Construction of cDNA representational difference analysis based on two cDNA libraries and identification of garlic inducible expression genes in human gastric cancer cells.

Abstract

AIM: To elucidate molecular mechanism of chemopreventive efficacies of garlic against human gastric cancer (HGC). METHODS: HGC cell line BGC823 was treated with Allitridi (a kind of garlic extract) and Allitridi-treated and parental BGC823 cDNA libraries were constructed respectively by using lambdaZAP II vector. cDNA Representational Difference Analysis (cDNA RDA) was performed using Bam H I cutting-site and abundant cDNA messages provided by the libraries. Northern blot analysis was applied to identify the obtained difference products. RESULTS: Two specific cDNA fragments were obtained and characterized to be derived from homo sapiens folate receptor alpha (FRalpha) gene and calcyclin gene respectively. Northern blot results showed a 4-fold increase in FRalpha gene expression level and 9-fold increase in calcyclin mRNA level in BGC823 cells after Allitridi treatment for 72h. CONCLUSION: The method of cDNA RDA based on cDNA libraries combines the high specificity of cDNA RDA with abundant cDNA messages in cDNA library; this expands the application of cDNA library and increases the specificity of cDNA RDA. Up-regulation of FRalpha gene and calcyclin gene expressions induced by Allitridi provide valuable molecular evidence for the efficacy garlic in treating HGC as well as other diseases.

Total synthesis of flustramine C via dimethylallyl rearrangement.

The marine natural product flustramine C from the bryozoan Flustra foliacea was synthesized in five steps and 38% yield starting from Nb-methyltryptamine. The key step is the biomimetic oxidation of the natural product deformylflustrabromine causing selective 1,2-rearrangement of the inverse prenyl group. By 1H,15N HMBC experiments, it is unambiguously shown that the reaction with t-BuOCl commences with chlorination of the side chain nitrogen. Deformylflustrabromine itself was synthesized via Danishefsky inverse prenylation. [reaction: see text].

Tandem oxidation of allylic and benzylic alcohols to esters catalyzed by N-heterocyclic carbenes.

N-heterocyclic carbenes catalyze the oxidation of allylic, propargylic, and benzylic alcohols to esters with manganese(IV) oxide in excellent yields. A variety of ester derivatives can be synthesized, including protected carboxylates. This one-pot tandem oxidation represents the first organocatalytic oxidation of alcohols to esters. Saturated esters can also be accessed from aldehydes using this method. Through the utilization of a chiral catalyst, the acyl-heteroazolium intermediate becomes a chiral acylating agent, which can desymmetrize meso-1,2-diols. [reaction: see text].

Surface seals reduce 1,3-dichloropropene and chloropicrin emissions in field tests.

Reducing emissions is essential for minimizing the impact of soil fumigation on the environment. Water application to the soil surface (or water seal) has been demonstrated to reduce 1,3-dichloropropene (1,3-D) emissions in soil column tests. This study determined the effectiveness of water application to reduce emissions of 1,3-D and chloropicrin (CP) in comparison to other surface seals under field conditions. In a small-plot field trial on a Hanford sandy loam soil (coarse-loamy, mixed, superactive, nonacid, thermic Typic Xerorthents) in the San Joaquin Valley, CA. Telone C35 (61% 1,3-D and 35% CP) was shank-applied at a depth of 46 cm at a rate of 610 kg ha-1. Soil surface seal treatments included control (no tarp and no water application), standard high density polyethylene (HDPE) tarp over dry and pre-irrigated soil, virtually impermeable film (VIF) tarp, initial water application by sprinklers immediately following fumigation, and intermittent water applications after fumigation. The atmospheric emissions and gas-phase distribution of fumigants in soil profile were monitored for 9 d. Among the surface seals, VIF and HDPE tarp over dry soil resulted in the lowest and the highest total emission losses, respectively. Intermittent water applications reduced 1,3-D and CP emissions significantly more than HDPE tarp alone. The initial water application also reduced emission peak and delayed emission time. Pre-irrigated soil plus HDPE tarp reduced fumigant emissions similarly as the intermittent water applications and also yielded the highest surface soil temperature, which may improve overall soil pest control.
