Leveraging Interradiomic Feature Relationships for Enhanced Prediction of Distant Metastasis and Characterization of Heterogeneity in Head and Neck Cancer.

PURPOSE: Distant metastasis remains a major cause of treatment failure in head and neck (HN) cancer, highlighting the need for more accurate early risk stratification. This study developed and validated a deep radiomics framework to characterize tumor heterogeneity from pretreatment computed tomography (CT) images and improve prediction of distant metastasis-free survival (DMFS). METHODS AND MATERIALS: This multicenter study included 3421 patients with HN cancer from 4 cohorts across 12 institutions. Radiomics features were extracted from primary tumors and transformed into OmicsMaps, a structured representation that spatially organizes interfeature relationships to facilitate learning of complex prognostic patterns. A convolutional neural network was trained to derive prognostic signatures, which were integrated with key clinical variables to construct an OmicsMap-clinical fusion model for patient risk stratification. Model performance was assessed using the concordance index (C-index) and time-dependent area under the receiver operating characteristic curve (AUC) in the CT Images from Large Head and Neck Cohort (RADCURE), HEAD-NECK-RADIOMICS-HN1 (HN1), and Head-Neck-Positron Emission Tomography-Computed Tomography (HN-PET-CT) cohorts. Radiogenomic analyses using RNA-seq data were conducted in the Cancer Genome Atlas Head-Neck Squamous Cell Carcinoma (TCGA-HNSC) cohort to investigate biological characteristics associated with the imaging-defined risk groups. RESULTS: The OmicsMap achieved C-index values of 0.742, 0.768, and 0.671 in the RADCURE, HN1, and HN-PET-CT cohorts, outperforming the conventional radiomics approach by 5.40%-6.37%. Incorporating clinical variables further improved generalizability, yielding a C-index of 0.864 (HN1) and 0.730 (HN-PET-CT), with time-dependent AUC of 0.727-0.895. The fusion model consistently stratified patients into distinct high- and low-risk groups for both DMFS and overall survival across cohorts (P <.01). Radiogenomic analyses revealed enrichment of immune-related pathways in the low-risk group, whereas the high-risk group exhibited a more aggressive phenotype enriched for proliferation, hypoxia, and epithelial-mesenchymal transition pathways, along with a fibrosis-prone tumor microenvironment characterized by extracellular matrix remodeling. CONCLUSIONS: Modeling interradiomic feature relationships using the OmicsMap representation substantially improves CT-based prediction of DMFS and characterization of tumor heterogeneity in HN cancer, supporting precision risk stratification in clinical oncology.

Two-dimensional multipole solitons in nonlocal nonlinear media.

We present the experimental observation of scalar multipole solitons in highly nonlocal nonlinear media, including dipole, tripole, quadrupole, and necklace-type solitons, organized as arrays of out-of-phase bright spots. These complex solitons are metastable, but with a large parameters range where the instability is weak, permitting their experimental observation.

Gap solitons supported by optical lattices in photorefractive crystals with asymmetric nonlocality.

We address the impact of the asymmetric nonlocal diffusion nonlinearity of gap solitons supported by photorefractive crystals with an imprinted optical lattice. We reveal how the asymmetric nonlocal response alters the domains of existence and the stability of solitons originating from different gaps. We find that in such media gap solitons cease to exist above a threshold of the nonlocality degree. We discuss how the interplay between nonlocality and lattice strength modifies the gap soliton mobility.

Stabilization of vector soliton complexes in nonlocal nonlinear media.

We introduce vector soliton complexes in nonlocal Kerr-type nonlinear media. We discover that under proper conditions the combination of nonlocality and vectorial coupling has a remarkable stabilizing action on multihumped solitons. In particular, we find that stable bound states featuring several field oscillations in each soliton component do exist. This affords stabilization of vector soliton trains incorporating a large number of humps, a class of structures known to self-destroy via strong instabilities in scalar settings.

Upper threshold for stability of multipole-mode solitons in nonlocal nonlinear media.

