Biphasic tautomerization dynamics of excited 7-hydroxyquinoline in reverse micelles.

The excited-state tautomerization dynamics of 7-hydroxyquinoline in the water pools of reverse micelles has been investigated by monitoring time-resolved fluorescence spectra and kinetics as well as static absorption and emission spectra with a variation of water content and isotopic fractionation. The normal and the tautomeric species are found to reside preferentially in the bound- and the free-water regions of the micelles, respectively. The excited-state tautomerization of the normal species in the bound-water layers is suggested to occur via two different channels, depending on rotamers at the moment of excitation. The cis tautomerizes via proton relay from the enol group to the imino group along a hydrogen-bonded water bridge, unusual in water but common in alcohols, whereas the trans tautomerizes via the stepwise individual acid-base reactions of two prototropic groups as found in bulk water. Proton relay can take place because water in the pools has substantially reduced polarity and disrupted hydrogen-bond networks compared with bulk water.

Kinetic study of the riboflavin-sensitised photooxygenation of two hydroxyquinolines of biological interest.

4-Hydroxyquinoline (4-OHQ) and 8-hydroxyquinoline (8-OHQ), two compounds of interest because of their bioactivity and their structural relation with bioactive products, are effectively photooxygenated when irradiated with visible light in the presence of riboflavin (Rf) (vitamin B2) in solution in air-saturated water-methanol (9:1). Rf behaves as a dye-sensitiser, since both quinolines are transparent to visible light. 8-OHQ degrades about five times faster than 4-OHQ. Kinetic data obtained through time-resolved and stationary detection of Rf-electronically excited states indicate that a superoxide radical anion-mediated mechanism exclusively operates for 4-OHQ, whereas singlet molecular oxygen--mainly--plus superoxide radical anion is the species that reacts with 8-OHQ. The sensitiser Rf, which is known to photodegrade under visible-light aerobic irradiation, is regenerated in the presence of any of the quinolines through an electron transfer process that produces superoxide radical anion. The overall picture indicates that both quinolines act as sacrificial scavengers of the photogenerated oxygen species, thus preventing the photodegradation of Rf.

A stable isotope dilution assay for the quantification of the Pseudomonas quinolone signal in Pseudomonas aeruginosa cultures.

A stable isotope dilution method was developed to analyse 2-heptyl-3,4-dihydroxyquinoline, also called the Pseudomonas quinolone signal (PQS), directly in Pseudomonas aeruginosa cultures by liquid chromatography coupled to mass spectrometry (LC/MS). PQS, along with the isobaric 2-heptyl-4-hydroxyquinoline N-oxide (HQNO), were quantified in various Pseudomonas liquid cultures using a deuterated PQS analog as internal standard. The kinetic of production of these quinolines in a growing culture of P. aeruginosa PA14 showed that their production starts at the end of the logarithmic growth phase and is maximal at the onset of the stationary growth phase. The concentration of PQS reached a maximum at 13 mg/l and then decreased, while the HQNO concentration reached 18 mg/l and then remained stable. Culture supernatants of P. aeruginosa strains PAO1 and PA14 produced similar concentrations of PQS whereas no PQS or HQNO could be detected in culture supernatants of the P. aeruginosa strain PAK or in the other Pseudomonas species tested, including phytopathogenic pseudomonads.
