Functionally graded fluoridated apatites.

Abstract

Fluoridated hydroxyapatite was synthesized at 80 +/- 1 degree C and pH 7.4 +/- 0.2 using a gradient fluoride supply system. X-ray diffraction analysis showed a typically apatitic pattern, although the (3 0 0) reflection was broader than that of homogeneous fluorapatite. Scanning electron micrographic observation indicated that the apatite was composed of rod-like crystals similarly to fluorapatite. High-resolution transmission electron microscopy showed electron damage in the core of the crystal. When the apatite pellet was prepared, electron spectroscopy for chemical analysis showed a negative gradient of fluoride concentration with depth in the crystals. The apparent solubility in 0.5 mol/l acetate buffer solution (37 degrees C and pH 4.0) was 9.16 +/- 0.39 mmol/l, much less than that of homogeneous hydroxyapatite 32.3 +/- 1.9 mmol/l, and less than that of heterogeneous two-layer fluoridated apatite with an outer fluoride-rich layer 12.5 +/- 0.6 mmol/l, which was synthesized previously by supplying fluoride during the latter half of the experimental period. These results suggest that graded fluoridated apatite may be formed by this process and have higher acid resistance than two-layer fluoridated apatite.

A new series of estrogen receptor modulators that display selectivity for estrogen receptor beta.

A series of 1,3,5-triazine-based estrogen receptor (ER) modulators that are modestly selective for the ERbeta subtype are reported. Compound 1, which displayed modest potency and selectivity for ERbeta vs ERalpha, was identified via high-throughput screening utilizing an ERbeta SPA-based binding assay. Subsequent analogue preparation resulted in the identification of compounds such as 21 and 43 that display 25- to 30-fold selectivity for ERbeta with potencies in the 10-30 nM range. These compounds profile as full antagonists at ERbeta and weak partial agonists at ERalpha in a cell-based reporter gene assay. In addition, the X-ray crystal structure of compound 15 complexed with the ligand binding domain of ERbeta has been solved and was utilized in the design of more conformationally restrained analogues such as 31 in an attempt to increase selectivity for the ERbeta subtype.

4-Hydroxypyridine-2,6-dicarboxylatodioxovanadate(V) complexes: solid state and aqueous chemistry.

The aqueous solution and solid state properties of (4-hydroxypyridine-2,6-dicarboxylato)dioxovanadate(V) (also referred to as (4-hydroxydipicolinato)dioxovanadate(V) or (chelidamato)dioxovanadate(V) and abbreviated [VO(2)(dipic-OH)](-)) were investigated. By using (1)H, (13)C, (17)O, and (51)V NMR 1D and 2D spectroscopy, the species present in solution, together with pK(a) values, equilibrium constants, and labilities, were characterized. The complex is most stable at acidic pH down to pH 1 where it is protonated. The stability of this complex is higher than that of the parent dipicolinatodioxovanadate(V) complex. The dipic-OH ligand is coordinated in a tridentate manner throughout the pH range studied, and the vanadium(V) atom is five-coordinate. Solid state structures of (NMe(4))[VO(2)(dipic-OH)].H(2)O (monoclinic, P2(1)/n) and Na[VO(2)(dipic-OH)].2H(2)O (triclinic, P1) were determined. The discrete complex anions in (NMe(4))[VO(2)(dipic-OH)].H(2)O are connected by hydrogen bonding between the hydroxyl group, a water molecule, and a carboxylate oxygen atom. Changing the counterion from NMe(4)(+) to sodium ion in Na[VO(2)(dipic-OH)].2H(2)O leads to the formation of a polymeric structure. Dynamic processes in solution were explored by using (1)H and (13)C EXSY NMR spectroscopy; exchange between complex and free ligand below pH 4 was observed. The differences between the dipicolinatodioxovanadate(V) parent complex and the [VO(2)(dipic-OH)](-) complex in the solid state and in solution demonstrate the subtle consequences of the one substitutional difference between the two ligands. The insulin-mimetic properties of this compound are likely to be of mechanistic interest in developing an understanding of the mode of action of the few known insulin-mimetic vanadium(V) complexes.

Kinase inhibitors and the case for CH...O hydrogen bonds in protein-ligand binding.

Although the hydrogen bond is known to be an important mediator of intermolecular interactions, there has yet to be an analysis of the role of CH...O hydrogen bonds in protein-ligand complexes. In this work, we present evidence for such nonstandard hydrogen bonds from a survey of aromatic ligands in 184 kinase crystal structures and 358 high-resolution structures from the Protein Data Bank. CH groups adjacent to the positively charged nitrogen of nicotinamide exhibit geometric preferences strongly suggestive of hydrogen bonding interactions, as do heterocyclic CH groups in kinase ligands, while other aromatic CH groups do not exhibit these characteristics. Ab initio calculations reveal a considerable range of CH...O hydrogen bonding potentials among different aromatic ring systems, with nicotinamide and heterocycles preferred in kinase inhibitors showing particularly favorable interactions. These results provide compelling evidence for the existence of CH...O hydrogen bonds in protein-ligand interactions, as well as information on the relative strength of various aromatic CH donors. Such knowledge will be of considerable value in protein modeling, ligand design, and structure-activity analysis.
