Cardioacceleratory responses to hypocretin-1 injections into rostral ventromedial medulla.

Intracisternal injections of hypocretin-1 (hcrt-1) have been shown to elicit sympathoexciatory responses. However, the location of central sites that may mediate these cardiovascular effects have not been clearly elucidated. This study was done in male Wistar rats to investigate the effects of microinjections of hcrt-1 into the rostral ventromedial medulla (RVMM) on mean arterial pressure (MAP), heart rate (HR) and the arterial baroreflex. An initial series of experiments was done to provide a detailed mapping of the location of hcrt-1- and hcrt-1 receptors (hcrtR-1)-like immunoreactivity (i.r.) in the RVMM region. Hcrt-1 and hcrtR-1 ir were found throughout the RVMM region, but primarily within the magnocellular reticular nucleus and the adjacent nucleus paragigantocellularis lateralis. In the second series, this region containing hcrt-1 and hcrtR-1 ir was explored for sites that elicited changes in MAP and HR in the anaesthetized rat. Microinjection of hcrt-1 (0.5-2.5 pmol) into the region of magnocellular reticular nucleus elicited a dose-dependent increase in HR, with little or no change in MAP. Administration (i.v.) of the muscarinic receptor antagonist atropine methyl bromide significantly attenuated ( approximately 62%) the HR response whereas, the total autonomic blockade abolished the HR response. Finally, unilateral or bilateral microinjection of hcrt-1 into the magnocellular reticular nucleus significantly attenuated the reflex bradycardia resulting from the activation of the baroreflex following the increase in MAP from an iv injection of phenylephrine. These data suggest that hcrt-1 in the RVMM region activates neuronal circuits that both inhibit vagal activity and increase sympathetic activity to the heart, and that it alters the excitability of central circuits that reflexly control the circulation.

Molecular QCA cells. 1. Structure and functionalization of an unsymmetrical dinuclear mixed-valence complex for surface binding.

Utilization of binary information encoded in the charge configuration of quantum-dot cells (the quantum-dot cellular automata, QCA, paradigm) requires molecule-sized dots for room temperature operation. Molecular QCA cells are mixed-valence complexes, and the evaluation and functionalization of an unsymmetrical heterobinuclear, two-dot, Fe-Ru molecular QCA cell is described. The solid state structures of trans-RuCl(dppm)(2)(C[triple bond]CFc) (1) (dppm = methylbis(diphenylphosphane), Fc = (eta(5)-C(5)H(5))Fe(eta(5)-C(5)H(4))) and mixed-valence [trans-RuCl(dppm)(2)(C[triple bond]CFc)][BF(4)] (1a) as well as XPS and spectroscopic data suggest class II behavior suitable for the intended application. Utilization of the trans-Cl position of 1 permits functionalization for surface binding. Two "tailed" complexes of 1, trans-Ru(dppm)(2)(C[triple bond]CFc)(C[triple bond]CPhOCH(3)) (2) and trans-[Ru(dppm)(2)(C[triple bond]CFc)(N[triple bond]CCH(2)CH(2)NH(2))][PF(6)] (3), have been prepared and characterized. The solid state structure of 3 and multinuclear NMR experiments define the structures. In addition, the spectroscopic properties of all complexes and their mixed-valence species are used to define the effect of the substituent "tail" on mixed-valence properties. Further, the electrochemistry of these compounds permits assessment of the extent of perturbation of the substituents on the comproportionation constants and overall electrochemical stability. The complexes possess properties necessary for candidate QCA molecules.

Molecular QCA cells. 2. Characterization of an unsymmetrical dinuclear mixed-valence complex bound to a Au surface by an organic linker.

Utilization of binary information encoded in the charge configuration of quantum-dot cells (the quantum-dot cellular automata, QCA, paradigm) requires surface-bound molecule-sized dots for room temperature operation. Molecular QCA cells are mixed-valence complexes, and the evaluation of a surface-bound unsymmetrical, heterobinuclear, two-dot, Fe-Ru molecular QCA cell is described. The tailed complex, trans-[Ru(dppm)(2)(C[triple bond]CFc)(N[triple bond]CCH(2)CH(2)-NH(2))][PF(6)] (dppm = methylbis(diphenylphosphane), Fc = (eta(5)-C(5)H(5))Fe(eta(5)-C(5)H(4))), is covalently modified with the molecular adapter, HS(CH(2))(10)COOH, for binding to a Au surface. Preparation and characterization of the films by AFM, XPS, and electrochemical techniques are reported. Cyclic voltammetric techniques are used to assess film growth, coverage and uniformity, effects of thiol diluents on areal densities of the complex, and stabilities of the accessible redox states. Amperometric techniques are used to investigate the efficiency of both chemical and electrochemical oxidation in producing the mixed-valence dication on the surface.

