Totally synthetic medium for susceptibility testing.

Abstract

The results of disk diffusion and plate dilution susceptibility testing of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Proteus mirabilis on media gelled with agar-agar or with a synthetic hydrogel were compared. Synthetic hydrogel can be combined with a totally defined synthetic amino acid medium to yield a reproducible, totally defined, synthetic solid medium without the antagonistic or booster effects of some components of agar. Such a medium could be used as a reference medium for susceptibility testing.

Karyotype instability and anchorage-independent growth in telomerase-immortalized fibroblasts from two centenarian individuals.

Several reports have shown that the ectopic expression of the human telomerase catalytic subunit gene (hTERT) leads to an indefinite extension of the life span of human fibroblasts cultured in vitro without the appearance of cancer-associated changes. We infected two fibroblast strains derived from centenarian individuals with an hTERT containing retrovirus and isolated transduced massive populations (cen2tel and cen3tel). In both populations, hTERT expression reconstituted telomerase activity and extended the life span. In cen2tel, a net telomere lengthening was observed while, in cen3tel, telomeres stabilized at a length lower than that detected in senescent parental cells. Interestingly, both cen2tel and cen3tel cells developed chromosome anomalies, numerical first and structural thereafter. Moreover, cen3tel cells acquired the ability to grow in the absence of solid support, a typical feature of transformed cells. The results we present here highlight an unexpected possible outcome of cellular immortalization driven by telomerase reactivation, and indicate that, in some cases, an artificial extension of cellular replicative capacity can increase the probability of occurrence of genomic alterations, which can lead to cellular transformation.

CHROMagar Candida medium as a practical tool for the differentiation and presumptive identification of yeast species isolated from salads.

CHROMagar Candida medium was used to study the diversity of yeast biota of salad samples, and to presumptively identify the isolates. This medium was originally developed for the selective isolation and presumptive identification of some clinically important yeast species such as Candida albicans, Candida tropicalis, Candida krusei, and Candida glabrata on the basis of differences in colour and surface of colonies. Ninety three yeast strains representing 33 species from the culture collection and 39 fresh isolates from different mayonnaise-based mixed salads showed a wide range of hue of colony colours ranging from white to yellow, orange, red, pink, purple, blue, green, etc., as well as different morphological appearances on the CHROMagar Candida medium. Therefore, CHROMagar Candida medium facilitates the detection of mixtures of yeast species from different samples on a single isolation plate and this medium can be a practical method for the differentiation and rapid presumptive identification of many yeast species occurring frequently in different kind of foods.
