Discovery of a dsRNA virus infecting the marine photosynthetic protist Micromonas pusilla.

We report the isolation of the first double-stranded (ds) RNA virus in the family Reoviridae that infects a protist (microalga Micromonas pusilla, Prasinophyceae). The dsRNA genome was composed of 11 segments ranging between 0.8 and 5.8 kb, with a total size of approximately 25.5 kb. The virus (MpRNAV-01B) could not be assigned to the genus level because host type, genome size, and number of segments smaller than 2 kb did not correspond to either of the two existing 11-segmented dsRNA genera Rotavirus and Aquareovirus. MpRNAV-01B has a particle size of 65-80 nm, a narrow host range, a latent period of 36 h, and contains five major proteins (120, 95, 67, 53, and 32 kDa). MpRNAV-01B was stable to freeze-thawing, resistant to chloroform, ether, nonionic detergents, chelating and reducing agents. The virus was inactivated at temperatures above 35 degrees C and by ionic detergent, ethanol, acetone, and acidic conditions (pH 2-5).

A DFT study of model complexes of zinc hydrolases and their inhibition by hydroxamic acids.

DFT calculations carried out on zinc acetate and zinc hydroxamates using the Hartree-Fock and B3LYP methods with the 6-311+G basis set give a series of stable pseudotetrahedral chelates (ZnL(2)) (L = OAc, FA, AA, NMeAA, GA, SA). Addition of a water molecule to these chelates gives the hydrates, ZnL(2).H(2)O, which in all cases are energetically more stable than the corresponding chelate. Hydrates formed from O,O coordinated hydroxamate species with a five-membered chelate ring contain water molecules occupying vacant coordination sites of the zinc atom. In contrast, those formed from zinc chelates with four-membered chelate rings contain a water molecule inserted into the chelate ring to give a six-membered ring in which one hydrogen of the water molecule is H-bonded to an oxygen atom of the zinc chelate with the water oxygen strongly bonded to the zinc. A slight lengthening of the H-bonded O-H bond suggests incipient hydroxide activation of water by zinc. In contrast, the O,O bonded hydroxamates do not incorporate water into the chelate ring nor activate the water in accordance with the ability of hydroxamic acids to inhibit zinc containing metalloenzymes.

Cysteine mediated multimerization of a recombinant dengue E fragment fused to the P64k protein following immobilized metal ion affinity chromatography.

A fragment of the Envelope protein of Dengue 2 virus encoding the amino acid (aa) 286-426 was fused to the P64k protein from Neisseria meningitidis. The hybrid gene (PD5) was expressed in Escherichia coli. The influence of using immobilized metal ion affinity chromatography (IMAC) in the purification process of PD5 was examined. After the elution, PD5 protein formed aggregates of high molecular weight depending on disulfide-bonds. The study of different conditions affecting the redox potential in the system revealed the influence of the copper ions on the multimerization of the protein, whereas metals with minor redox potential-for instance, zinc or nickel ions-did not cause this effect. It was also demonstrated that cysteines involved in this process belonged to the P64k protein. Finally, a PD5 purification process was established reaching a 85% of purity using the Zn(2+) as metal ion in the IMAC. This is a very useful finding due to the wide use of P64k as a carrier protein and the advantages of the IMAC as a chromatographic process.
