On-line coupling flow injection microcolumn separation and preconcentration to electrothermal atomic absorption spectrometry for determination of (ultra)trace selenite and selenate in water.

Abstract

A flow injection manifold with an air-segmented and air-transported operational sequence for on-line coupling of microcolumn separation and preconcentration to electro-thermal atomic absorption spectrometry (ETAAS) was developed for the determination of (ultra)trace selenite and selenate in water. The determination of selenite was achieved by selective reaction with pyrrolidine dithiocarbamate (PDC), sorption of the resultant Se-PDC compound onto a conical microcolumn (10.2 microL) packed with RP C18 sorbent, elution with ethanol, and detection by ETAAS. The concentration of selenate was obtained as the difference between the concentrations of selenite after and before prereduction of selenate to selenite. With the developed manifold and operation sequence, the dispersion during elution and eluate transport and the eluent volume required for complete elution of the sorbed analyte were minimized. As a result, the sorbed analyte was quantitatively eluted from the column with only 26 microL of ethanol, and all the eluate was automatically introduced into the graphite tube by an air flow without the need of preheating the graphite tube or precise timing. Pretreatment of the graphite tube with iridium as a long-term "permanent" modifier effectively prevented analyte loss arising from the high volatility of the Se-PDC compound and greatly improved the precision, sensitivity, and detection limit. One thermal pretreatment of the graphite tube with injection of 150 microgram of iridium made possible at least 200 repetitive atomization cycles. With a preconcentration time of 180 s and a sample flow rate of 1.4 mL min(-1), an enhancement factor of 112 was achieved in comparison with direct injection of 30 microL of aqueous solution. The detection limit (3s) was 4.5 ng L(-1)Se. The RSD (n = 7) was 3.8% at 20 ng L(-1)Se. The concentrations of selenite and selenate determined in synthetic aqueous mixtures were in good agreement with the expected values. The recoveries for selenite from spiked seawater samples ranged from 98 to 102%. The concentrations of selenite in several seawater reference materials obtained with simple aqueous standard solutions for calibration agreed well with the certified and information values, respectively. In addition, the developed method was successfully applied to the certification of selenite and selenate in water.

Widespread marine and freshwater distributions of active sulfoquinovose-degrading bacteria.

Sulfoquinovose (SQ), a sulfonated sugar produced on a gigaton scale each year, contributes to global sulfur cycling, yet the microbes and pathways mediating its turnover in the environment have been inferred largely from genomic potential rather than direct activity. Here, we coupled incubations of environmental samples with 13C-labeled SQ to DNA-stable isotope probing to identify active SQ carbon assimilators across estuary, mangrove, and lake ecosystems. In estuarine communities, Vibrio and Cognatishimia incorporated SQ-derived 13C; Novosphingobium dominated in the mangrove, and Agrobacterium in the lake. Pure-culture experiments, coupled with comparative proteomics and gene knockout validation, demonstrated that Vibrio strains degrade SQ via modified sulfoglycolytic Embden-Meyerhof-Parnas and Entner-Doudoroff pathways to produce the environmentally significant organosulfur 2,3-dihydroxypropanesulfonate. Comparative genomic analyses suggested that closely related genome representatives of Novosphingobium, Cognatishimia, and Agrobacterium encode the sulfolytic SQ monooxygenase pathway. A global survey of aquatic microbial genomes indicated that over 9% harbor SQ degradation clusters, supporting a widespread distribution of bacterial SQ catabolic potential in aquatic environments.

Relative effects of land use and near-stream chemistry on phosphorus in an urban stream.

Elevated levels of P in urban streams can pose significant water quality problems. Sources of P in urban streams, however, are difficult to identify. It is important to recognize both natural and anthropogenic sources of P. We investigated near-stream chemistry and land use factors on stream water P in the urbanizing Johnson Creek watershed in Portland, OR, USA. We sampled stream water and shallow groundwater soluble reactive P (SRP) and total P (TP) and estimated P flux at 13 sites along the main stem of Johnson Creek, with eight sites in urban land use areas and five sites in nonurban land use areas. At each site, we sampled the A and B horizons, measuring soil pH, water-soluble P, acid-soluble P, base-soluble P, total P, Fe, and Al. We found continuous input of P to the stream water via shallow groundwater throughout the Johnson Creek watershed. The shallow groundwater P concentrations were correlated with stream water P within the nonurban area; however, this correlation was not found in the urban area, suggesting that other factors in the urban area masked the relationship between groundwater P and stream water P. Aluminum and Fe concentrations were inversely correlated with shallow groundwater P, suggesting that greater P adsorption to Al and Fe oxides in the nonurban area reduced availability of shallow groundwater P. Using stepwise multiple regression analysis, however, we concluded that while riparian soil chemistry was related to stream water P, land use patterns had a more significant relationship with stream water P concentrations in this urbanizing system.

Phosphorus load reduction goals for Feitsui Reservoir Watershed, Taiwan.

The present paper describes an effort for developing the total maximum daily load (TMDL) for phosphorus and a load reduction strategy for the Feitsui Reservoir in Northern Taiwan. BASINS model was employed to estimate watershed pollutant loads from nonpoint sources (NPS) in the Feitsui Reservoir watershed. The BASINS model was calibrated using field data collected during a 2-year sampling period and then used to compute watershed pollutant loadings into the Feitsui Reservoir. The simulated results indicate that the average annual total phosphorus (TP) loading into the reservoir is 18,910 kg/year, which consists of non-point source loading of 16,003 kg/year, and point source loading of 2,907 kg/year. The Vollenweider mass balance model was used next to determine the degree of eutrophication under current pollutant loading and the load reduction needed to keep the reservoir from being eutrophic. It was estimated that Feitsui Reservoir can becoming of the oligotrophic state if the average annual TP loading is reduced by 37% or more. The results provide the basis on which an integrated control action plan for both point and nonpoint sources of pollution in the watershed can be developed.
