Chlropyrifos-methyl shows anti-androgenic activity without estrogenic activity in rats.

Chlorpyrifos-methyl (CPM), an organophosphate insecticide, widely used for grain storage and agriculture, has been suspected as endocrine disrupter by a few in vitro studies. This study was performed to investigate the (anti-) estrogenicity and (anti-) androgenicity of CPM in vivo using immature rat uterotrophic assay and rat Hershberger assay. CPM with or without 17beta-estradiol were administered to 20 days old female rats to investigate its (anti-) estrogenic activity. Uterine and vaginal weight, uterine epithelial cell height were not affected by the treatment of CPM (2, 10, 50, 250 mg/kg). CPM 250 mg/kg potentiated relative vagina weight in 17beta-estradiol treated immature female rats without any changing of uterine weight. Relative liver weight was increased with decrease of body weight by CPM 250 mg/kg treatment. Uterine cell proliferation tested with bromodeoxyuridine labeling index was not observed in CPM treated rats. CPM with or without testosterone propionate were administered to castrated rat of 51 days old for 10 days to investigate the (anti-)androgenic activity,. The weight of relative and absolute androgen-dependent accessory sex organs; seminal vesicle with coagulating glands (SV/CG), ventral prostate gland (VP), glans penis (GP), levator ani plus bulbocarvernosus muscle (LABC) and Cowper's gland (CG,) were unchanged by the treatment of CPM alone. While CPM induced the increase of relative adrenal gland weight, CPM 50mg/kg decreased the weights of CV/CG, VP, CG and LABC without change of GP without changing of GP when it was treated with TP. In conclusion, CPM dose not show estrogenic and anti-estrogenic activity in immature female rats, but it represents anti-androgenic activity by inhibition of the TP-stimulated increase of the weight of accessory sex organs.

Immunocytochemical localization of the endogenous vasoactive peptide apelin to human vascular and endocardial endothelial cells.

Apelin, the proposed endogenous peptide ligand of the novel G-protein-coupled receptor APJ, has been shown to possess potent vasodilator and positive inotropic effects in rats and humans in vivo. However, in humans, no endogenous source of apelin has been reported. Therefore, based on the presence of APJ and mRNA encoding apelin in human tissues, we investigated the expression of apelin in fresh-frozen human tissue from right atrium, left ventricle, lung, kidney, adrenal and large conduit vessels using immunocytochemistry. Apelin-like immunoreactivity (apelin-LI) was detected in vascular endothelial cells lining blood vessels in the human heart, kidney, adrenal gland and lung and in endothelial cells of large conduit vessels. Apelin-LI was also present in endocardial endothelial cells lining recesses of the right atrium. Apelin-LI was not present or below the level of detection in cardiomyocytes, Purkinje's cells, pulmonary or renal epithelial cells, secretory cells of the adrenal gland, vascular smooth muscle cells, adipocytes, nerves and connective tissue. The restricted presence of apelin-LI in endothelial cells suggests that endothelial apelin may play a role as a locally secreted cardiovascular mediator acting on APJ receptors present on the vascular smooth muscle and on cardiac myocytes to regulate vascular tone and cardiac contractility.

Evaluation of adrenal function in serum and feces of Steller sea lions (Eumetopias jubatus): influences of molt, gender, sample storage, and age on glucocorticoid metabolism.

Fecal corticosterone concentrations, measured via radioimmunoassay (RIA), were validated as a method to monitor adrenal function in Steller sea lion physiology. Quantification of adrenal response to an acute stressor and relevance of data produced by developed methodologies was determined through physiological challenge with exogenous administration of adrenocorticotropic hormone (ACTH) to captive adult, reproductively intact, Steller sea lions of both sexes (n=3, 1 male, 2 female) during seasonal molt. Following ACTH administration, serial blood and fecal samples were collected and analyzed by RIA to determine adrenal response. Storage regimens and weather exposure were examined to establish external impact on fecal corticosterone concentrations. High-pressure liquid chromatography (HPLC) of both serum and feces of Steller sea lions was employed to explore potential gender-based differences extant in either sample media. ACTH challenges produced >3-fold increases in serum cortisol concentrations which were reflected in >18-fold increases in fecal corticosterone concentrations post-injection at 3.25 and 32 h, respectively, and fecal corticosterone concentrations returned to baseline 52 h post-injection. Neither outdoor exposure to weather nor variation in duration and temperature of freezer storage impacted fecal corticosterone concentrations. HPLC of individual fecal samples produced eluate immunoreactivity profiles that differed consistently with both sex and age class. Techniques developed herein effectively detected physiologically relevant corticosterone data in Steller sea lion feces, unaffected by conditions likely to be encountered with field collection samples. Additionally, results quantify an acute response to ACTH and provide methodology for examining chronically heightened adrenal activity in Steller sea lions.
