Protein synthesis in the medial preoptic area is important for the mating-induced decrease in estrus duration in hamsters.

Abstract

Sexual receptivity in female hamsters potentially lasts for about 16 h. However, vaginal cervical stimulation (VCS) from a male during mating eventually reduces receptivity and can shorten the duration of behavioral estrus. The process by which this change in response to the male takes place is unknown. Recently, detection of the Fos protein has indicated that the medial preoptic area (POA) is one of the brain regions particularly responsive to VCS. Additionally, the POA may have an inhibitory effect on sexual receptivity. To determine if protein synthesis in the POA is required to initiate the VCS-induced decrease in estrus duration, a protein synthesis inhibitor (anisomycin, 0.50 microg) or a control substance (cholesterol) was applied bilaterally to the POA of steroid-primed ovariectomized female hamsters. Females were tested with a sexually active male at five time points following the initial test for sexual receptivity (hour 1, 2, 6, 12, and 24). Half of the females tested were allowed to receive VCS from a male, while half were fitted with vaginal masks to prevent penile intromission. Each group receiving VCS showed a significant decrease in lordosis duration evident between hour 2 and hour 6, except the group which received anisomycin in the POA. In this respect the POA anisomycin group was similar to animals which did not receive VCS. Hamsters with vaginal masks and the anisomycin/POA animals allowed to receive VCS exhibited their first decrease in lordosis duration between hour 6 and hour 12. These results indicate that protein synthesis is important for VCS-induced decrease in estrus duration in the POA.

Enhanced cortical extracellular levels of cholecystokinin-like material in a model of anticipation of social defeat in the rat.

The involvement of cholecystokinin (CCK) in the mechanisms of stress and/or anxiety was assessed by in vivo microdialysis in rats subjected to a social stress paradigm. During the initial 30 min period of each conditioning session, a male Sprague Dawley rat (intruder) was placed in a protective cage inside the cage of a male Tryon Maze Dull rat (resident), allowing unrestricted visual, olfactory, and auditory contacts but precluding close physical contact between them. During the following 15 min period, both the protective cage and the resident were removed (nondefeated intruders) or only the protective cage was removed allowing the resident to attack the intruder (defeated rats). This procedure was repeated once daily for 4 d. On the fifth day, a guide cannula was implanted into the prefrontal cortex of intruders. During a single 30 min test session, performed 4 d later, intruders were subjected to only the 30 min protected confrontation to the resident. Anxiety-like behavior (immobility, ultrasonic vocalizations, and defensive postures), associated with an increase (approximately +100% above baseline) in cortical outflow of CCK-like material (CCKLM), were observed in defeated intruders. Pretreatment with diazepam (5 mg/kg, i.p.), but not buspirone (0.5-2 mg/kg, i.p.), prevented both the anxiety-related behavior and CCKLM overflow. The selective CCK-B receptor antagonist CI-988 (2 mg/kg, i.p.) reduced the anxiety-like behavior without affecting the increase in CCKLM outflow. These data indicate that anticipation of social defeat induces a marked activation of cortical CCKergic neurons associated with anxiety-related behaviors in rats.

Suppressing aggressive behavior with analogs of allopregnanolone (epalon).

3alpha-Hydroxy-20-oxo-5alpha-pregnan-21-yl hemisuccinate (8) was produced by partial acylation of 3alpha,21-dihydroxy-5alpha-pregnan-20-one (14). 3alpha-Fluoro-5alpha-pregnan-20-one (9) was prepared by treatment of 3beta-hydroxy-5alpha-pregnan-20-one (11) with DAST and by solvolysis of tosylate 12 with tetrabutylammonium fluoride. A behavioral test on mice was performed using 3alpha-hydroxy-5alpha-pregnan-20-one (1) and compounds 8 and 9. Compound 8 was found to be inactive, while the fluoro derivative 9 selectively reduced aggressive behavior in mice more than the corresponding 3alpha-hydroxy compound 1; locomotion and other behavioral features were not affected.

Dating violence: a critical review of the literature.

The investigation of dating violence has previously been underrepresented in the interpersonal violence literature. Within the past 2 decades, however, researchers have significantly advanced our knowledge of the variables associated with dating violence. This critical article provides a comprehensive review of the current body of literature on dating aggression. Research on dating violence is presented, progressing from prevalence rates and types of violence to an exploration of victim and perpetrator characteristics. There is an emphasis on the necessity to establish a theoretical model of typology to allow investigation of the distinct subgroups of violent individuals. The deficits in the present body of literature are presented and include sampling methods, dependent measures, and data analyses. Finally, implications for future research and prevention are provided.
