Optical factors determined by the T-matrix method in turbidity measurement of absolute coagulation rate constants.

Turbidity measurement for the absolute coagulation rate constants of suspensions has been extensively adopted because of its simplicity and easy implementation. A key factor in deriving the rate constant from experimental data is how to theoretically evaluate the so-called optical factor involved in calculating the extinction cross section of doublets formed during aggregation. In a previous paper, we have shown that compared with other theoretical approaches, the T-matrix method provides a robust solution to this problem and is effective in extending the applicability range of the turbidity methodology, as well as increasing measurement accuracy. This paper will provide a more comprehensive discussion of the physical insight for using the T-matrix method in turbidity measurement and associated technical details. In particular, the importance of ensuring the correct value for the refractive indices for colloidal particles and the surrounding medium used in the calculation is addressed, because the indices generally vary with the wavelength of the incident light. The comparison of calculated results with experiments shows that the T-matrix method can correctly calculate optical factors even for large particles, whereas other existing theories cannot. In addition, the data of the optical factor calculated by the T-matrix method for a range of particle radii and incident light wavelengths are listed.

Study on improving the turbidity measurement of the absolute coagulation rate constant.

The existing theories dealing with the evaluation of the absolute coagulation rate constant by turbidity measurement were experimentally tested for different particle-sized (radius = a) suspensions at incident wavelengths (lambda) ranging from near-infrared to ultraviolet light. When the size parameter alpha = 2pi a/lambda > 3, the rate constant data from previous theories for fixed-sized particles show significant inconsistencies at different light wavelengths. We attribute this problem to the imperfection of these theories in describing the light scattering from doublets through their evaluation of the extinction cross section. The evaluations of the rate constants by all previous theories become untenable as the size parameter increases and therefore hampers the applicable range of the turbidity measurement. By using the T-matrix method, we present a robust solution for evaluating the extinction cross section of doublets formed in the aggregation. Our experiments show that this new approach is effective in extending the applicability range of the turbidity methodology and increasing measurement accuracy.

A combined effect of the KPNA3 and KPNB3 genes on susceptibility to schizophrenia.

Recent studies suggest that both the KPNB3 gene and the KPNA3 gene in the long arm of chromosome 13 (13q) are associated with schizophrenia. Because these two genes belong to the same family of karyopherins, their combined effect on illness was investigated among 238 Chinese family trios consisting of fathers, mothers and affected offspring with schizophrenia. We detected three single nucleotide polymorphisms (SNPs), including rs626716 at the KPNB3 locus, and rs3782929 and rs3736830 at the KPNA3 locus. The transmission disequilibrium test (TDT) showed allelic association for rs626716 (X2=10.77, P=0.001) and for rs3782929 (X2=4.89, P=0.027) but not for rs3736830 (X2=0.29, P=0.59). Although the conditional test did not show association either for the rs626716-rs3782929 combinations (X2=1.329, d.f.=2, P=0.514) or for the rs626716-rs3736830 combinations (X2=0.606, d.f.=2, P=0.739), the 1-d.f. test showed association for the rs626716(C)-rs3782929(G) combination (X2=10.79, P=0.001) and for the rs626716(C)-rs3736830(G) combination (X2=8.64, P=0.003). The present work suggests that the combination of the KPNA3 gene and the KPNB3 gene may increase a genetic risk for schizophrenia.

Improved procedure on the microscopic approach to determine colloidal stability.

We present an improved procedure on the approach to determine the stability of polystyrene spheres at microscopic particle levels by means of artificially induced particle collisions with the aid of optical tweezers [J. Chem. Phys. 119, 2399 (2003)]. The basic consideration on this new development is that the major contribution to the sticking probability for a particle pair caught into the optical trap for a short period is from its single collision; therefore, if the trapping duration for the pair is taken to be short, the accumulated sticking probability will be a good approximation for the single collision. The experimental procedure associated with this approximation does not resort to exactly controlling the short trapping duration or request the trapping duration correction as previously reported, and therefore it is more practical and applicable for a broader range of the stability ratio. The experimental results under different electrolyte concentrations by the new procedure are consistent with those from the turbidity measurements.

[A study on DNA damage of mouse livers induced by methylmercury].

OBJECTIVE: To study the DNA damage induced by methylmercury (MeHg) in mouse hepatocytes. METHODS: The single cell gel electrophoresis (SCGE) was used to study the DNA damage of mouse hepatocytes when treated with different doses in vivo and in vitro. RESULTS: After treated with high, middle and low doses of MeHg for 12 h in vivo (i.p.), the proportion of DNA damage was increased and the ratio of living cells was decreased; and both effects showed significantly dose-effect relationships. Similar effects were found when different MeHg doses were administered to hepatocytes in vitro for 1 h. CONCLUSION: MeHg induces DNA damage in mouse hepatocytes.
