[Inhibition of HPV16 E6 oncogene in cervical cancer by RNA interference].

OBJECTIVE: The efficiency of HPV16 E6 gene silenced by RNA interference in vitro and in vivo was assessed. METHODS: The specific siRNA of HPV16 E6 was designed and transfected into CaSki cells by liposome. Cell apoptotic rates and the changes in HPV16 E6 mRNA and protein before and after transfection were measured. Cervical cancer nude mice models were set up, siRNA was injected directly into subcutaneous tumor. The function of siRNA was evaluated by the changes in tumor volume, HPV16 E6 protein expression and apoptosis of tumor cells. RESULTS: In vitro research, the cell apoptotic rates were 7.7%, 11.8%, 37.4% and 12.6% respectively at 24 h, 48 h, 5th day and 9th day after transfection. The HPV16 E6 mRNA was reduced by 77%, 83%, 59% and 41% at 24 h, 48 h, 5th day and 9th day after transfection. The inhibition rates of E6 protein measured by Flow cytometry were 79.7%, 80.4%, 71.3% and 57.4% at 24 h, 48 h, 5th day and 9th day after transfection, which were confirmed by the results of Western blot. In vivo research, E6 siRNA administration groups had great power in inhibiting tumor growth, restraining E6 protein expression, increasing tumor necrosis and apoptosis. The result of repeated injections of siRNA was better than that of single injection. CONCLUSION: RNA interference with HPV16 E6 is specific and highly efficient in vitro and in vivo.

[Evaluation of the effect of fertility-saving surgery on young patients with malignant ovarian tumors].

OBJECTIVE: To evaluate the impact of fertility-saving surgery and adjuvant chemotherapy on survival and fertility of young patients with ovarian malignant tumors. METHODS: A retrospective analysis was done on 38 patients with ovarian malignant germ cell tumors, 22 patients with malignant epithelial tumors and 4 patients with sexual cord mesenchymal tumors receiving conservative treatments. Outcomes such as menstruation and reproduction ability were assessed. RESULTS: Fifty-nine among 64 patients have been alive up to now (92%). The overall survival rate for ovarian epithelial malignancies, malignant germ cell tumors and sexual cord mesenchymal tumors were 95% (21/22), 89% (34/38) and 4/4, respectively. Fifteen patients received second operation and recurrence was found in 6 patients. Among the 59 surviving patients, 53 had normal menstruation. Thirteen patients among 20 patients who wanted to get pregnant had 15 pregnancies and 9 successful deliveries. CONCLUSIONS: The management of fertility-saving surgery on patients with ovarian malignant germ cell tumors, whatever the stagings are, is a safe option. For patients with ovarian epithelial carcinomas, fertility-saving surgery is only indicated for low-stage (stage I), high-grade (G1), and patients who hope to maintain fertility function eagerly. Cisplatinum-based combination chemotherapy is necessary. Standardized chemotherapy has no effect on fertility function.

[Reversal of multi-drug resistance in ovarian cancer cell by RNA interference].

OBJECTIVE: To investigate the effects of small interference RNA (siRNA) on the inhibition of MDR1 mRNA and P-gp expression of ovarian cancer cells with high expression of MDR1 gene, and reversal of drug resistance. METHODS: siRNA was synthesized and transfected into human ovarian cancer cell line OVCAR8/TR by liposome. The expression of MDR1 mRNA at different times after transfection was measured by real time RT-PCR and the P-gp expression was detected by flow cytometry. Adenosine triphosphate (ATP)-bioluminence assay was applied to check the drug sensitivity to four different chemotherapeutic agents before and after transfection. RESULTS: The suppression rates of MDR1 mRNA were 26.42%, 84.00%, 78.43%, 45.85% and 0 respectively at 24, 48, 72, 96 and 120 hours after transfection. The P-gp suppression rates were 16.71%, 49.64%, 85.23%, 65.98%, 9.44% respectively at 24, 48, 72, 96 and 120 hours after transfection. The maximal suppression rates of MDR1 mRNA and P-gp occurred at 48 and 72 hours after transfection respectively. ATP-bioluminence assay showed that OVCAR8/TR cells were sensitive to fluorouracil, resistant to cisplatin, doxorubicin (adriamycin) and paclitaxel (taxol). After siRNA treatment, OVCAR8/TR cells were sensitive to paclitaxel and doxorubicin, but the resistance to cisplatin could not be reversed. CONCLUSIONS: RNA interference (RNAi) presents in human ovarian cancer cells. siRNA can effectively inhibit the expression of mRNA and P-gp of the multidrug resistance gene MDR1, and can reverse the drug resistance to chemotherapeutic agents which are transferred by P-gp.

