A channel model with fluctuating barrier structures.

Abstract

Following the theory 'Fluctuations of barrier structure in ionic channels' (Läuger, P., Stephan, W. and Frehland, E. (1980) Biochim. Biophys. Acta 602, 167-180), we constructed a model of a channels with several conformational states. The origin of these conformational states and the source for the transitions from one to the other are given explicitly for the presented model. In this work the effect of multiple conformational states on the ion transport process is analyzed. We considered a channel protein with two main barriers and one binding site. The site is surrounded by dipolar groups. The dipole moment of these groups can be reoriented by thermal activity and also by electrical interaction with the transported ions. Differently polarized states generate different activation energy barriers for the ions. The set of conformational states of the channel is constituted by all the possible polarized states of the binding site. Using the rate-theory analysis of ion transport (Glässtone, S., Laider, K.J. and Eyring, H. (1941) The theory of rate processes, McGraw-Hill, New York), the possible coupling between ion flux and the channel conformational transitions has been incorporated into the model by considering the dependence of the rate constants on the heights of the energy barriers. The resulting multistate kinetic equations have been solved numerically. It was shown that the simple saturation characteristic of the flux-concentration curve was obtained. For certain values of the model parameters, the channel shows a strongly different conductance for anions compared to cations. In fact, the model contains an interesting mechanism that exhibits selectivity with respect to the charge of the ions.

Charge inversion at high ionic strength studied by streaming currents.

We report charge inversion, the sign reversal of the effective surface charge in the presence of multivalent counterions, for the biologically relevant regimes of divalent ions and mixtures of monovalent and multivalent ions. Using streaming currents, the pressure-driven transport of countercharges in the diffuse layer, we find that charge inversion occurs in rectangular silica nanochannels at high concentrations of divalent ions. Strong monovalent screening is found to cancel charge inversion, restoring the original surface charge polarity. An analytical model based on ion correlations successfully describes our observations.

Synthetic ion channels and pores (2004-2005).

This critical review covers synthetic ion channels and pores created between January 2004 and December 2005 comprehensively. The discussion of a rich collection of structural motifs may particularly appeal to organic, biological, supramolecular and polymer chemists. Functions addressed include ion selectivity and molecular recognition, as well as responsiveness to light, heat, voltage and membrane composition. The practical applications involved concern certain topics in medicinal chemistry (antibiotics, drug delivery), catalysis and sensing. An introduction to principles and methods is provided for the non-specialist; some new sources of inspiration from fields beyond chemistry are highlighted.

Affinity of 1-aryl-1,2,3,4-tetrahydroisoquinoline derivatives to the ion channel binding site of the NMDA receptor complex.

A series of 1-aryl-1,2,3,4-tetrahydroisoquinoline and 8-methyl-1-aryl-1,2,3,4-tetrahydroisoquinoline derivatives was evaluated for affinity to the PCP binding site of the NMDA receptor complex. The (S)-configured tetrahydroisoquinoline derivative (S)-4 e x HCl bearing a 2-methylphenyl substituent in position 1 of the heterocyclic ring system and a methyl group in position 8 was found to exhibit the highest affinity among the derivatives with a K(i)-value of 0.0374 microM. In addition, this compound shows a remarkable enantioselectivity of binding by being almost 90 times more potent than the corresponding (R)-enantiomer (R)-4 e x HCl. Additionally, a convenient and efficient synthetic approach to racemic 1-aryl-1,2,3,4-tetrahydroisoquinoline derivatives is described.
