Dimethyl sulfoxide (DMSO)--an overview.

Abstract

The solvent dimethyl sulfoxide (DMSO) has received intense lay publicity sporadically over the last two decades. The compound has been particularly promoted as an analgesic and anti-inflammatory agent. Its membrane penetrant-carrier properties have also been of prime interest. The history, physical/chemical and pharmacological properties, pharmacokinetics, clinical trials, therapeutic uses, side effects and dosage/administration of DMSO are reviewed. Legal implications and concerns with regard to DMSO use are presented in addition to advice for patients.

Alkamide stability in Echinacea purpurea extracts with and without phenolic acids in dry films and in solution.

Degradation of the major alkamides in E. purpurea extracts was monitored under four different accelerated storage conditions, phenolic-depleted and phenolic-rich dry E. purpurea extracts and phenolic-depleted and phenolic-rich DMSO E. purpurea extracts at 70, 80, and 90 degrees C. Degradation of alkamides followed apparent first-order reaction rate kinetics. Alkamides degraded faster in dry films than in DMSO solution. The phenolic acids acted as antioxidants by limiting the loss of the alkamides in dry E. purpurea extracts. In contrast, E. purpurea alkamides in DMSO degraded faster when the phenolic fraction was absent. The overall order of degradation rate constants was alkamides 1 approximately 2 approximately 6 > 9 approximately 8 > 3 approximately 5 approximately 7. The energy of activation (Ea) predicted for alkamide degradation averaged 101 +/- 12 kJ/mol in dry films +/- phenolic acids, suggesting the oxidation mechanism was the same under both conditions. In DMSO solutions, Ea values were about one-half of those in dry films (61 +/- 14 kJ/mol), suggesting a different mechanism for alkamide oxidation in solution compared to dry. Predicted half-lives for alkamides in extracts suggested very good stability.

Molecular basis for dimethylsulfoxide (DMSO) action on lipid membranes.

Dimethylsulfoxide (DMSO) is an aprotic solvent that has the ability to induce cell fusion and cell differentiation and enhance the permeability of lipid membranes. It is also an effective cryoprotectant. Insights into how this molecule modulates membrane structure and function would be invaluable toward regulating the above processes and for developing chemical means for enhancing or hindering the absorption of biologically active molecules, in particular into or via the skin. We show here by means of molecular simulations that DMSO can induce water pores in dipalmitoyl-phosphatidylcholine bilayers and propose this to be a possible pathway for the enhancement of penetration of actives through lipid membranes. DMSO also causes the membrane to become floppier, which would enhance permeability, facilitate membrane fusion, and enable the cell membrane to accommodate osmotic and mechanical stresses during cryopreservation.

Synthesis and characterization of stable organosols of silver nanoparticles by electrochemical dissolution of silver in DMSO.

Stable organosols of silver nanoparticles (AgNPs) without any capping agents have been synthesized by an electrochemical dissolution of a sacrificing silver electrode in dimethyl sulfoxide (DMSO). The peak at 425 +/- 5 nm observed in the UV-vis spectra was attributed to the surface plasmon resonance for silver. The formation of nanoparticles of silver was confirmed by X-ray diffraction analysis (XRD). In TEM, three ranges of particle size were observed, namely, 3.75 +/- 0.8, 6.25 +/- 0.5, and 9.25 +/- 0.3 nm. The correlation among these sizes was explained by a new model based on a droplet coalition. Based on that, sizes correlation fits very well in the empirical formula 5d(1)(3)n' + (3 - n')d(2)(3) = d(3)(3) where d(1), d(2), and d(3) are three sizes of particles and n' is an integer having values 0, 1, 2, 3, .... The sols prepared in DMSO were stable against flocculation for months. An unusual solution stability without any capping agents was attributed to the formation of Ag(I)DMSO complex on particle surface, which was confirmed by FTIR, fluorescence spectroscopy, and thermal analysis.