[Comparative study of that synergistic effect of cisplatin combined with compound herbal medicinal prescription for tonic quality and activating blood circulation is on SKOV3 cell proliferation and apoptosis].

OBJECTIVE: To investigate the synergistic effects on cell apoptosis and growing restriction of SKOV3 cells by the combination of compound herbal medicinal prescription (CHMP) with cisplatin (DDP). METHODS: Cisplatin and two CHMP for tonic quality(CHMP1) and activating blood circulation (CHMP2), which was medicated serum, were prepared and used to treat the human ovarian carcinoma cell line SKOV3. By serum pharmacologic method, the growth and apoptosis of SKOV3 cell were observed at different time points(24,48,72, 96 h) with different concentrations of medicated serum. Coefficient of drug interaction (CDI) between CHMP, and CHMP2 was studied by MTT method. The effects of control group(A group),CHMP1 group(B group),CHMP2 group (C group), DDP group(D group), CHMP1 + DDP group(E group), CHMP2 + DDP group(Fgroup)to SKOV3 cell were studied by flow cytometry; and the cell apoptosis was observed by agarose electrophoresis; the expressions of TNFR1, caspase-8 on each group were analyzed by Western blot method. RESULTS: Synergistic effects were found between herbal medicinal mixtures and DDP, Restraining rate of SKOV3 and CHMP serum concentration was not in a dose-dependent manner as DDP was. CDI between CHMPI and CHMPS was found to be significant difference (CDI of CHMP1, CHMP2 and DDPwas 0.66, 0.58 respectively). It showed that the combined treatment was able to get better effect than single drug treatment. The performed agarose electrophoresis revealed the extracted DNA to show a typical ladder patterns for cell apoptosis. The analysis results of western blot showed the increased expressions of TNFR1 and caspase-8 after combined using of medicine, which were accord to the rates of apoptosis. CONCLUSIONS: CHMP drug granules show the synergistic effects with DDP, and the suppressing functions in the course of cell proliferation, and the inducing effect on apoptosis of human ovarian carcinoma cell line SKOV3 in vitro. And this mechanism is showed to be sponsored by the activation of TNFR1 and Caspase-8.

[A clinicopathologic analysis of primary ovarian non-Hodgkin's lymphoma].

OBJECTIVE: To observe the clinicopathologic manifestations and to investigate the clinicopathologic features and diagnosis of primary ovarian non-Hodgkin's lymphoma. METHODS: A retrospective study of clinicopathology was made for 15 cases of primary ovarian non-Hodgkin's lymphomas. The histological classifying was based on the WHO classification for tumors of hematopoietic and lymphoid tissues (2001). Immunophenotype analysis was made by SP method. RESULTS: This kind of tumor was accounted for 0.56% of all non-Hodgkin's lymphoma (NHL) received in the same period. In all 15 cases, the patients of stage III and IV were 12 (80%), and stage I and II were 3 cases (20%). Histological classification: all 15 cases (100%) were diffuse large B cell lymphoma (DLBCL), centroblast or immunoblast types. All patients had received operation; three of them also received chemotherapy of CHOP or COMP. The follow-up data was available for 4 cases, and all of them died within period of 21 days to 18 months. CONCLUSION: Primary ovarian non-Hodgkin's lymphoma is rare and the prognosis is usually poor. Establishment of the diagnosis is based on pathological biopsy and immunophenotype analysis. Owing to the diagnosis made usually after most cases operated, the surgical treatment is still the essential, and the chemotherapy and radiotherapy are also adjuvant for the tumor.

[Effects of nonsteroidal anti-inflammatory drug celecoxib on expression of cyclooxygenase-2 (COX-2) in ovarian carcinoma cell].

OBJECTIVE: To explore the effects of nonsteroidal anti-inflammatory drug Celecoxib on the expression of cyclooxygenase-2 (COX-2) in the SKOV3 cell line and the xenografted nude mice of ovarian carcinoma. METHODS: The expression of COX-2 in the SKOV3 cell was determined by reverse transcription polymerase chain reaction (RT-PCR), flow cytometry (FCM), and Western blot analysis. The expression of COX-2 in tumor cells was measured with Immunocytochemistry. RESULTS: RT-PCR showed that the expression COX-2 mRNA was strongly down-regulated in SKOV3 cells after treatment with Celecoxib or Aspirin. FCM and Western blot analysis showed that the protein product of COX-2 was strongly decreased by Celecoxib or Aspirin. The Celecoxib was more potential effects than Aspirin. The immunocytochemistry result showed that the expression of COX-2 in 10, 25, 50 mg/kg x d of Celecoxib were lower obviously than it in the control group in Xenografted nude mice. CONCLUSION: The anticarcinogenic effects of Celecoxib is probably related to the down-regulation of COX-2, and can be explained to both COX-2-dependent and -independent mechanisms.

