosomal RNA (rRNA) genes and are markers of aggressiveness for certain tumors. PATIENTS AND METHODS: This study tested the correlation between plasma cell morphology and PCNA and NOR expression in 50 multiple myeloma (MM) patients prior to therapy. 50 MM bone marrow trephine biopsies were studied using the monoclonal antibody PC-10 and the colloid silver nitrate method for identification of PCNA and AgNors, respectively. RESULTS: PCNA positive cells ranged from 2% to 100%, higher PCNA expression was observed in patients with immature type of MM (mean 29.5%, SD = 5.5), the highest expression was seen in plasmablastic type MM (mean 60.5%, SD = 22.6). The AgNOR count was also higher in MM with plasmablasts in the bone marrow (p < 0.001). In addition, the localization of AgNOR changed according to the type of MM: in 84% of mature and intermediate MM NORs were present in the central nucleus whereas in immature and plasmablastic MM NORs were dispersed or near the membrane. Elevated PCNA expression and AgNOR counts was also seen in MM patients with hyperviscosity coma as well as in patients with renal failure. CONCLUSION: PCNA expression and AgNOR count in myeloma indicates the myeloma's proliferative activity and correlates positively with the cytomorphology of plasma cells and clinical outcome.
