Determination of betamethasone disodium phosphate in the in vitro media of PLGA microspheres by high-performance liquid chromatography.

Betamethasone disodium phosphate is a kind of corticosteroid, which is commonly used in the intralesional treatment of keloids. In this work, polymeric microspheres loaded with betamethasone disodium phosphate were fabricated to design a sustained-release system. A validated HPLC technique for determination of betamethasone disodium phosphate in in vitro media of the polymeric microspheres has been developed. The operation conditions were optimized. The analytical column was ZORBAX Bonus-RP column with 4.6 x 150 mm ID and a particle size of 5 microm. The mobile phase consisted of acetonitrile-0.01% phosphoric acid water solution (40:60, v/v). The flow rate was 1.0 ml/min and injection volume was 50 microl. The elutes were detected at 240 nm. Linearity, repeatability, inter- and intra-assay precision and accuracy of the method were evaluated. The liner range was obtained in a concentrated range of 20-1000 microg/ml, with a coefficient of correlation r=0.999978. The limit of detection for betamethasone disodium phosphate in the in vitro test samples was 0.25 microg. Recovery of betamethasone disodium phosphate from the in vitro test samples was 99.7+/-5.2% (mean+/-SD). Stability of betamethasone disodium phosphate at different pH values and temperatures were also investigated.

Simultaneous determination of terbinafine HCL and triamcinolone acetonide by UV derivative spectrophotometry and spectrodensitometry.

A binary mixture of terbinafine hydrochloride and triamcinolone acetonide was determined by three different methods. The first one concerned with determination of both drugs using first derivative (D(1)) spectrophotometric technique at 297 and 274 nm over concentration ranges of 5-30 and 4-24 microg ml(-1), with mean percentage accuracies 99.90+/-0.67 and 100.25+/-0.49, respectively. The second method depends on ratio-spectra 1st derivative (RSD(1)) spectrophotometry at 298 and 248 nm over the same concentration ranges with mean percentage accuracies 100.22+/-0.51 and 99.93+/-0.56, respectively. The spectrodensitometric analysis provides a rapid and precise method for the separation and quantitation of both terbinafine hydrochloride and triamcinolone acetonide. The method depends on the quantitative densitometric evaluation of thin layer chromatogram of terbinafine hydrochloride and triamcinolone acetonide at 283 and 238 nm over concentration ranges of 5-25 and 2.5-22.5 microg spot(-1), with mean percentage accuracies 100.66+/-0.51 and 100.27+/-0.73, respectively. The suggested procedures were checked using laboratory prepared mixtures and were successfully applied for the analysis of their pharmaceutical preparations. The three methods retained their accuracy and precision when applying the standard addition technique. The results obtained by applying the proposed methods were statistically analysed and compared with those obtained by a reported method.

Arachidonic acid-derived bioactive lipids: their role and the role for their inhibitors in dermatology.

BACKGROUND: In addition to corticosteroids, there are increasing numbers of anti-inflammatory agents that specifically target bioactive lipids generated from arachidonic acid. Knowledge of the diverse mechanisms of action of these different bioactive lipids holds promise in the therapy of a wide spectrum of cutaneous and systemic disorders. OBJECTIVE: Therapeutic manipulations of these lipid molecules through inhibition, stimulation, or direct replacement have broad physiologic effects. These therapeutic strategies not only modulate inflammation, pain, and hemostatic parameters, they also play a role in cardiac, respiratory, renal, and gastrointestinal function and disease, as well as in angiogenesis and in factors that control cell growth and apoptosis important in carcinogenesis. CONCLUSION: Newer drug discovery methods, including combinatorial chemistry with molecular modeling, have made it possible to develop inhibitors and analogs with increasing specificity and bioactivity and decreasing toxicity. Although the application of these analogs and inhibitors for cutaneous disease is limited today, either as primary agents or adjuvant therapy, these drugs will have a place in our therapeutic regimes of the future. We present a review of the therapeutic agents now available from manipulation of these bioactive lipids, and their role and future in dermatology.