Elevated plasma homocysteine was associated with hemorrhagic and ischemic stroke, but methylenetetrahydrofolate reductase gene C677T polymorphism was a risk factor for thrombotic stroke: a Multicenter Case-Control Study in China.

BACKGROUND AND PURPOSE: It is still controversial whether elevated plasma homocysteine and the C677T polymorphism of methylenetetrahydrofolate reductase (MTHFR) gene are risk factors for stroke. The aim of the present study was to investigate the association between the 2 factors and stroke in Chinese in a large case-control study. METHODS: We recruited 1823 stroke patients (807 cerebral thrombosis, 513 lacunar infarction, 503 intracerebral hemorrhage) and 1832 controls. Total plasma homocysteine was determined by high-performance liquid chromatography. C677T polymorphism was genotyped by polymerase chain reaction and HinfI digestion. RESULTS: Total plasma homocysteine levels were significantly higher in cases than controls (median, 14.7 versus 12.8 micromol/L; P<0.001) and associated with an increased risk of 1.87-fold (95% confidence interval [CI], 1.58 to 2.22) for overall stroke, 1.72-fold (95% CI, 1.39 to 2.12) for cerebral thrombosis, 1.89-fold (95% CI, 1.50 to 2.40) for lacunar infarction, and 1.94-fold (95% CI, 1.48 to 2.55) for intracerebral hemorrhage. The C677T mutation of the MTHFR gene was positively correlated with plasma homocysteine levels in both controls (beta=0.250, P<0.001) and cases (beta=0.272, P<0.001) and more frequently in cases than in controls (47.0% versus 44.2%, P=0.017). The TT genotype was associated with an increased risk for overall stroke (odds ratio, 1.27; 95% CI, 1.04 to 1.56) and thrombotic stroke (odds ratio, 1.37; 95% CI, 1.06 to 1.78). CONCLUSIONS: The C677T polymorphism of the MTHFR gene was associated with increased risk of cerebral thrombotic stroke in Chinese. Total plasma homocysteine was correlated with both ischemic and hemorrhagic stroke, suggesting potential initiation of homocysteine-lowering therapy in this population.

Estrogen alters the bradycardia response to hypocretin-1 in the nucleus tractus solitarius of the ovariectomized female.

Experiments were performed to investigate the effect of 17beta-estradiol (E; 30 pg/ml plasma) treatment (15-25 days) in the ovariectomized (OVX) female Wistar rat on the cardiovascular responses to hypocretin-1 (hcrt-1) in the nucleus tractus solitarius (NTS). In an initial series of experiments, the distribution of hcrt-1-like immunoreactivity within the region of the NTS was mapped in both OVX only and OVX+E animals. Hcrt-1 immunoreactivity was found throughout the NTS region in both groups of females, predominantly within the caudal interstitial, commissural, medial and lateral subnuclei of the NTS. The relative density of hcrt-1 immunoreactivity in all NTS subnuclei was similar in both female groups. Microinjections of hcrt-1 (0.5-10 pmol) into the caudal lateral and medial subnuclei of the NTS complex of the alpha-chloralose of the urethane-anaesthetized E-treated OVX rat elicited a dose-related decrease in heart rate (HR). On the other hand, although a dose-response effect on arterial pressure was evident, significant arterial pressure responses were observed only at the higher dose of hcrt-1 (>2.5 pmol). In the OVX only female rat, microinjection of hcrt-1 into similar NTS sites elicited a bradycardia and depressor response only at the highest dose of hcrt-1, and these responses were significantly smaller in magnitude than those elicited in the OVX+E animal. In addition, in the OVX only animals, a few sites within the caudal commissural subnucleus of the NTS complex were found at which hcrt-1 elicited tachycardia and pressor responses. Finally, it was found that the reflex bradycardia to the activation of arterial baroreceptors as a result of increasing systemic arterial pressure with phenylephrine (2-4 microg/kg) was significantly potentiated in the OVX+E animals only. These data suggest that hcrt-1 in the NTS of the female activates a neuronal circuit that controls the circulation and that the circulating level of E alters the sensitivity of these cardiovascular circuits to hcrt-1.