We address the stability of multipole-mode solitons in nonlocal Kerr-type nonlinear media. Such solitons comprise several out-of-phase peaks packed together by the forces acting between them. We discover that dipole-, triple-, and quadrupole-mode solitons can be made stable, whereas all higher-order soliton bound states are unstable.

Soliton mobility in nonlocal optical lattices.

We address the impact of nonlocality in the physical features exhibited by solitons supported by Kerr-type nonlinear media with an imprinted optical lattice. We discover that the nonlocality of the nonlinear response can profoundly affect the soliton mobility, hence all the related phenomena. Such behavior manifests itself in significant reductions of the Peierls-Nabarro potential with an increase in the degree of nonlocality, a result that opens the rare possibility in nature of almost radiationless propagation of highly localized solitons across the lattice.

Multicolor vortex solitons in two-dimensional photonic lattices.

We report on the existence and stability of multicolor lattice vortex solitons constituted by coupled fundamental frequency and second-harmonic waves in optical lattices in quadratic nonlinear media. It is shown that the solitons are stable almost in the entire domain of their existence, and that the instability domain decreases with the increase of the lattice depth. We also show the generation of the solitons, and the feasibility of the concept of lattice soliton algebra.

Antimitotic and antifungal C-3/C-3''-biflavanones from Stellera chamaejasme.

Two new C-3/C-3'' biflavanones, chamaejasmenin D (1) and isochamaejasmenin B (2), were isolated from the roots of Stellera chamaejasme, together with five known biflavanones, chamaejasmenin A (3), chamaejasmenin B (4), neochamaejasmin A (5), sikokianin A (6), and chamaejasmenin C (7). Their structures were elucidated by spectroscopic methods, including 2D NMR techniques. Among these compounds, 1-3 demonstrated potent antimitotic and antifungal activity with minimum inhibitory concentration (MIC) values of 6.25, 6.25, and 3.12 microg/ml, respectively.

Spatiotemporal discrete multicolor solitons.

We have found various families of two-dimensional spatiotemporal solitons in quadratically nonlinear waveguide arrays. The families of unstaggered odd, even, and twisted stationary solutions are thoroughly characterized and their stability against perturbations is investigated. We show that the twisted and even solitons display instability, while most of the odd solitons show remarkable stability upon evolution.

Exact soliton solutions for the core of dispersion-managed solitons.

We consider the averaged dispersion-managed (DM) fiber system equation, which governs the dynamics of the core of the DM solitons. For a special case of such a system equation, we derive the exact soliton solutions using the Darboux transformation. Further, we discuss the interaction scenario between two neighboring solitons. Finally, we derive a dark soliton solution for such a system by assuming an ansatz, and the interaction between neighboring dark solitons is discussed.

Modulation instability and solitons on a cw background in an optical fiber with higher-order effects.

We construct the Lax pair for a higher-order nonlinear Schrödinger equation that includes terms accounting for the third-order dispersion, the self-steepening effect, and the delayed nonlinear response effect. Two exact analytic solutions that describe (i) modulation instability and (ii) soliton propagation on a continuous wave background are obtained by using the Darboux transformation. In addition, we analyze the amplification-absorption and quintic nonlinearity effects on the second solution in the adiabatic approximation.

Gray optical dips in the subpicosecond regime.

Narrow optical dip solutions are investigated when, besides self-phase modulation and group velocity dispersion, also third-order dispersion, nonlinear dispersion, and stimulated Raman scattering are taken into account. By using the inverse scattering transform for the higher-order optical nonlinear Schrödinger (HNLS) equation under Hirota parameter conditions, the dark N-soliton solution is constructed. The explicit forms of the one- and two-soliton solutions are investigated in detail. The results show an interesting property of the gray two-soliton solution. Two gray dips do not interact provided their modulation depths are appropriately chosen. In addition, when generalizing the HNLS equation (to regions beyond the Hirota parameter conditions), it can be shown that also quite stable generalized two-dip solitary wave solutions exist. The latter, although not belonging to integrable systems, approximately preserve most of the interesting properties detected for the integrable Hirota equation.
