[Estimation of postmortem interval using myofibril fragmentation index].

OBJECTIVE: To study the relationship between postmortem interval and the myofibril fragmentation index of skeletal muscle. METHODS: Rabbit skeletal muscle were left in the room temperature for different postmortem intervals, and the protein concentration of each sample was detected by using biuret method. Furthermore, the myofibril fragmentation index of each sample was measured under the protein concentration level of 0.5 mg/mL. RESULTS: The myofibril fragmentation index increased obviously according to the postmortem interval prolongation. CONCLUSION: The myofibril fragmentation index may be used on the estimation of early postmortem interval.

[Primary study in rabbit penile corpus cavernosum smooth muscle cell culture and RyRs expression].

OBJECTIVE: To investigate expression of ryanodine receptors (RyRs) in rabbit penile corpus cavernosum smooth muscle (CCSM) cells. METHODS: New Zealand White Rabbit CCSM cells were cultured by primary tissue culture. Using CCSM cells and fibroblast have different adherence velocity, CCSM cell can be purified. Identification of CCSM cell was by inverted microscope and immunofluorescence technique. The cDNA sequence of RyRs was found out by searching genebank. Three pair of primers were designed by Primer Premier 5.0. The RyRs subunits mRNA was detected by reverse transcription PCR in cultured CCSM cells. RESULTS: After 7d, we found growth of cultured cells. While 15 to approximately 20 d, cells filled the bottom of culture flask. They were identified by inverted microscope and immunofluorescence technique. After purification, purity of CCSM cells was near to 100%. It suggested only RyRs1 subunit was expressive in CCSM by RT-PCR. CONCLUSION: RyRsl subunit is expressed in CCSM cells. It suggests that RyRs contribute to the regulation of Ca2+ in CCSM cells.

[Study of CTnI immunohistochemistry on acute myocardial infarction in rabbits].

OBJECTIVE: To investigate the signification of CTnI in acute myocardial infarction. METHODS: The model of myocardial infarction was established by ligating the left ventricle branch. Immunohistochemistry and image analysis were used to detect the change of CTnI in the model, and The sensitivity of immunohistochemistry and HE coloration were also compared. RESULTS: The acute myocardial infarction tissue showed obvious depletion of CTnI, there was no characterization of myocardial infarction in HE coloration. CONCLUSION: CTnI is sensitive to diagnosis of acute myocardial infarction.

[Estimation of postmortem intervals by using terminal deoxynucleotidyl transferase].

OBJECTIVE: To investigate the relationship between DNA degradeation and postmortem interval. METHODS: dUTP was transferred to 3'terminal of DNA by using terminal deoxynucleotidyl transferase (TDT), then the reminders of dUTP after experimental reaction, as indicator of quantity of DNA degradation, were detected. RESULTS: The reminders of dUTP were decreasing along with the postmortem interval. CONCLUSION: Postmortem DNA degradation may be used in postmortem interval judgment.

[The relationship between expression of FN in human skin injury and early injury time].

OBJECTIVE: In order to supply an effective reference of early injury time estimation and explore the time limit of detection of EDA\EDB mRNA in human skin samples, the expression of alternative splicing segment of fibronectin--EDA\EDB in incised wound of human skin were studied. METHODS: Using in situ hybridization with DIG-labeled anti-sense RNA probe, the expression of FN EDA\EDB domain was detected in human skin incised wound at the early stage of injury (from 30 min to 3 h). RESULTS: The positive expression rates of FN-EDA\EDB immediately after injury and area far away from wound were same as the control group. The expression of FN-EDA\EDB in human skin incised wound showed a gradually increased tendency in early injury time (within 3 h). The positive expression cells were mainly distributed in basement cells of epidermis and the expression of EDA is much higher than EDB. It's difficult to detect EDA\EDB mRNA when the samples were deposited in air for 4 hour. CONCLUSION: FN-EDA\EDB may be used as a sensitive mark for the estimation of early injury time. The in-situ hybridization technique is not applicable in the application.

[The expression of FN-IIIcs in the wound healing and its relationship with injury time].

OBJECTIVE: To research the expression of alternative splicing segment of fibronectin-IIIcs in incised wound of skin, to offer the experimental data for early injury time judgement. METHODS: Using in situ hybridization with DIG-labeled anti-sense RNA probe, the expression of FN-IIIcs domain was detected in both human and rat skin incised wound. RESULTS: (1) The expression of FN-IIIcs improved from the time of 30 min after injury, reached peak at 6 h and then reduced gradually. (2) The positive expression cells were mainly distributed in hair follicles, sebaceous glands and endothelium, however, in human beings they were basement cells of epidermis. CONCLUSION: The expression of FN-IIIcs domain would be a supposed useful criteria for early injury time judgement.

[The in situ hybridization effort estimation between antisense RNA probe and random primed DNA probe].

OBJECTIVE: To estimate the effort between antisense RNA probe and random primed DNA probe. METHODS: The in situ hybridization was conducted on parafin section from wounding model of rat skin. RESULTS: Although both probes appeared positive staining, RNA probes was superior to DNA probes in terms of depth of staining and background. CONCLUSION: RNA probe showed more satisfactorily on ISH.

[Tryptase and fatal anaphylaxic reaction].

OBJECTIVE: To investigate the relationship between tryptase in serum and anaphylaxis. METHODS: The concentrations of tryptase in the sera of heart blood in three persons died from anaphylaxis shock were detected by ELISA. The first sample was obtained from a man, aged 38, died of injecting Amikacin. The second sample was obtained from a man, aged 42, died of injecting Cephradine. The third sample was from a woman, aged 39, died of injecting Lincomycin. All samples were stored in -20 degrees C. RESULTS: The concentrations of tryptase in sera were 52 ng/ml, 121 ng/ml and 0.73 ng/ml. It was unknown why the concentration of tryptase in the third sample was normal. CONCLUSION: In fetal anaphylaxia reaction tryptase measurement is a useful indicator, but the diagnosis is not to be based on the test alone.