Pentanucleotide TTTTA repeat polymorphism of apolipoprotein(a) gene and plasma lipoprotein(a) are associated with ischemic and hemorrhagic stroke in Chinese: a multicenter case-control study in China.

BACKGROUND AND PURPOSE: It is still inconclusive whether high plasma lipoprotein(a) [Lp(a)] level is a risk factor for stroke. Small sample size and different ethnic groups and methodologies might be contributors to the conflicts in study results. The purpose of the present study was to investigate the association between plasma Lp(a) levels, pentanucleotide TTTTA repeat (PNTR) polymorphism of the apolipoprotein(a) [apo(a)] gene, and Chinese stroke in a case-control study. METHODS: We recruited 1825 cases with stroke (44.3% cerebral atherothrombosis, 28.3% lacunar infarction, and 27.3% intracerebral hemorrhage) and 1817 controls from 7 centers in China. Lp(a) concentrations were quantified by enzyme-linked immunosorbent assay. The PNTR polymorphism of the apo(a) gene was determined by polymerase chain reaction-polyacrylamide gel electrophoresis. Conditional multivariate logistic regression analysis was used to identify independent risk factors for stroke and its subtypes. RESULTS: Lp(a) levels were significantly higher in cases than in controls (median, 28.5 versus 23.1 mg/dL; P<0.001), leading to a 1.97-fold (95% CI, 1.64 to 2.37) increase in risk for overall stroke, 2.0-fold (95% CI, 1.59 to 2.52) increase for atherothrombotic type, 2.05-fold increase (95% CI, 1.59 to 2.63) for lacunar type, and 1.64-fold increase (95% CI, 1.21 to 2.21) for hemorrhagic type. The number of PNTR negatively correlated with Lp(a) levels. Low-number repeats (sum of both alleles <16) of apo(a) PNTR were associated with both atherothrombotic stroke (odds ratio, 1.41; 95% CI, 1.04 to 1.91) and hemorrhagic stroke (odds ratio, 1.62; 95% CI, 1.09 to 2.37). CONCLUSIONS: Our results indicate for the first time that low numbers of apo(a) PNTR and plasma Lp(a) levels are independently associated with both ischemic and hemorrhagic stroke in Chinese.

[Expression of Fas, FasL and IFN-gamma in gastric cancer].

OBJECTIVE: To study the expression of Fas, Fas ligand (FasL) and IFN-gamma in gastric cancer and its possible significance. METHODS: Fifty-eight gastric paraffin wax embedded cancer tissues and fifty-three normal tissues adjacent gastric cancer were tested for the expression of Fas and FasL protein by immunohistochemistry and IFN-gamma mRNA by in situ hubridisation respectively. RESULTS: The positive rate of Fas in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(19.0% and 64.2%, respectively; chi 2 = 23.46, P = 0.00). The positive rate of FasL in cancer cells of gastric cancer tissues was significantly higher than that in gastric epithelial cells of the tissues adjacent cancer(63.8% and 45.3%, respectively; chi 2 = 3.83, P = 0.05). The positive rate of IFN-gamma in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(0.0% and 49.1%, respectively; chi 2 = 37.16, P = 0.00). CONCLUSION: The Fas-FasL system was unbalanced, and the expression of IFN-gamma was low in gastric cancer cells in this study. These may be related to the carcinogenesis of gastric epithelial cells and might be responsible for the immune escape of these cells.

Synthetic, structural, electrochemical, and theoretical studies of heterometallic aggregates with a [Pt(2)(mu-S)(2)M] core (M = Hg, Au).

