Peroxisomal remnant structures in Hansenula polymorpha Pex5 cells can develop into normal peroxisomes upon induction of the PTS2 protein amine oxidase.

Abstract

We have analyzed the properties of peroxisomal remnants in Hansenula polymorpha pex5 cells. In such cells PTS1 matrix protein import is fully impaired. In H. polymorpha pex5 cells, grown on ethanol/ammonium sulfate, conditions that repressed the PTS2 protein amine oxidase (AMO), peroxisomal structures were below the limit of detection. In methanol/ammonium sulfate-grown cells, normal peroxisomes are absent, but a few small membranous structures were observed that apparently represented peroxisomal ghosts since they contained Pex14p. These structures were the target of a Pex10p.myc fusion protein that was produced in pex5 cells under the control of the homologous alcohol oxidase promoter (strain pex5::P(AOX).PEX10.MYC). Glycerol/methanol/ammonium sulfate-grown cells of this transformant were placed in fresh glucose/methylamine media, conditions that fully repress the synthesis of the Pex10p.myc fusion protein but induce the synthesis of AMO. Two hours after the shift Pex10p.myc-containing structures were detectable that had accumulated newly synthesized AMO protein and which during further cultivation developed in normal peroxisomes. Concurrently, the remaining portion of these structures was rapidly degraded. These findings indicate that peroxisomal remnants in pex5 cells can develop into peroxisomes. Also, as for normal peroxisomes in H. polymorpha, apparently a minor portion of these structures actually take part in the development of these organelles.

Polyamine metabolism in sunflower plants under long-term cadmium or copper stress.

The effect of different doses of cadmium and copper was studied in relation to growth and polyamine (Pas) metabolism in shoots of sunflower plants. Cadmium accumulated to higher levels than copper and shoot length was reduced by 0.5 and 1 mM Cd, but only by 1 mM Cu. At 1 mM of Cd or Cu, Put content increased 270% and 160% with Cd2+ and Cu2+, respectively. Spermidine (Spd) was modified only by 1 mM Cd, while spermine (Spm) declined after seeds germinated, increasing thereafter but only with 1 mM Cd or Cu (273% over the controls for Cd and 230% for Cu at day 16). Both ADC and ODC activities were increased by 1 mM Cd, whereas 1 mM Cu enhanced ADC activity, but reduced ODC activity at every concentration used. The role of Pas as markers of Cd or Cu toxicity is discussed.

Absence of tissue-bound semicarbazide-sensitive amine oxidase activity in carp tissues.

We have previously reported that carp (Cyprinus carpio) tissue mitochondria contain a novel form of monoamine oxidase (MAO), which belongs neither to MAO-A nor to MAO-B of the mammalian enzyme. This conclusion results from the findings that the carp MAO was equally sensitive to a selective MAO-A inhibitor clorgyline and to the MAO-B selective inhibitor l-deprenyl, when tyramine, a substrate for both forms, serotonin or beta-phenylethylamine, a substrate for either A or B-form of mammalian MAO, was used. In the present study, we tried to detect another amine oxidase, termed tissue-bound semicarbazide-sensitive amine oxidase (SSAO), activity in carp tissues. As definition of SSAO was used, such as insensitivity to inhibition of the kynuramine oxidizing activity by an MAO inhibitor pargyline and high sensitivity to the SSAO inhibitor semicarbazide. The results indicated that the oxidizing activity was selectively and almost completely inhibited by 0.1 mM pargyline alone or a combination of 0.1 mM pargyline plus 0.1 mM semicarbazide, but not by 0.1 mM semicarbazide alone. We also tried to detect any SSAO activity by changing experimental conditions, such as lower incubation temperature, higher enzyme protein concentration, a lower substrate concentration and different pH's in the reaction, as the enzyme source. However, still no SSAO activity could be detected in the tissues. These results conclusively indicate that carp tissues so far examined do not contain SSAO activity.
