Dynamic multivalent lactosides displayed on cyclodextrin beads dangling from polymer strings.

Abstract

[reaction: see text] Lactose-appended cyclodextrin derivatives have been synthesized and threaded onto hydrophobic polymers in aqueous solution to form dynamic multivalent lactosides for binding to lectins. The threading process, which proceeds quickly, can be observed by one- and two-dimensional NMR spectroscopies.

Chemo-enzymatic synthesis of 1,4-oxazepanyl sugar as potent inhibitor of chitinase.

N-acetyl glucosamine 1 is selectively converted into 2 without protection of the other hydroxyl groups by allylation of the anomeric alkoxide in N,N-dimethylformamide containing lithium bromide. We use cell density cultures to produce the allylated derivative of penta-N-acetyl-chitopentaose by using 2 as the initial acceptor for the synthesis of 3 in vivo. Upon periodate oxidation, 3 is transferred to 4. Compound 4 is quickly subjected to sodium borohydride reduction and NH3 amination, which afforded the target compound 5. In 5-binding chitinase assay, it indicates that the chitinase is obviously inactivated by 5 with IC50 = 4.7 micromol/L.

Building up key segments of N-glycans in the gas phase: intrinsic structural preferences of the alpha(1,3) and alpha(1,6) dimannosides.

The intrinsic conformer specific vibrational spectra of two important subunits of the core pentasaccharide of N-linked glycans, the alpha(1,3) and alpha(1,6) dimannosides, have been recorded in the gas phase. Coupling these measurements with a computational exploration of their conformational landscapes has enabled their conformational assignment and has identified characteristic vibrational signatures associated with particular conformational families-including those that do or do not display inter-ring hydrogen bonding across the glycosidic linkage. In addition, the IR spectra of the monosaccharide moieties provide benchmarks, through which the conformational assignments can be refined. This introduces a general concept of modularity and secondary structure in oligosaccharides--essential for the success of similar studies on larger oligosaccharides in the future.

Stereocontrolled synthesis of spiroketals via Ti(Oi-Pr)4-mediated kinetic spirocyclization of glycal epoxides with retention of configuration.

A Ti(Oi-Pr)4-mediated kinetic spiroketalization reaction has been developed for the stereocontrolled target- and diversity-oriented synthesis of spiroketals. In contrast to most existing methods for spiroketal synthesis, this reaction does not rely upon thermodynamic control over the stereochemical configuration at the anomeric carbon. Stereochemically diverse glycals are first alkylated at the C1-position to introduce a hydroxyl-bearing side chain, then epoxidized stereoselectively. Treatment with Ti(Oi-Pr)4 leads to an unusual kinetic epoxide-opening spirocyclization (spirocycloisomerization) with retention of configuration at the anomeric carbon. The reaction is proposed to proceed via a chelation-controlled mechanism and has been used to form five-, six-, and seven-membered rings with stereochemically diverse substituents. This approach may also be useful for the related intermolecular beta-mannosidation reaction. This Ti(Oi-Pr)4-mediated spirocyclization is stereochemically complementary to our previously reported MeOH-induced spirocyclization, which proceeds with inversion of configuration, and together, these reactions provide comprehensive access to systematically stereochemically diversified spiroketals.
