Milestone's microwave labstation.

Abstract

In the actual research world, the laboratory has different needs, correlated to the field and level of research. Some researchers work with small volumes, others with large ones; in homogeneous or heterogeneous conditions, with solvent or dry media; at normal pressure or under pressure. And if we look at the field of drug discovery, during the last decade combinatorial chemistry, along with combinatorial and parallel synthesis, has held a fundamental role. This technique is becoming more and more popular because it concerns the quick synthesis of a high number of compounds with pharmaceutical activity. In this context, the efficency of microwave heating to speed up chemical reactions matches perfectly the need of short and clean chemical reactions. Milestone's MicroSYNTH labstation combined with the microwave technology, satisfies the need of the various chemical research laboratories. In fact, the different accessories that can be fitted inside the multimode microwave cavity cover a large ramp of volume, from 2 ml to 500 ml, different values of pressure, from 1 to 50 bar, and give the possibility to reach temperature up to 250 degrees C. And this is not all, you can also choose to perform one reaction at a time or to perform parallel synthesis with Milestone's rotors for up to 24 reactions at the same time.

Stereochemical identification of (R)- and (S)-ibuprofen using residual dipolar couplings, NMR, and modeling.

In this work, we describe an NMR-based method that utilizes an orientation media composed of the chiral polypeptide liquid crystal poly-gamma-benzyl-L-glutamate (PBLG) dissolved in CDCl(3), to measure the (1)H-(1)H, (1)H-(13)C and (13)C-(13)C residual dipolar couplings (RDCs) of (R) and (S)-ibuprofen. Calculated RDCs, obtained from the lowest energy conformers, are then compared with the experimentally measured RDCs to predict the stereochemistry of each enantiomer. Excellent agreement between calculated and experimental RDCs was found when the lowest energy structure of each enantiomer, obtained in a simulated PBLG/CDCl(3) environment, was used to back-calculate the RDCs. This method is generally useful for small molecular weight molecules that possess either one or two chiral centers, are soluble in low viscosity organic solvents, and will not crystallize (Clegg, Crystal Structure Analysis. Principles and Practice. New York: Oxford University Press; 2002) or cannot be derivatized with a Mosher's reagent (Dale and Mosher, J Am Chem Soc 1973;95:512-519).

A cyclic triphenylamine dimer for organic field-effect transistors with high performance.

A cyclic ethylene linked triphenylamine dimer formed highly crystalline thin film via vapor deposition. Meanwhile, the corresponding linear molecule only resulted in amorphous films under the same condition. The performance as FET semiconductor also improved significantly when the molecular structure derived from linear to cyclic type. The cyclic molecule displayed mobilities in excess of 10-2 cm2 V-1 s-1 and high on/off ratios up to 107.

Stereoselective syntheses of piperidinones and their modification by organometallic coupling reactions.

Dehydropiperidinones stereoselectively obtained from N-arabinosyl imines were iodinated at the enaminone structure. Knochel iodine-magnesium exchange afforded Grignard compounds of these piperidinone derivatives which reacted, either directly or after transmetalation to zinc or copper intermediates, with alkyl-, aryl- or acylhalides to give correspondingly substituted piperidinones. Stereoselective conjugate allyl cuprate addition to a thus obtained 5-allyl dehydropiperidinone and ring-closing metathesis of the product gave a hydroquinolinone containing three stereogenic centers.
