Hyaluronidase from two different strains of Streptococcus agalactiae was purified and characterized. The purification was performed successively by chromatography and rechromatography on phenylsepharose, gel filtration with FPLC on Superdex G 200 and isoelectric focusing. The purified hyaluronidase had an isoelectric point of 8.75 and a molecular weight of approximately 116,000 D. It showed maximal enzyme activity at pH 6.30 and 40 degrees C. The Michaelis constant was estimated to be 8.17 x 10(-2) mg/ml. Hyaluronidase was stimulated only by Mg++ and inhibited by Zn++, Al , Cu++ and Fe++ at a final concentration of 10 mmol/l, respectively. The enzyme splitted hyaluronic acid and in low amounts dermatan sulphate and chondroitin sulphate A. Additionally, synthetic polyanions (like polymers of gentisic acid with formaldehyde and hydroxy sulphonic acid with formaldehyde) turned out to be also potent inhibitors of the enzyme.
Using the Streptococcus equi decapsulation test, 879 strains of S. aureus, 212 strains of S. intermedius and 214 coagulase-negative staphylococci were examined for hyaluronidase production; six of the coagulase-negative staphylococci belonged to S. hyicus subsp. hyicus. Positive hyaluronidase production was recorded in all strains of S. aureus and in the strains of S. hyicus subsp. hyicus. No hyaluronidase was produced by 208 coagulase-negative staphylococci and the strains of S. intermedius. The decapsulation test is recommended as a reliable method of the differentiation of S. aureus and S. intermedius.
The data on the capacity of 50 melanogenic and 50 amelanogenic P. aeruginosa strains to produce hemolysins, gelatinase, caseinase, DNAase, RNAase, lecithinase, elastase, neuraminidase and to form extracellular slime, obtained in the comparative study of these strains in vitro, are presented. Melanogenic P. aeruginosa cultures were found to have a higher lecithinase and neuraminidase activity. The strains incapable of melanogenesis formed slime more frequently. The properties of the strains in respect to other pathogenicity characteristics under study were identical.
