An acid-catalyzed macrolactonization protocol.

Abstract

[reaction: see text] An efficient macrolactonization protocol devoid of any base was developed derived from the use of vinyl esters in transesterification. Subjecting a hydroxy acid and ethoxyacetylene to 2 mol % [RuCl(2)(p-cymene)](2) in toluene followed by addition of camphorsulfonic acid or inverse addition provided macrolactones in good yields.

LC determination of impurities in azithromycin tablets.

A LC method with UV detection for determining azithromycin impurities in tablets as pharmaceutical form has been developed. It is to be employed in routine and stability tests. A linear gradient elution was employed starting with 47% A and 53% B to reach 28% A and 72% B at 48 min. Mobile phase A was KH2PO4 10 mM (H2O) at pH 7.00. B was a mixture methanol:acetonitrile 1:1 (v/v). UV detection was performed at 210 nm. The chromatographic column was Phenomenex Synergi MAX-RP 4 microm 250 x 460 mm kept at 50 degrees C. Six impurities were separated and identified and it was possible to quantify five out of the six with reasonable accuracy and precision.

A review of the different techniques for solid surface acid-base characterization.

In this work, various techniques for solid surface acid-base (AB) characterization are reviewed. Different techniques employ different scales to rank acid-base properties. Based on the results from literature and the authors' own investigations for mineral oxides, these scales are compared. The comparison shows that Isoelectric Point (IEP), the most commonly used AB scale, is not a description of the absolute basicity or acidity of a surface, but a description of their relative strength. That is, a high IEP surface shows more basic functionality comparing with its acidic functionality, whereas a low IEP surface shows less basic functionality comparing with its acidic functionality. The choice of technique and scale for AB characterization depends on the specific application. For the cases in which the overall AB property is of interest, IEP (by electrokinetic titration) and H(0,max) (by indicator dye adsorption) are appropriate. For the cases in which the absolute AB property is of interest such as in the study of adhesion, it is more pertinent to use chemical shift (by XPS) and the heat of adsorption of probe gases (by calorimetry or IGC).

Immunocytochemical identification of type I collagen in acid-etched dentin.

The hypothesis tested in this study is that the application of phosphoric acid prevents collagen fibrils (CF) from maintaining their structural morphology, as assessed by their immunochemical antigenicity. Dentin was conditioned with EDTA and with 35% H3PO4 for 15, 30, and 60 s. For a control there was no treatment. A pre-embedding immunohistochemical procedure was evaluated under high-resolution SEM and a postembedding immunolocalization technique was performed on ultrathin sections at TEM level. Conditioning with EDTA resulted in a weaker labeling signal than for dentin conditioned with H3PO4. The most intense labeling was observed after dentin had been etched with H3PO4 for 15 s. The least intensive labeling was evident when the acid was applied for 30 s or for 60 s. A very weak signal was detected on the untreated dentin surface. These results provide evidence that a 15-s application of 35% H3PO4 causes mineral dissolution of the crystals enveloping the superficial CF without damaging the ultrastructure of the CF while longer applications of 35% H3PO4 cause alterations in the CF that decrease their antigenicity.
