Structural and physicochemical characterisation of branched dextrans produced by an active α-(1→2) branching sucrase from Apilactobacillus kunkeei PDER37.

Recently, branching sucrases encoded in the genomes of certain Lactic Acid Bacteria (LAB) strains have become novel enzymes to obtain branched α-glucans. In this study an active α-(1 → 2) branching sucrase from Apilactobacillus kunkeei PDER37 was expressed, characterised and distinct branched dextrans was obtained with reactions under different sucrose: dextran ratio. Structural characterisation by 1H and 13C NMR analysis demonstrated the branching of the dextran with (1 → 2)-linked α-d-glucose units with no alteration in the final structure depending on sucrose: dextran ratio (D0) but this ratio was effective for the determination of the molecular weights of the branched dextrans (D1, D2 and D3). FTIR analysis further supported the dextran structures and suggested the higher accumulation of the α-Glc units in the branched dextrans. Thermal characterisation of the branched dextrans obtained by TGA and DSC analysis suggested the increased hygroscopicity of the branching units. Both SEM and AFM analysis demonstrated more porous chain like structures in the branched dextrans. This study provides valuable information on the role of active α-(1 → 2) branching sucrase (BS37) for the production of branched dextrans with potential increased physicochemical status applicable for food and other industries.

Partitioning behavior of silica in the Triton X-100/dextran/water aqueous biphasic system.

The partitioning behavior of silica particles was investigated in the Triton X-100/dextran/water system. It was found that both electrostatic effects and interactions between phase-component species and the solid surface played important roles in determining the distribution of solids. Silica partition was highly pH-dependent, which was interpreted in terms of the pH dependence of the Triton X-100/SiO(2) interaction and the weak acidity of dextran. The presence of sodium dodecyl sulfate (SDS) moved the particles from the top surfactant-rich phase to the interface and the bottom phase, while dodecyltrimethylammonium bromide (DTAB) had the opposite effect. These trends are attributable to the electrostatic interaction between the charged mixed micelles in the top phase and the particles and to the fact that the ionic surfactants modified the adsorption density of the nonionic surfactant on silica.

Preparation and characterization of polybutylcyanoacrylate magnetic nanoparticles.

OBJECTIVE: To Prepare and characterize polybutylcyanoacrylate (PBCA) magnetic nanoparticles. METHODS: The suspension of PBCA magnetic nanoparticles was prepared by emulsion polymerization method. The particle size, size distribution, polydispersity and other parameters were determined by photon correlation spectroscopy and transmission electron microscopy, and the iron content in the nanoparticles by ultraviolet spectroscopy. RESULTS: The data of the particle size and size distribution of aqueous decanoic acid stabilized Fe3O4 magnetic fluids were acquired, and the influences of concentrations of butylcyanoacrylate (BCA) and Dextran-70 on the particle size and size distribution of PBCA magnetic nanoparticles were characterized. CONCLUSION: We have for the first time successfully prepared stable suspension of PBCA magnetic nanoparticles by emulsion polymerization method at pH 6.3-6.4 in aqueous medium.
