Crystalline solids.

Abstract

Many drugs exist in the crystalline solid state due to reasons of stability and ease of handling during the various stages of drug development. Crystalline solids can exist in the form of polymorphs, solvates or hydrates. Phase transitions such as polymorph interconversion, desolvation of solvate, formation of hydrate and conversion of crystalline to amorphous form may occur during various pharmaceutical processes, which may alter the dissolution rate and transport characteristics of the drug. Hence it is desirable to choose the most suitable and stable form of the drug in the initial stages of drug development. The current focus of research in the solid-state area is to understand the origins of polymorphism at the molecular level, and to predict and prepare the most stable polymorph of a drug. The recent advances in computational tools allow the prediction of possible polymorphs of the drug from its molecular structure. Sensitive analytical methods are being developed to understand the nature of polymorphism and to characterize the various crystalline forms of a drug in its dosage form. The aim of this review is to emphasize the recent advances made in the area of prediction and characterization of polymorphs and solvates, to address the current challenges faced by pharmaceutical scientists and to anticipate future developments.

Colorimetric and fluorimetric methods for determination of panthenol in cosmetic and pharmaceutical formulation.

Two methods are suggested for determination of panthenol. The first is colorimetric method where panthenol is subjected to alkaline hydrolysis; the resulting beta-alanol is allowed to react with vanillin (Duquenois reagent) in presence of McIlvain buffer pH 7.5. The color developed is measured at 406 nm. The linearity range was found to be 50-500 microg/ml while the lower limit of detection was about 10 microg/ml. The second is a sensitive and reliable modified fluorimetric method is also suggested, whereas panthenol, after alkaline hydrolysis is treated with ninhydrin. The fluorescent product was found to have excitation lambda(max) at 385 nm and emission lambda(max) at 465 nm. The method showed high sensitivity with linearity range from 0.01 to 3 microg/ml. The lower limit of detection (LOQ) reached 0.005 microg/ml. Validation of both methods was carried out and the two methods were applied for determination of panthenol in some cosmetic and pharmaceutical formulations.

In vitro release properties of etonogestrel and ethinyl estradiol from a contraceptive vaginal ring.

The release properties of steroids from a combined contraceptive vaginal ring have been investigated. The product design is based on a coaxial fiber consisting of two types of polyethylene vinylacetate copolymers. Inside the core of the fiber, two steroids are present in a molecularly dissolved state. In order to design a controlled release system with specified release characteristics, values of diffusion coefficient and solubility are required. These data can either be determined during pre-formulation studies on e.g. polymeric flat films or from in-vitro release measurements of the actual coaxial fibers. It can be concluded from this study that polyethylene vinylacetate copolymers exhibit suitable properties to develop a controlled release system with the two steroids etonogestrel and ethinyl estradiol. It has been found that the permeability data obtained in the pre-formulation studies are useful in semi-quantitative terms, but deviate from the permeability data found from the in-vitro release of coaxial fibers. This is most likely due to differences in the polymeric structure of films and coaxial fibers. As a consequence, further studies should be initiated to evaluate the relationship between the manufacturing process and the resulting polymeric structure. It has also been found that the solubility and release of etonogestrel are influenced by the concentration of ethinyl estradiol. By investigating this phenomenon by thermoanalysis, it was shown that the steroids form an eutectic. The lower melting point of the steroids results in an increase in solubility and hence in altered permeability properties.

Nanosized cationic hydrogels for drug delivery: preparation, properties and interactions with cells.

A new family of nanoscale materials on the basis of dispersed networks of cross-linked ionic and nonionic hydrophilic polymers is being developed. One example is the nanosized cationic network of cross-linked poly(ethylene oxide) (PEO) and polyethyleneimine (PEI), PEO-cl-PEI nanogel. Interaction of anionic amphiphilic molecules or oligonucleotides with PEO-cl-PEI results in formation of nanocomposite materials in which the hydrophobic regions from polyion-complexes are joined by the hydrophilic PEO chains. Formation of polyion-complexes leads to the collapse of the dispersed gel particles. However, the complexes form stable aqueous dispersions due to the stabilizing effect of the PEO chain. These systems allow for immobilization of negatively charged biologically active compounds such as retinoic acid, indomethacin and oligonucleotides (bound to polycation chains) or hydrophobic molecules (incorporated into nonpolar regions of polyion-surfactant complexes). The nanogel particles carrying biological active compounds have been modified with polypeptide ligands to enhance receptor-mediated delivery. Efficient cellular uptake and intracellular release of oligonucleotides immobilized in PEO-cl-PEI nanogel have been demonstrated. Antisense activity of an oligonucleotide in a cell model was elevated as a result of formulation of oligonucleotide with the nanogel. This delivery system has a potential of enhancing oral and brain bioavailability of oligonucleotides as demonstrated using polarized epithelial and brain microvessel endothelial cell monolayers.