[TRAIL impacts in growth and apoptosis of human ovarian carcinoma cell line SKOV3].

OBJECTIVE: To investigate the effect of TRAIL on growth and apoptosis of ovarian carcinoma cell line SKOV3. METHODS: The effects of TRAIL alone and/or associating with DDP to treat on SKOV3 cells growth inhibition were measured by MTT assay, and the changes of DR4 and DR5 mRNA were detected by RT-PCR technique. The cell cycle and apoptosis index were analyzed by technique of Flow Cytometry (FCM). RESULTS: The soluble TRAIL protein had the inhibiting effect on the growth SKOV3 cells. The DDP increased the ability of TRAIL to SKOV3 cell growth inhibition. The apoptotic indexes (AI) of SKOV3 cells detected by FCM were 5.9%, 13.4% and 39.5% for control group, TRAIL group and TRAIL plus DDP group, respectively. The DDP increased the DR5 expressing in SKOV3 cells by 1.95 folds than control group and by 1.54 times for the DR4 expressing than the control group (P < 0.05). CONCLUSIONS: The soluble TRAIL has the effect on inhibiting the growth of SKOV3 cells. The combination use of TRAIL and DDP increases significantly the growth-inhibiting ratio to SKOV3 cells. The AI of SKOV3 cell is raised after TRAIL intervening in. The DDP can increase the expressions of DR4 and DR5 in SKOV3 cells.

[Relation between the expression of hypoxia inducible factor-1alpha and angiogenesis in ovarian cancer using tissue microarray].

OBJECTIVE: To study the relation between the expression of hypoxia inducible factor-1alpha (HIF-1alpha)and angiogenesis in ovarian cancer. METHODS: The expressions of HIF-1alpha mRNA, vascular endothelial growth factor (VEGF), and CD(34) in 295 patients with epithelial ovarian tumors were analyzed by tissue microarray technology, in situ hybridization and immunohistochemistry, and compared with those of 13 normal ovarian tissue samples. RESULTS: The expressions of HIF-1alpha mRNA were observed in 0, 13.2%, 42.1% and 81.9% of normal ovarian tissue, benign, borderline and malignant ovarian tumors respectively. Expression rates of HIF-1alpha mRNA in borderline and invasive tumors were significantly higher than those in normal ovarian tissues and benign tumors (P < 0.01). Statistical analysis revealed that the expression of HIF-1alpha mRNA was not related to surgical stages and histological subtypes. Close positive relation was observed between the expression of HIF-1alpha mRNA and tumor histological grade (r = 0.246, P < 0.01). There were significant correlations between the expression of HIF-1alpha mRNA and VEGF (r = 0.206, P = 0.01) and microvessel density (MVD) level (r = 0.451, P < 0.01). CONCLUSION: HIF-1alpha may play a role in angiogenesis of ovarian carcinoma, and may promote the development of the carcinoma.

[Study of thalidomide on the growth and angiogenesis of ovary cancer SKOV3 transplanted subcutaneously in nude mice].

OBJECTIVE: To study the effect of thalidomide (Thd) used alone and in combination with cytoxan (CTX) on the growth and angiogenesis of human ovarian cancer transplanted subcutaneously in nude mice. METHODS: Human ovarian cancer model transplanted subcutaneously in nude mice was established, and divided into 3 groups: control group, Thd group, and Thd + CTX group. Tumor volume and weight, vascular endothelial growth factor (VEGF) mRNA, VEGF protein, microvascular density (MVD) were detected. The level of VEGF mRNA in tumor tissue was determined by relative quantative reverse transcription polymerase chain reaction. VEGF protein level in serum was determined by enzyme-linked immunosorbent assay (ELISA). MVD was calculated by immunohistochemistry. RESULTS: (1) Tumor volumes in Thd group and Thd + CTX group were smaller than those in control group (P < 0.05). (2) Expression of VEGF mRNA level in Thd group (55 +/- 9) and Thd + CTX group (26 +/- 7) was significantly lower than that in control group (79 +/- 7, P < 0.01). Serum VEGF level in Thd group, [(29 +/- 10) pg/ml] and Thd + CTX group [(12 +/- 6) pg/ml] was significantly lower than that in control group, [(71 +/- 16) pg/ml, P < 0.01]. (3) MVD in Thd group (12.6 +/- 3.3) and Thd + CTX group (10.6 +/- 1.9) was significantly smaller than that in control group (19.3 +/- 2.8, P < 0.01). CONCLUSIONS: Thalidomide can inhibit the growth and angiogenesis of human ovarian cancer transplanted subcutaneously in nude mice. Treatment with thalidomide is a potentially useful antitumor therapy for ovarian cancer.

