Hydrothermal crystallization of carbonate-containing hydroxyapatite coatings prepared by radiofrequency-magnetron sputtering method.

Carbonate-containing hydroxyapatite (HA) films were prepared by low-temperature hydrothermal annealing from carbonate-containing calcium phosphate amorphous coatings on titanium substrates. The biocompatibility of the carbonate-containing HA layers was estimated by in vitro tests using simulated body fluid (SBF). Precursory amorphous coatings were deposited with rf-magnetron sputtering apparatus, using calcium phosphate glass target in Ar/CO2 atmosphere. The carbonate-containing HA coatings were successfully formed by the annealing at above 130 degrees C for 20 h. On the basis of SEM observation, about 2-microm thickness films coated rigidly were durable enough for the hydrothermal treatment. The coating layer was revealed to consist of single phase of PO4(-) and OH- partially carbonated HA by XRD and IR analyses. Overgrowing of bone-like apatite layers on the carbonate-containing HA surfaces in the SBF implied that the obtained films acquired a sufficient osteoconductivity, while it was still unclear that activity was enhanced, compared to pure HA coatings. The low-temperature hydrothermal annealing method was effective for preparation of rigid HA coatings on titanium as well as modification of their chemical compositions.

Very early prenatal diagnosis of genetic diseases based on coelomic fluid analysis: a feasibility study.

BACKGROUND: Coelocentesis may represent the ideal technique for very early prenatal diagnosis. Although cell density in coelomic fluid (CF) is very low, the results of analyses on the cellular compartment have been proposed for prenatal diagnosis. METHODS AND RESULTS: We aimed to evaluate the amount of total DNA (i.e. cellular and cell-free) in 14 samples (0.4-0.8 ml) of CF, taken from women at 8- to 9-week gestation, who are about to undergo termination of pregnancy, and to assess the feasibility of multiple single-gene analyses using multiplex real-time PCR. We found that the amount of total DNA in the CF was very low and varied widely. Genetic testing using multiplex real-time PCR was successfully achieved in 10 of 14 samples (71%). However, when considering samples that could provide a reliable prenatal diagnosis (i.e. successful PCR analysis and no marked maternal contamination), reliable CF-DNA-based prenatal diagnoses were obtained in only 8 of the 14 (58%) samples. CONCLUSION: The development of highly reliable procedures adapted to pauci-cellular CF is crucially needed before coelocentesis could be proposed for early prenatal diagnosis of genetic diseases before 10 weeks.

Biomineralization of calcium disilicide in porous polycaprolactone scaffolds.

The incorporation of CaSi(2) grains within a polycaprolactone (PCL) framework results in bioactive and biodegradable scaffolds which may be used in bone tissue regeneration. Porous PCL scaffolds were prepared via a combination of salt-leaching and microemulsion methods. To provide markedly different structural environments for the inorganic phase, calcium disilicide powder was either added to a mixed-composition porogen during a given scaffold preparation, or alternatively added to pre-formed scaffolds. Selective fluorescent labeling, scanning electron microscopy (SEM), and energy dispersive X-ray (EDX) analysis were employed to assess scaffold calcification in vitro. The process of CaSi(2)/PCL scaffold calcification under zero bias, during which calcium phosphate growth is significantly dependent on the structural degradation of CaSi(2) grains, has a similar mechanism as the calcium phosphate growth on bioactive glasses/ceramics. The biomineralization of these scaffolds is initiated solely by the silicide phase and can be accelerated by the degradation of the polymer matrix.
