Imaging of the fluorescence spectrum of a single fluorescent molecule by prism-based spectroscopy.

We have devised a novel method to visualize the fluorescence spectrum of a single fluorescent molecule using prism-based spectroscopy. Equipping a total internal reflection microscope with a newly designed wedge prism, we obtained a spectral image of a single rhodamine red molecule attached to an essential light chain of myosin. We also obtained a spectral image of single-pair fluorescence resonance energy transfer between rhodamine red and Cy5 in a double-labeled myosin motor domain. This method could become a useful tool to investigate the dynamic processes of biomolecules at the single-molecule level.

Superquenching in cyanine pendant poly(L-lysine) dyes: dependence on molecular weight, solvent, and aggregation.

A series of poly(L-lysines) ranging in number of repeat units (N(PRU)) from 6 to 900 has been synthesized and the photophysics of the series and monomer cyanine dye have been studied in solution. In water or aqueous dimethyl sulfoxide, the oligomers and polymers exhibit high sensitivity to fluorescence quenching by oppositely charged electron acceptors; in this study, 9,10-anthraquinone-2,6-disulfonate was used as a quencher for the cationic fluorescent polyelectrolytes. Quenching constants (K(SV)) measured in 50:50 (v/v) dimethyl sulfoxide-water increase monotonically with increase in N(PRU) ranging from 630 M(-1) for monomer to 1.2 x 10(9) M(-1) for dilute solutions of the polymer having N(PRU) approximately 900. The polymers having N(PRU) > 100 exhibit predominantly J-aggregate absorption and fluorescence and enhanced susceptibility to quenching. For the polymers exhibiting strong J-aggregation, the effective exciton domain quenched by a single quencher reaches approximately 100 PRU. The results of this study permit a semiquantitative analysis of superquenching of fluorescent polyelectrolytes in solution and the factors that control it.

Morphology and dynamics of chromosome territories in living cells.

Chromosome territories formed by fluorescence-labeled sub-chromosomal foci were analyzed in time-lapse series of 3D confocal data sets of living HeLa and human neuroblastoma cells. The quantitative analysis of the chromosome territory morphology confirmed previous results obtained by visual observation [Zink et al., Hum. Genet. 102 (1998) 241-251] that chromosome territories persisted as stable entities over an observation time >4 h. The changes in morphology with time of single chromosome territories were found to be less pronounced than differences in morphology of different chromosome territories in fixed cells. The analysis of the individual motion of chromosome territories recently showed 'Brownian' diffusion-like motion at very slow rates [Bornfleth et al., Biophys. J. 77 (1999) 2871-2886]. Here, we show that the mutual motion of different chromosome territories was independent and also 'Brownian' diffusion-like.
