Modified inter-simple sequence repeat PCR protocol for use in conjunction with the LI-COR Gene ImagIR2 DNA analyzer.

Abstract

Inter-simple sequence repeat (inter-SSR) PCR was assessed for use in variety testing of chrysanthemum. This method was modified to allow detailed analysis of DNA profiles on a LI-COR Gene ImagIR2 DNA analyzer system. Protocols for unlabeled PCR were unsuccessful in producing labeled products when using infrared (IR) dye-labeled primers. Various modifications to the known protocols were investigated: (i) different ratios of labeled to unlabeled primer; (ii) various annealing temperatures; (iii) the use of an IR genotyping kit; (iv) end labeling; and (v) direct incorporation and cycle labeling. Successful amplification using labeled primers only occurred when two consecutive reactions were performed. The first PCR was performed using standard protocols for unlabeled reactions. The second PCR used a dilution of these reaction products as a template and 50% IR-labeled and unlabeled primer. The complete procedure leading to a high-resolution analysis of inter-SSR PCR products on a LI-COR system is reported for the first time. This system allows high-throughput fingerprinting with the potential for applications on a commercial scale.

Triterpenoids from pyrethrum tatsienense.

Two novel and eight known triterpenoids were isolated from alcoholic extract of the whole plant of Pyrethrum tatsienense. The structures of new compounds were elucidated as olean-12-en-3beta,11alpha,16beta-triol-3-O-palmitate (1), ursa-12-en-3beta,11beta,16beta-triol-3-O-palmitate (2) by spectroscopic methods including intensive 2D NMR techniques (H1-H1 COSY, gHMQC, gHMBC) and FAB-MS, and the known compounds were identified on the basis of comparing their NMR data with those of corresponding compounds in the literature.

Xylem hydraulic conductivity related to conduit dimensions along chrysanthemum stems.

The stem xylem conduit dimensions and hydraulic conductivity of chrysanthemum plants (Dendranthema x grandiflorum Tzvelev cv. Cassa) were analysed and quantified. Simple exponential relations describe conduit length distribution, height dependency of conduit length distribution, and height dependency of stem hydraulic conductivity. These mathematical descriptions can be used to model the xylem water transport system. Within a chrysanthemum stem of 1.0 m, the conduit half-length (the length within which 50% of the conduits have their end) was 0.029 m at soil surface and decreased by half at a height of 0.6 m. With each 0.34 m increase in height up the stem, the hydraulic conductivity decreased by 50%. The resistance calculated from conduit lumen characteristics was 70% of the measured resistance. The remaining unexplained part of the hydraulic resistance is at least partly caused by inter-conduit connections.
