Development of a new methanethiol quantification method using ethanethiol as an internal standard.

Abstract

Development of a new method to quantify methanethiol in which ethanethiol was employed as an internal standard is reported. Recovery yields for methanethiol from an aqueous model system and a soy protein concentrate (SPC) aqueous slurry determined with this method ranged from 97 to 107% and from 103 to 121%, respectively. The methanethiol content of two commercial SPCs and two commercial soy protein isolate (SPI) samples, on a dry basis, ranged from 835 to 1190 times greater than the odor threshold for methanethiol. Relative standard deviations for quantifying methanethiol with the method from these samples were <5%, indicating its good reproducibility in quantifying methanethiol from soy protein products. Also investigated in the current study was the feasibility of using 5',5-dithiobis(2-nitrobenzoic acid) (DTNB, Ellman's reagent) to determine the concentration of methanethiol in the aqueous solutions used to prepare the standard curve for quantifying methanethiol from soy protein products. The concentrations of methanethiol obtained from Ellman's reagent method were comparable with those from a weighing method and theoretical calculation.

Identification of a stable flavin-thiolate adduct in heterotetrameric sarcosine oxidase.

Heterotetrameric sarcosine oxidase (TSOX) is a complex bifunctional flavoenzyme that contains two flavins. Most of the FMN in recombinant TSOX is present as a covalent adduct with an endogenous ligand. Enzyme denaturation disrupts the adduct, accompanied by release of a stoichiometric amount of sulfide. Enzyme containing>or=90% unmodified FMN is prepared by displacement of the endogenous ligand with sulfite, a less tightly bound competing ligand. Reaction of adduct-depleted TSOX with sodium sulfide produces a stable complex that resembles the endogenous TSOX adduct and known 4a-S-cysteinyl flavin adducts. The results provide definitive evidence for sulfide as the endogenous TSOX ligand and strongly suggest that the modified FMN is a 4a-sulfide adduct. A comparable reaction with sodium sulfide is not detected with other flavoprotein oxidases. A model of the postulated TSOX adduct suggests that it is stabilized by nearby residues that may be important in the electron transferase/oxidase function of the coenzyme.

Polymeric vesicle permeability: a facile chemical assay.

We present a simple method to characterize vesicles and determine, at the same time, the membrane permeability to specific molecules. The method is based on encapsulating highly hydrophilic 3,3',3' '-phosphinidynetris-benzenesulfonic acid (PH) into vesicles and subsequently monitoring its reaction with 5,5'-dithiobis-2-nitrobenzoic acid (DTNB). We tested the method by measuring the membrane permeability of vesicles formed from a series of poly(ethylene oxide)-co-polybutylene oxide (EB) copolymers and egg yolk phosphatidylcholine. We found that the experimental data are in good agreement with calculations based on Fick's first law. We therefore quantified the DTNB permeability across vesicle membranes, finding that polymeric EB membranes have a more selective permeability toward polar molecules compared to phospholipids membranes.

[Determination of human serum glutathione peroxidase activity, by using hydrogen peroxide and 5,5'-dithio-bis(2-nitrobenzoic acid)].

The method has been developed for the assay of human serum glutathione:H2O2-oxidoreductase activity. The reaction medium comprises tris-HCl buffer (0.04 M; pH 8.5), 4.4 mM EDTA, 10 mM NaN3, 1 mM GSH, 400 microM Y2O2. The incubation time of 60 seconds at 37 degrees C was chosen. There was a decrease in GSH in the reaction with 5,5'-dithio-bis(2-nitrobenzoic acid). This method is recommended for the application to the assay of selenium-dependent glutathione peroxidase because hydrogen peroxide (H2O2) used in this method is a more specific substrate than commonly used organic hydroperoxides.