Novel electroactive multimetallic compounds based on the [Pt(2)(mu(2)-S)(2)M] core, viz. [Pt(2)(PPh(3))(4)(mu(3)-S)(2)HgFc]PF(6) (1) [Fc = (eta(5)-C(5)H(4))Fe(eta(5)-C(5)H(5))] and [Pt(2)(PPh(3))(4)(mu(3)-S)(2)Hg(2)Fc'](PF(6))(2) (2) [Fc' = Fe(eta(5)-C(5)H(4))(2)], have been synthesized under the guide of electrospray mass spectrometry. The electrochemistry of these ferrocene funtionalized compounds together with the reported [Pt(2)(PPh(3))(4)(mu(3)-S)(2)HgPPh(3)](PF(6))(2) (3), [Pt(2)(PPh(3))(4)(mu(2)-S)(mu(3)-S)HgPh]PF(6) (4), and [Pt(2)(PPh(3))(4)(mu(2)-S)(mu(3)-S)AuPPh(3)]PF(6) (5) have been investigated using cyclic voltammetry and DFT calculations. These results point to a prominent ligand-based oxidation.

[Relation of pentanucleotide repeat polymorphism of apolipoprotein (a) gene to plasma lipoprotein (a) level among Chinese patients with myocardial infarction and cerebral infarction].

OBJECTIVE: Investigate the distribution and characteristics of 5'pentanucleotide TTTTA repeat (PNTR) polymorphism of apolipoprotein (a) [apo (a)] gene among Chinese patients with myocardial infarction (MI) and cerebral infarction, and its relationship to plasma lipoprotein (a) [Lp (a)] level. METHODS: DNA was extracted from the peripheral blood of 438 patients with myocardial infarction (MI, documented angiographically) and 218 controls, and 809 patients with cerebral infarction (documented by CT) and 1 817 controls. Plasma Lp (a) level was measured by ELISA. PNTR polymorphism of apo (a) gene was determined by polymerase chain reaction (PCR) and subsequent polyacrylamide gel electrophoresis. RESULTS: Eight alleles and 29 genotypes in apo (a) PNTR were detected in the four groups. The frequency of (TTTTA)(4 - 7) allele in the cerebral infarction group was 7.0%, significantly higher than that in control group (5.4%, P < 0.05). The frequency of genotype (TTTTA)(5/8) in MI group was 13.2%, significantly higher than that in the control group (7.8%, P < 0.05). No significant difference was seen in other alleles and genotypes among different groups (all P > 0.05). The level of triglyceride (TG) in patients with MI was 5.13 mmol/L +/- 1.15 mmol/L, significantly higher than that in controls (4.90 mmol/L +/- 0.99 mmol/L, P < 0.05). The level of TG in patients with cerebral infarction was 1.84 mmol/L +/- 1.36 mmol/L, not significantly different from that in controls (1.83 mmol/L +/- 1.51 mmol/L). The level of total cholesterol (TC) in patients with MI was 4.85 mmol/L +/- 1.04 mmol/L, significantly lower than that in controls (4.97 mmol/L +/- 1.00 mmol/L, P < 0.001). The level of TC in patients with cerebral infarction was 1.97 +/- 1.11 mmol/L, not significantly different from that in controls (1.79 mmol/L +/- 1.23 mmol/L). A negative correlation between the number of PNTR and Lp (a) level was observed in both MI group (P < 0.001) and cerebral infarction group (P < 0.01), and not in the control group. Small fragment repeat of apo (a) PNTR was correlated with cerebral infarction (OR = 1.4, 95% CI: 1.040 approximately 1.910), not significantly with MI (P > 0.05). Multiple logistic regression analysis showed that individuals with plasma Lp (a) > 300 mg/L had higher risk of MI (OR = 2.4, 95% CI: 1.630 - 3.594), and a higher risk of cerebral infarction (OR = 1.6, 95% CI: 1.013 - 2.021) after adjustment for age, sex, SBP, DBP, BMI, smoking, TC, TG, HDL-C, LDL-C and Glu. CONCLUSION: Plasma Lp (a) is an independent risk factor for MI and cerebral infarction. PNTR of apo (a) gene was negatively correlated with Lp (a) levels both in MI group and cerebral infarction group. Association with PNTR genotype was only found in cerebral infarction, but not in MI.

Enhanced perchloroethylene reduction in column systems using surfactant-modified zeolite/zero-valent iron pellets.

