Through-shell alkyllithium additions and borane reductions.

Abstract

The through-shell borane reduction and methyllithium addition to benzaldehyde (1), benzocyclobutenone (2), and benzocyclobutenedione (3) incarcerated inside a hemicarcerand (4) with four tetramethylenedioxy bridges are reported. All guests could be reduced and methylated. Selective monoreduction and monomethylation were observed for 3. In the methyllithium addition to 4[symbol: see text]3, the initially formed lithium alcoholate underwent a Moore rearrangement. The reactivity of the incarcerated guests toward methyllithium increased in the order 1 < 2 << 3 and toward borane in the order 1 << 2 approximately equal 3. Guest reactivity was correlated with the inner-phase location of the reacting carbonyl group in the preferred guest inner-phase orientation. The latter was determined from the X-ray structures of 4[symbol: see text]1, 4[symbol: see text]2, and 4[symbol: see text]3, from molecular mechanical calculations, and from the hemicarcerand-induced upfield shift of the guest proton resonances. In the methyllithium and n-butyllithium addition to 4[symbol: see text]1 and 4[symbol: see text]3 at elevated temperatures, selective cleavage of a host's spanner or tetramethylenedioxy bridge, respectively, was observed. The cleavage of one spanner also took place in the methyllithium addition to the 1-methyl-2-pyrrolidinone hemicarceplex. These scission reactions are initiated by the initially formed lithium alcoholates, which show enhanced basicity and nucleophilicity in the inner phase as compared to the bulk phase. Mechanisms for the host scission reactions are discussed.

Polar group enhanced gas-phase acidities of carboxylic acids: an investigation of intramolecular electrostatic interaction.

We studied the effects of polar groups on the gas-phase acidities of carboxylic acids experimentally and computationally. In this connection, the gas-phase acidities (DeltaH(acid), the enthalpy of deprotonation, and DeltaG(acid), the deprotonation free energy) of borane-complexed methylaminoacetic acid ((CH(3))2N(BH(3))CH(2)CO(2)H) and methylthioacetic acid (CH(3)S(BH(3))CH(2)CO(2)H) were measured using the kinetic method in a flowing afterglow-triple quadrupole mass spectrometer. The values of DeltaH(acid) and DeltaG(acid) of (CH(3))2N(BH(3))CH(2)CO(2)H were determined to be 328.8 +/- 1.9 and 322.1 +/- 1.9 kcal/mol, and those of CH(3)S(BH(3))CH(2)CO(2)H were determined to be 325.8 +/- 1.9 and 319.2 +/- 1.9 kcal/mol, respectively. The theoretical enthalpies of deprotonation of (CH(3))2N(BH(3))CH(2)CO(2)H (329.2 kcal/mol) and CH(3)S(BH(3))CH(2)CO(2)H (325.5 kcal/mol) were calculated at the B3LYP/6-31+G(d) level of theory. The calculated enthalpies of deprotonation of N-oxide-acetic acid (CH(3)NOCH(2)CO(2)H, 329.4 kcal/mol) and S-oxide-acetic acid (CH(3)SOCH(2)CO(2)H, 328.6 kcal/mol) are comparable to the experimental results for borane-complexed methylamino- and methylthioacetic acids. The enthalpy of deprotonation of sulfone-acetic acid (CH(3)SO2CH(2)CO(2)H, 326.1 kcal/mol) is about 2 kcal/mol lower than the S-oxide-acetic acid. The calculated enthalpy of deprotonation of sulfoniumacetic acid, (CH(3))2S+CH(2)CO(2)H, is 243.0 kcal/mol. Compared to the corresponding reference molecules, CH(3)NHCH(2)CO(2)H and CH(3)SCH(2)CO(2)H, the dipolar group and the monopolar group substituted carboxylic acids are stronger acids by 11-14 and 97 kcal/mol, respectively. We correlated the changes of the acidity upon a polar group substitution to the electrostatic free energy within the carboxylate anion. The acidity enhancements in polar group substituted carboxylic acids are the results of the favorable electrostatic interactions between the polar group and the developing charge at the carboxyl group.

Solution behavior of a sugar-based carborane for boron neutron capture therapy: a nuclear magnetic resonance investigation.

The physico-chemical properties of beta-lactosyl-closo-orthocarborane in water solution were investigated by multinuclear NMR, (13)C NMR relaxation, and ab-initio calculations. This molecule represents a potentially selective boron carrier in Boron Neutron Cancer Therapy (BNCT) and exhibits amphiphilic characteristics. Its structural and dynamic features were studied comparing NMR data acquired in both aggregating and non-aggregating conditions. Aggregates are characterized by rapid exchange with the bulk and by high sensitivity to temperature conditions. An unusually stable intra-molecular CH...O hydrogen bond was found to persist in water solution both for the free molecules and after aggregate formation. At the same time, inter-molecular specific CH()O interactions do not seem effective in the aggregate formation process, which appears to take place only on non-specific hydrophobic basis.

Nucleophilic substitution at the anomeric position of 1,2-O-isopropylidenefuranose derivatives. A novel stereoselective synthesis of cyclic phosphates analogous to cAMP.

1,2-O-Isopropylidenefuranose derivatives were treated with various nucleophiles in the presence of either BF(3).OEt(2) or trimethylsilyl trifluoromethanesulfonate (TMSOTf) leading to substitution products in a regio- and stereoselective manner. In particular, nucleophilic substitution of 1,2-O-isopropylidenefuranose derivatives when treated with allyltrimethylsilane was controlled by steric and electronic factors (similar to Woerpel's stereoelectronic model). On the other hand, when 1,2-O-isopropylidenefuranose derivatives were treated with trimethylsilane, in the presence of bis-O-trimethylsilyl-5-iodouracil or bis-O-trimethylsilyl-thymidine, substitution products were generated in high regio- and stereoselectivities via an unusual nucleophilic substitution with opening of the furanose ring. Based on these results, a stereoselective method for the synthesis of neutral cyclic phosphates analogous to cAMP was developed.
