Increased PRSS56 expression is a causal factor and therapeutic target for human axial high myopia.

High myopia (HM), characterized by significant ocular axial length elongation, affects hundreds of millions of people and is often inherited, particularly in cases that develop during childhood or adolescence. Although numerous myopia loci (MYP) have been identified, most causative genes remain undefined. Here, we analyzed two large HM pedigrees and refined the critical region through haplotype linkage analysis to a 3.9-Mb interval on 2q37.1, which was previously reported as MYP12 with an unknown pathogenic gene. Whole-genome sequencing identified the noncoding promoter variants c.-187G>T and c.-187G>C in PRSS56, encoding a trypsin-like serine protease, which exclusively co-segregated with all affected members in both pedigrees. Compared with matched controls, increased PRSS56 expression was observed in both patient-derived iPSCs carrying c.-187G>T and knock-in mice (c.-155G>T, corresponding to human c.-187G>T) that faithfully recapitulate myopia phenotypes. Noncoding PRSS56 variants promote self-expression via enhanced binding to the transcription factor EGR1, as confirmed by dual-luciferase assays. Notably, we demonstrated that higher PRSS56 levels directly increase ocular axial length in a dose- and activity-dependent manner in multiple transgenic mouse models. Guinea pig myopia models consistently exhibited high Prss56 expression, and short-wave light exposure reduced Prss56 mRNA levels and attenuated further axial elongation. Mechanistically, higher PRSS56 expression was associated with reduced abundance of myosin-4 in the sclera and with molecular signatures of scleral remodeling, which were in turn correlated with axial elongation. In conclusion, our findings provide strong genetic and functional evidence for the pathogenic role of noncoding PRSS56 variants in HM and highlight PRSS56 as a promising therapeutic target for juvenile HM.

Sorption of organic pollutants by marine sediments: implication for the role of particulate organic matter.

The objective of this study was to quantify sorption properties for kerogen/black carbon (BC)-bearing sediments. Single-solute sorption isotherms were measured for five pristine marine sediments using phenanthrene, naphthalene, 1,3,5-trichlorobenzene, and 1,4-dichlorobenzene as the sorbates. The results showed that the sorption isotherms were nonlinear and that the organic carbon normalized single point K(OC) values were comparable to those reported in the literature for the purified kerogen and BC, but are much higher than the data reported for HA and kerogen/BC-containing terrestrial soils and sediments. It is likely that kerogen and BC associated with these pristine marine sediments may not be encapsulated with humic acids or Fe and Mn oxides and hydroxides as often do in terrestrial soils and sediments. As a result, they may be fully accessible to sorbing molecules, exhibiting higher sorption capacities. The study suggests that competition from background HOCs and reduced accessibility when kerogen and BC are associated with terrestrial sediments may dramatically increase variability of sorption reactivities of geosorbents. Such variability may lead to large uncertainties in the prediction of sorption from the contents of kerogen and/or BC along with TOC.

Fast simultaneous determination of urinary 1-hydroxypyrene and 3-hydroxybenzo[a]pyrene by liquid chromatography-tandem mass spectrometry.

A fast analysis method using liquid chromatography-atmospheric pressure chemical ionization tandem mass spectrometry was developed for the simultaneous determination of the 1-hydroxypyrene (1-OHP) and 3-hydroxybenzo[a]pyrene (3-OHBaP) in urine. Mass transitions were monitored at m/z 219.3-200.0 for 1-OHP and m/z 269.2-252.2 for 3-OHBaP. Only 10 min was needed for the analysis. The recovery was 60% for 3-OHBaP and 91% for 1-OHP, respectively. And the method detection limits were 0.49 microg/L for 1-OHP and 1.03 microg/L for 3-OHBaP. The inter- and intra-day relative standard deviations were in the range of 2.8-8.9% for 1-OHP and 9.7-20.8% for 3-OHBaP, respectively. The developed method was successfully used to measure urinary PAH metabolites of student volunteers in a high school.

Differential roles of humic acid and particulate organic matter in the equilibrium sorption of atrazine by soils.

Recent studies have indicated that soil organic matter (SOM) may consist of physically and chemically different fractions, including particulate organic matter (POM), such as black carbon and unburned coal materials. The present study examined the differential roles of three different SOM fractions isolated from a peat and a topsoil in the equilibrium sorption of the herbicide atrazine (ATZ). The SOM fractions isolated from the two samples included humic acids (HAs), base-extracted humin (HM), and POM after demineralization of HM. A batch technique was employed to measure both the nonequilibrium ATZ sorption on the original and HA samples and the equilibrium ATZ sorption and desorption. The results showed that the phase-distribution relationships measured under nonequilibrium conditions were more linear and had lower sorption-capacity parameters compared with their respective isotherms measured under equilibrium conditions. The sorption isotherms were variously nonlinear, with POM exhibiting the greatest organic carbon-normalized sorption capacity. There existed apparent sorption-desorption hysteresis for each sorbent-sorbate system. It appeared that the extracted HAs could facilitate hydrolysis of ATZ when the reaction time extended to 4 d or longer. The equilibrium sorptive behavior of the HAs therefore was not examined. The present study indicated that both original samples showed lower organic carbon-normalized sorption distribution coefficients compared with their respective SOM fractions, suggesting that a fraction of sorption sites in soil aggregates were not accessed by ATZ.

