Multicommuted flow-through fluorescence optosensor for determination of furosemide and triamterene.

Multicommutation implemented with flow-through optosensors is a very promising area of research. This recent approach benefits from the advantages of both methods and results in high sensitivity, selectivity, and speed, and little waste generation. This paper reports the simultaneous determination of furosemide and triamterene, two widely used diuretics, by measurement of their native fluorescence. The system has been proved to be useful for determination of both analytes in pharmaceutical preparations and for determination of triamterene in human urine and serum. A minicolumn filled with Sephadex SPC-25 microbeads was used to achieve separation of both analytes before detection in a flow-through cell filled with the same resin. The sensor is linear in the range 50-1200 and 0.4-8 ng mL(-1) with detection limits of 15 and 0.1 ng mL(-1) for furosemide and triamterene, respectively.

'The measurement of thermal neutron flux depression for determining the concentration of boron in blood'--suggestions as to further development of the method.

Suggestions about determining the concentration of 10B in blood via the thermal neutron flux depression measurement (NFDM) are made. The use of a measuring set-up consisting of a 252Cf neutron source, polyethylene moderator and a slim BF3 counter surrounded by an annular sample is examined. It is shown experimentally that using 6 ml samples and the source emitting 1.4 x 10(7) neutrons s(-1), one can determine the concentration of 10B in water at the level of 10 ppm with a statistical precision of 10% in about 20 min. Monte Carlo simulations performed with the use of MCNP-4C code revealed a potential for further improvements of the NFDM technique both in respect of the sample volume and counting period.

Sensitive LC MS quantitative analysis of carbohydrates by Cs+ attachment.

The development of a sensitive assay for the quantitative analysis of carbohydrates from human plasma using LC/MS/MS is described in this paper. After sample preparation, carbohydrates were cationized by Cs(+) after their separation by normal phase liquid chromatography on an amino based column. Cesium is capable of forming a quasi-molecular ion [M + Cs](+) with neutral carbohydrate molecules in the positive ion mode of electrospray ionization mass spectrometry. The mass spectrometer was operated in multiple reaction monitoring mode, and transitions [M + 133] --> 133 were monitored (M, carbohydrate molecular weight). The new method is robust, highly sensitive, rapid, and does not require postcolumn addition or derivatization. It is useful in clinical research for measurement of carbohydrate molecules by isotope dilution assay.