Surfactant- (hexadecyltrimethylammonium, HDTMA) modified zeolite (SMZ)/zero-valent iron (ZVI) pellets having high hydraulic conductivity (9.7 cm s(-1)), high surface area (28.2 m2 g(-1)), and excellent mechanical strength were developed. Laboratory column experiments were conducted to evaluate the performance of the pellets for perchloroethylene (PCE) sorption/reduction under dynamic flow-through conditions. PCE reduction rates with the surfactant-modified pellets (SMZ/ZVI) were three times higher than the reduction rates with the unmodified pellets (zeolite/ZVI). We speculate that enhanced sorption of PCE directly onto iron surface by iron-bound HDTMA and/or an increased local PCE concentration in the vicinity of iron surface due to sorption of PCE by SMZ contributed to the enhanced PCE reduction by the SMZ/ZVI pellets. Trichloroethylene and cis-dichloroethylene production during PCE reduction increased with the surfactant-modified pellets, indicating that the surfactant modification may have favored hydrogenolysis over beta-elimination. PCE reduction rate constants increased as the travel velocity increased from 0.5 to 1.9 m d(-1), suggesting that the reduction of PCE in the column systems was mass transfer limited.

Investigating shape and function relationship in retinal ganglion cells.

This article addresses the investigation of the relationship between neural shape and function in cat retinal ganglion cells in terms of representative morphological features. More specifically, a series of geometrical measures is extracted from two-dimensional images of these cells, and pattern recognition methods are applied in order to quantify the differentiation between the two classes (i.e., alpha, beta). The morphological measures cover several of the more meaningful geometrical features of neuronal cells, including: (a) the distribution of angles along the cell contours considering several smoothing degrees; (b) the overall interaction between the cell arborization and the surrounding space, quantified in terms of the multiscale fractal dimension; and (c) the distribution of width and extent of the dendritic processes. Several combinations of such morphological measures are assessed with respect to the separability of the classes. The obtained results indicate that the methods based on statistic relation between segment length and segment diameter, and the method of multiscale angle entropy not only successfully encapsulated a large amount of experimental data into relatively compact patterns but also marked off various ganglion cells into befit groups. On the other hand, the method of neuron classification based on fractal dimension resulted relatively less effective for class separation.

Scanning tunneling microscopy and spectroscopy investigations of QCA molecules.

Quantum-dot cellular automata (QCA), a computation paradigm based on the Coulomb interactions between neighboring cells. The key idea is to represent binary information, not by the state of a current switch (transistor), but rather by the configuration of charge in a bistable cell. In its molecular realization, the QCA cell can be a single molecule. QCA is ideally suited for molecular implementation since it exploits the molecule's ability to contain charge, and does not rely on any current flow between the molecules. We have examined using an UHV-STM some of the QCA molecules like silicon phthalocyanines and Fe-Ru complexes on Au (111) and Si (111) surfaces, which are suitable candidates for the molecular QCA approach.

Influence of quaternary ammonium on sorption of selected metal cations onto clinoptilolite zeolite.

Clay minerals and zeolites have large cation exchange capacities, which enable them to be modified by cationic surfactant to enhance their sorption of organic and anionic contaminants. In this study, the influence of quaternary ammonium surfactants on sorption of five metal cations (Cs+, Sr+, La3+, Pb2+, and Zn2+) onto a clinoptilolite zeolite was investigated. Generally, the metal cation sorption capacity and affinity for the zeolite decreased, indicating that presorbed cationic surfactants blocked sorption sites for metal cations, as the surfactant loading on the zeolite increased. Cesium and Pb2+ sorption was affected to a small extent, indicating that selective sorption for Cs+ and specific sorption for Pb2+ play an important role in addition to cation exchange. Sorption of cationic surfactants on zeolite preloaded with different metal cations showed a strong correlation with the chain length of the surfactant tail group, while the roles of the charges and types of the metal cations were minimal. As the chain length increases, the critical micelle concentration decreases and the surfactant molecules become more hydrophobic, resulting in progressive bilayer coverage. Desorption of presorbed metal cations by cationic surfactants was strongly affected by the surfactant chain length and metal type. More metal cations, particularly Sr2+ and Zn2+, desorbed with an increase in surfactant chain length. The results, in combination with those from organic and oxyanion sorption on surfactant-modified zeolite, may be used for future surfactant modification to target sorption and desorption of a specific type of contaminant or a mixture of different types of contaminants.