Oxidation kinetics of pentachlorophenol by manganese dioxide.

This study examined the abiotic transformation kinetics of pentachlorophenol (PCP) by manganese dioxide (MnO2) at different solution chemistry and initial concentrations of PCP and MnO2. The measured PCP transformation rates were found to be on the order of 1.07 with respect to [PCP] and 0.91 and 0.87 with respect to [MnO2] and [H+], respectively. Dissolved Mn2+ and Ca2+ as background electrolytes considerably decreased the reaction rate because of their adsorption and hence blocking of active sites on MnO2 surfaces. The dechlorination number, 0.59 chloride ions per transformed PCP after a 1-h reaction, suggests that a fraction of the transformed PCP was not dechlorinated and may be coupled directly to dimeric products. Gas chromatography/ mass spectrometry and liquid chromatography/mass spectrometry/mass spectrometry techniques were used to identify two isomeric nonachlorohydroxybiphenylethers as major products and 2,3,5,6-tetrachloro-1,4-hydroquinone and tetrachlorocatechol as minor products. Product identification suggested that the reaction may include two parallel reactions to form either dimers or 2,3,5,6-tetrachloro-1,4-hydroquinone and tetrachlorocatechol via simultaneous dehydrochlorination and hydroxylation.

Competitive sorption between 17alpha-ethinyl estradiol and naphthalene/phenanthrene by sediments.

Steroid hormones such as 17alpha-ethinyl estradiol (EE2) have been frequently detected at various levels in surface waters downstream of many municipal wastewater treatment facilities. Their fate, transport, and environmental risk are currently not well characterized. This study examined the competitive sorption between EE2 and two aromatic hydrocarbon compounds, phenanthrene and naphthalene, by three sediments. The sorption isotherms of phenanthrene and naphthalene were measured at 22 +/- 0.5 degrees C using a batch technique with initial aqueous concentrations (Co) of EE2 at 0, 100, 500, and 2000 microg/L. Competitive sorption varied between EE2 and phenanthrene or naphthalene on the sediments. The linearity of the naphthalene sorption isotherm was found to increase as a function of the cosorbate EE2 concentration from 0 to 2000 microg/L. The single-point naphthalene KD value at equilibrium aqueous-phase naphthalene concentration (Ce) of 25 microg/L was reduced by 19-26% and 27-48% at Co (EE2) = 100 and 500 microg/L, respectively. The sorption of phenanthrene at its low Ce range was similarly affected by EE2, but to a much less extent, possibly because phenanthrene is more hydrophobic than EE2. At high phenanthrene Ce, no measurable change was observed even at CO (EE2) = 2000 microg/L. While the effect of naphthalene on EE2 sorption was insignificant, the competitive effect on the sorption of EE2 by phenanthrene was very significant at low EE2 concentrations. The measured single-point EE2 KD values decreased as much as 35% as the phenanthrene Ce increases from below 10 microg/L to slightly above 100 microg/L. This study suggests that the fate and transport of emerging pollutants such as EE2 could be affected in the presence of more hydrophobic pollutants in aquatic systems.

[A preoperative planning system for unilateral craniofacial malformation restoration surgery based on the volume measurement of eye sockets].

The surgical treatment of craniofacial malformation is a sophisticated task which needs careful and detailed preoperative planning. However, based on the patient's 2-D CT and X-ray images, the surgeons always feel hard to produce a convincible and satisfying surgical scheme. In this article, a craniofacial surgery simulation system based upon 3-D reconstruction and volume calculation of two eye sockets is presented. By using this system, the surgeon can virtually repose bone fragment according to the D-value between the volumes of two eye sockets. The D-value serves as a guideline to direct the bone reposing. The simulated key steps of real surgical procedure are presented, which demonstrate the efficiency of the system. The contrast images between postoperative and preoperative surgery are also given.

[The segmentation of eye socket].

In this paper, we propose a hybrid level set model with application to the segmentation of eye socket. Compared with other level set methods, the hybrid model is more robust and accurate due to the combination of region and boundary information. It solves the leaking problem of boundary based level set method. The segment results of hybrid level set model can be used in 3-D eye socket reconstruction and calculation of hydroxyapatite implant, which are useful in computer-aided surgery. Experiments have shown that hybrid level set model has very good performance in the segmentation of eye socket.

Black carbon and kerogen in soils and sediments. 2. Their roles in equilibrium sorption of less-polar organic pollutants.

