Chlropyrifos-methyl shows anti-androgenic activity without estrogenic activity in rats.

Chlorpyrifos-methyl (CPM), an organophosphate insecticide, widely used for grain storage and agriculture, has been suspected as endocrine disrupter by a few in vitro studies. This study was performed to investigate the (anti-) estrogenicity and (anti-) androgenicity of CPM in vivo using immature rat uterotrophic assay and rat Hershberger assay. CPM with or without 17beta-estradiol were administered to 20 days old female rats to investigate its (anti-) estrogenic activity. Uterine and vaginal weight, uterine epithelial cell height were not affected by the treatment of CPM (2, 10, 50, 250 mg/kg). CPM 250 mg/kg potentiated relative vagina weight in 17beta-estradiol treated immature female rats without any changing of uterine weight. Relative liver weight was increased with decrease of body weight by CPM 250 mg/kg treatment. Uterine cell proliferation tested with bromodeoxyuridine labeling index was not observed in CPM treated rats. CPM with or without testosterone propionate were administered to castrated rat of 51 days old for 10 days to investigate the (anti-)androgenic activity,. The weight of relative and absolute androgen-dependent accessory sex organs; seminal vesicle with coagulating glands (SV/CG), ventral prostate gland (VP), glans penis (GP), levator ani plus bulbocarvernosus muscle (LABC) and Cowper's gland (CG,) were unchanged by the treatment of CPM alone. While CPM induced the increase of relative adrenal gland weight, CPM 50mg/kg decreased the weights of CV/CG, VP, CG and LABC without change of GP without changing of GP when it was treated with TP. In conclusion, CPM dose not show estrogenic and anti-estrogenic activity in immature female rats, but it represents anti-androgenic activity by inhibition of the TP-stimulated increase of the weight of accessory sex organs.

Central and peripheral interactions between the agouti-related protein and leptin.

The agouti-related protein (AgRP) is a powerful appetite modulator expressed in the hypothalamus and the adrenal gland and regulated by leptin. Here we report the robust expression of AgRP in epididymal fat and its upregulation in this tissue by feeding rather than by fasting. This was observed in both the obesity-susceptible C57BL/6J and the obesity-resistant CAST/Ei mouse strains. Surprisingly, AgRP expression was higher in the hypothalamus and the adrenal gland in the leaner and obesity-resistant CAST/Ei strain. In vitro leptin treatment upregulated endogenous AgRP in mouse hypothalamus and adrenal cells, after an acute 6-h exposure, but it downregulated AgRP after a long-term 60-h exposure. AgRP, on the other hand, upregulated its own endogenous expression in the hypothalamus and the adrenal cells and also upregulated endogenous leptin in the adrenal cells. These results reveal a novel feedback loop and reciprocal transcriptional regulation between AgRP and leptin centrally and peripherally.

Protein preparation, crystallization and preliminary X-ray analysis of human adrenal gland protein AD-004.

The adrenal gland protein AD-004 was identified in the human adrenal gland. Full-length AD-004 contains 172 amino acids, with a predicted molecular weight of about 20 kDa. In attempts to crystallize human AD-004, the gene was subcloned into a modified pET vector, pET21-DEST, with an N-terminal His(5) tag using the Gateway cloning system, followed by protein expression in Escherichia coli strain BL21(DE3). The protein was purified in two steps to near-homogeneity and was crystallized. The crystals belong to space group P6(1) or P6(5), with unit-cell parameters a = b = 99.56, c = 57.19 A. A complete 2.0 A data set has been collected at a rotating-anode X-ray source and structure determination is under way.