[Study on the relationship between LOH and MI of FHIT gene and the development of cervical carcinoma].

OBJECTIVE: To explore the relationship of loss of heterozygosity (LOH) and microsatellite instability (MI) of fragile histidine triad (FHIT) gene to the development of cervical carcinoma. METHODS: Two sites of microsatellite polymophism in FHIT gene were selected to detect LOH and MI in 60 cases of primary invasive cervical carcinoma and 35 cases of cervical intra-epithelial neoplasia (CIN). RESULTS: At D3S1234 and D3S1300, the LOH rates of primary invasive cervical carcinomas were 45.0% (27/60) and 38.3% (23/60), the MI rates were 18.3% (11/60) and 11.7% (7/60), respectively. The LOH rates of CINs were 42.9% (15/35) and 37.1% (13/35), the MI rates were 11.4% (4/35), 8.6% (3/35), respectively. There were no significant differences between invasive cervical carcinomas and CINs in respect to their positive rates of LOH and MI at D3S1234 and D3S1300 (P>0.05). There were significant differences in LOH rates at D3S1234 and D3S1300 between the well/moderately differentiated cervical carcinomas and the poorly differentiated invasive cervical carcinomas (P<0.05). Significant differences were noted between the invasive cervical carcinomas with lymph node metastasis and those without lymph node metastasis in regard to their LOH and MI at the two sites (P < 0.05). The positive rates of LOH and MI for CIN III and noninvasive cervical carcinomas were higher than those for CIN I-II. CONCLUSION: The FHIT gene change is a relatively late event in CINs. The detection of the LOH of FHIT gene might be helpful to the early diagnosis and the screening of cervical carcinoma. It might also be useful for predicting the prognosis of cervical carcinoma.

[The expression of matrix metalloproteinase-2 and -9 in cervical cancer and a study of their relationship].

OBJECTIVE: To detect the expressions of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in cervical cancer and their correlation with clinicopathologic features. METHODS: With the use of specific monoclonal antibody to human MMP-2 and MMP-9, the formalin-fixed and paraffin-embedded specimens of 62 invasive carcinomas of cervix uteri, 5 carcinomas in situ, 10 normal cervical epithelia and 10 lymph node metastases were detected by immunohistochemical method. RESULTS: MMP-2 and MMP-9 protein expressions in tumor and peritumoral stromal cells were enhanced in invasive carcinoma, compared to those in carcinoma in situ and controls (P < 0.05). There was a low correlation between MMP-2 and MMP-9 (r = 0.34, P < 0.01); the expression level of MMP-9 was higher than MMP-2. The protein expressions of MMP-2 and MMP-9 did not correlate with tumor stage, histological grade or pathological subtype. MMP-9 was correlated with lymph node metastasis and lymphovascular space invasion. CONCLUSION: The over-expression of matrix metalloproteinase-2 and -9 was correlated with invasive behaviour of cervical cancer, MMP-9 was significantly correlated with the lymph node metastasis, it might become a useful prognostic indicator for early cervical cancer.

[Relationship between the reduction of FHIT expression and the development of cervical carcinoma].

OBJECTIVE: To explore the relationship between reduction of FHIT expression of fragile histidine triad (FHIT) and the development of cervical carcinoma. METHODS: Expression of the FHIT product was detected by immunohistochemistry in 22 normal cervices and 35 cervical intra-epithelial neoplasias (CINs) as well as 60 primary invasive cervical carcinomas. RESULTS: The rates for loss or reduction of expression of FHIT protein in the squamous epithelium of normal cervices, CIN I-II, CIN III and noninvasive carcinoma and invasive cervical carcinoma were 0% (0/22), 20% (4/20), 53.3% (8/15), 81.7% (49/60) respectively (P<0.05). Among the well differentiated, intermediately differentiated and poorly differentiated invasive cervical carcinoma, the rates for loss or reduction of expression of FHIT protein were 60.0% (6/10), 70.0% (14/20), and 96.7% (29/30) respectively (P<0.05). The rate of the impaired FHIT protein expression in the invasive cervical carcinoma with lymph node metastasis (90.9%) was higher than that without lymph node metastasis (79.6%). CONCLUSION: The impaired FHIT protein expression might be a useful indicator in identifying the possibility of the progression of advanced CINs into invasive cervical carcinoma. FHIT protein expression might indicate the clinical characteristic of cervical carcinoma cells and the prognosis of cervical carcinoma.