The first paper of this series reported that soil/sediment organic matter (SOM) can be fractionated into four fractions with a combined wet chemical procedure and that kerogen and black carbon (BC) are major SOM components in soil/sediment samples collected from the industrialized suburban areas of Guangzhou, China. The goal of this study was to determine the sorptive properties forthe four SOM fractions for organic contaminants. Sorption isotherms were measured with a batch technique using phenanthrene and naphthalene as the sorbates and four original and four Soxhlet-extracted soil/sediment samples, 15 isolated SOM fractions, and a char as the sorbents. The results showed that the sorption isotherms measured for all the sorbents were variously nonlinear. The isolated humic acid (HA) exhibited significantly nonlinear sorption, but its contribution to the overall isotherm nonlinearity and sorption capacity of the original soil was insignificant because of its low content in the tested soils and sediments. The particulate kerogen and black carbon (KB) fractions exhibited more nonlinear sorption with much higher organic carbon-normalized capacities for both sorbates. They dominate the observed overall sorption by the tested soils and sediments and are expected to be the most important soil components affecting bioavailability and ultimate fate of hydrophobic organic contaminants (HOCs). The fact that the isolated KB fractions exhibited much higher sorption capacities than when they were associated with soil/sediment matrixes suggested that a large fraction of the particulate kerogen and BC was not accessible to sorbing HOCs. Encapsulation within soil aggregates and surface coverage by inorganic and organic coatings may have caused large variations in the accessibility of fine kerogen and BC particles to HOCs and hence lowered the sorption capacity of the soil. This variability posts an ultimate challenge for precisely predicting HOC sorption by soils from the contents of different types of SOM.

Intercorrelations among degree of geochemical alterations, physicochemical properties, and organic sorption equilibria of kerogen.

Recent studies reported that kerogen is an important natural organic material dominating sorption of relatively hydrophobic organic contaminants (HOCs) by topsoils and river sediments collected from industrialized regions. Due to its chemical and structural heterogeneity, kerogen is expected to exhibit a spectrum of sorptive phenomena for HOCs. The goal of this study is to establish correlations between heterogeneous physicochemical properties of kerogen and its sorptive characteristics for HOCs. In this study, we simulated diagenetic alterations under laboratory conditions by thermally treating a low-grade lignite at 200, 250, 300, 350, 400, 450, and 500 degrees C, yielding a series of type III kerogen samples having the same parental material but different maturations and physicochemical properties. The treated samples and the original lignite were systematically characterized using different methods and were used as the sorbents for sorption equilibrium study. The results of characterization revealed that black carbon or charwas formed at 450 degrees C or above and that, as the treatment temperature (T) increases, both O/C and H/C atomic ratios decrease whereas aromaticity and reflectance index increase. The sorption and desorption isotherms measured for 1,3,5-trichlorobenzene and phenanthrene are nonlinear and hysteretic. The nonlinearity and apparent desorption hysteresis increase as a function of Tand correlate well with rigidity and aromaticity of the organic matrix. The sorption capacity for each sorbate increases initially as T increases, reaches a maximum at 300-350 degrees C, and then decreases rapidly as Tincreases beyond 350 degrees C. This study suggests that the highly heterogeneous kerogen-based coal materials may have varied elemental compositions, functionalities, and matrix rigidity and that they could play major roles in the isotherm nonlinearity and the apparent sorption-desorption hysteresis exhibited by soils and sediments.

Sorption of steroid estrogens to soils and sediments.

Steroid estrogens at sub-micrograms per liter levels are frequently detected in surface water, and increasingly cause public concern of their potential impacts on ecosystems and human health. Assessing the environmental fate and risks of steroid estrogens requires accurate characterization of various physicochemical and biological processes involving these chemicals in aquatic systems. This paper reports sorption of three estrogens, 17beta-estradiol (estradiol), estrone, and 17alpha-ethinyl estradiol (EE2), by seven soil and sediment samples at both equilibrium and rate-limiting conditions. The results indicated that attainment of sorption equilibrium needs about 2 d when aqueous estrogen concentrations (C(t)s) are 25 to 50% of their solubility limits (S(W)S), but equilibrium requires 10 to 14 d when the Ct is 20 times lower than the S(W). The measured sorption isotherms are all nonlinear, with the Freundlich model parameter n ranging from 0.475 to 0.893. The observed isotherm nonlinearity correlates to a gradual increase of single-point organic carbon-normalized sorption distribution coefficient (capacity) (K(OC)) as the equilibrium estrogen concentration (Ce) decreases. At Ce = 0.5S(W), all three estrogens have log K(OC) values of 3.14 to 3.49, whereas at Ce = 0.02S(W), the log K(OC) values for estrone, EE2, and estradiol are within ranges of 3.40 to 3.81, 3.45 to 3.85, and 3.71 to 4.12, respectively. This study suggests that, when at sub-micrograms per liter levels, these estrogenic chemicals may exhibit even slower rates and greater capacities of sorption by soils and sediments.
