A slotted waveguide applicator for continuous flow grain drying.

Abstract

A slotted waveguide antenna was designed and developed with series slots cut on the broad side that constituted a popular means of coupling microwaves with grain for drying. The radiating characteristics of the slotted wave guide antenna mounted in a setup that simulated the coupling to a grain conveying tube were studied at a frequency of 2.45 GHz. The antenna consisted of a WR-340 waveguide made of copper. The effect of the slot inclination angle O, moisture content and the shape of the slots were investigated. The results indicated 55 degrees as an optimum slot angle for slots having a width of 13 mm and a length 58 mm. Slots with a width of 6 mm did not exhibit any regular behavior and the effect of O was not clearly identified.

Comparative analysis of the heat stable proteome of radicles of Medicago truncatula seeds during germination identifies late embryogenesis abundant proteins associated with desiccation tolerance.

A proteomic analysis was performed on the heat stable protein fraction of imbibed radicles of Medicago truncatula seeds to investigate whether proteins can be identified that are specifically linked to desiccation tolerance (DT). Radicles were compared before and after emergence (2.8 mm long) in association with the loss of DT, and after reinduction of DT by an osmotic treatment. To separate proteins induced by the osmotic treatment from those linked with DT, the comparison was extended to 5 mm long emerged radicles for which DT could no longer be reinduced, albeit that drought tolerance was increased. The abundance of 15 polypeptides was linked with DT, out of which 11 were identified as late embryogenesis abundant proteins from different groups: MtEm6 (group 1), one isoform of DHN3 (dehydrins), MtPM25 (group 5), and three members of group 3 (MP2, an isoform of PM18, and all the isoforms of SBP65). In silico analysis revealed that their expression is likely seed specific, except for DHN3. Other isoforms of DNH3 and PM18 as well as three isoforms of the dehydrin Budcar5 were associated with drought tolerance. Changes in the abundance of MtEm6 and MtPM25 in imbibed cotyledons during the loss of DT and in developing embryos during the acquisition of DT confirmed the link of these two proteins with DT. Fourier transform infrared spectroscopy revealed that the recombinant MtPM25 and MtEm6 exhibited a certain degree of order in the hydrated state, but that they became more structured by adopting alpha helices and beta sheets during drying. A model is presented in which DT-linked late embryogenesis abundant proteins might exert different protective functions at high and low hydration levels.

Residues of azoxystrobin from grapes to raisins.

Azoxystrobin, a fungicide of the strobilurin group, has an European Union maximum residue level (MRL) of 2 mg/kg for grapes. This work aimed to assess residues on fresh and washed grapes and on raisins following processing with (i) alkali treatment and sun drying and (ii) sun drying only. QUADRIS 25% SC was applied according to good agricultural practice for two consecutive years on a typical cv. Thomson seedless and a seed-producing clone. Samples were collected 0, 15, and 21 days postapplication and analyzed using gas chromatography/electron capture detection; recoveries were 86 +/- 12% for grapes and 99 +/- 15% for raisins. Residues on grapes were 0.49-1.84 mg/kg, and washing removed 75% of the residue. Residues in raisins produced from seedless grapes were 0.51-1.49 (treatment 1) and 1.42-2.08 mg/kg (treatment 2), with residue transfer factors sometimes >1, even following alkali treatment, which reduced residues considerably. To avoid trade problems, a higher MRL for raisins is necessary.

Over-expression of tobacco NtHSP70-1 contributes to drought-stress tolerance in plants.

HSP70, a heat shock protein, is a molecular chaperone responsive to various environmental stresses. Here, NtHSP70-1 was a drought-/ABA-inducible gene. We monitored the expression of CaERD15 (early responsive to dehydration) with exposing plants to progressive drought stress. Its activity was used as an indicator of water-deficit conditions. To analyze the protective role of HSP70, we obtained transgenic tobacco plants that constitutively expressed elevated levels of the tobacco HSP70, NtHSP70-1, as well as transgenic plants containing either the vector alone or else having NtHSP70-1 in the antisense orientation. Plants with enhanced levels of NtHSP70-1 in their transgenic sense lines exhibited tolerance to water stress. Under progressive drought, the amount of leaf NtHSP70-1 was correlated with maintenance of optimum water content, with contents being higher in the leaves of dehydrated transgenic sense plants than in those of either the control (vector-only) or the transgenic antisense plants. Moreover, the expression of CaERD15 was considerably reduced in tobacco plants that over-expressed NtHSP70-1. These results suggest that elevated levels of NtHSP70-1 can confer drought-stress tolerance.
