Histological study of post-pollination events in Spathodea campanulata beauv. (Bignoniaceae), a species with late-acting self-incompatibility.

The reproductive biology of Spathodea campanulata was investigated by means of hand-pollination experiments, observations of pollen tube growth using fluorescence microscopy, and serial sections of ovules in selfed and crossed pistils. Only cross-pollinated flowers developed fruits, and all selfed flowers abscised within 3-4 d. However, self pollen tubes grew successfully to the ovary, penetrating and fertilizing the majority of ovules by 48 h, indicating that S. campanulata is a species with late-acting self-incompatibility. The incidences of ovule penetration, fertilization and endosperm initiation were all significantly slower in selfed vs. crossed pistils, although no other signs of malfunctioning were detected. The possible role of such slow self pollen tube effectiveness as a recognition event is discussed within the context of the slow but not entirely suppressed self pollen tube growth reported for some species with conventional homomorphic self-incompatibility.

Isolation and identification of four flavonoid constituents from the seeds of Oroxylum indicum by high-speed counter-current chromatography.

Four flavonoids, chrysin, baicalein, baicalein-7-O-glucoside, baicalein-7-O-diglucoside (Oroxylin B) and one unknown flavonoid have been isolated and purified for the first time in the seeds of Oroxylum indicum by high-speed counter-current chromatography with a two-phase solvent system composed of chloroform-methanol-water (8:10:5, v/v). Then, 50 mg baicalein-7-O-glucoside, 10.5 mg baicalein-7-O-diglucoside, 4.5 mg chrysin-7-O-diglucoside, 25 mg baicalein and 45 mg chrysin could be obtained after injecting 20 mg/ml sample extract ten times and their purities were 96, 90, 85, 95 and 98%, respectively. All these constituents were identified by high-performance liquid chromatography-mass spectrometry and nuclear magnetic resonance.

Micropropagation of Anemopaegma arvense: conservation of an endangered medicinal plant.

Anemopaegma arvense (Vell.) Stellfeld ex de Souza is a medicinal species also known as Catuaba. All commercially available formulations of Catuaba have been produced from wild crafting, the gathering of plant biomass from its native habitat. Nodal segments were used as explants and 4.8 new buds per explant were induced on MS media supplemented with 4.4 microM of kinetin in 30 days. The percentage of in vitro rooting was low, although acclimatization of unrooted plants into soil was successfully achieved. For germplasm maintenance, cultures of A. arvense were stored on 4 % (w/v) sorbitol and maintained viable at low growth rate without subcultures for six months.
