Abstract

The types of phyllotaxis have been considered. The types of phyllotaxis occurring in plants allow them to keep unchanged the position of leaves on the shoot during growth, despite unequal stretching of stem in length and diameter.

Non-destructive prediction of lead content in oilseed rape leaves by fluorescence hyperspectral technology based on neural network.

Based on fluorescence hyperspectral imaging (FHSI), this study targeted rapid, non-destructive quantification of lead (Pb) content in oilseed rape leaves treated with varying silicon (Si) concentrations, acquiring fluorescence spectra over the 484.43-1001.61 nm wavelength range. To optimize spectral data quality, preprocessing methods (Savitzky-Golay smoothing, first derivative, detrending) were comprehensively compared. Characteristic wavelengths were then selected via interval variable iterative shrinkage, which effectively compressed data dimensionality and reduced computational load. A hybrid SE-CL1DA model, fusing a 1D convolutional neural network, a long short-term memory network and SE attention mechanism was constructed, with Bayesian optimization tuning hyperparameters to boost stability. The BO-SE-CL1DA outperformed both traditional machine learning and insufficiently optimized deep learning model (Rp2=0.9609, RMSE = 0.0377 mg/kg, RPD = 5.1736), thus enabling accurate Pb estimation, supporting Si-regulated heavy metal stress management and facilitating agricultural contamination monitoring.

A single-nucleus and spatial transcriptomic atlas of poplar leaves reveals the regulation of leaf polarity and cuticle deposition.

Leaf adaxial-abaxial polarity is fundamental for plant morphogenesis and environmental adaptation through asymmetric cell differentiation. Emerging evidence reveals dorsoventral metabolic gradients act downstream of transcriptional networks to fine-tune cellular specialization. While conserved transcription factors (e.g., HD-ZIP III and KANADI) establish initial polarity, the molecular networks driving position-specific cellular differentiation and their integration with metabolic adaptation remain unclear. Leveraging single-nucleus and spatial transcriptomics, we resolve major cell classes (mesophyll, epidermal, and vascular-associated) and their adaxial-abaxial subtypes, revealing dorsoventral polarity in transcriptional profiles and metabolic pathways. Adaxial cells are enriched in phenylpropanoid/flavonoid biosynthesis, while abaxial cells show preferential activation of stress and hormone signaling. Notably, we identify MYC2 as a key regulator of adaxial cuticle biosynthesis, binding to promoters of lipid biosynthetic and transport genes (e.g., CER10 and LTPG1) and promoting cuticle thickening. Our study uncovers how positional identity shapes transcriptional and metabolic polarity in leaves, with MYC2 emerging as a central regulator coordinating organ-specific adaptations. These findings provide insights into the spatial regulation of plant development and stress resilience, offering potential strategies for engineering stress-tolerant woody crops.

Ultrasound-driven mechanophore activation in living plants.

This study presents a biocompatible, ultrasound-responsive platform for remotely activating mechanochemical reactions within live plant tissue. Fluorogenic Mechanophore-embedded silica NanoParticles (FMNPs) that are thermally stable were engineered to emit blue fluorescence at 440 nm upon mechanical activation. In Solanum lycopersicum (tomato) leaves, activation was achieved through the synergistic combination of gas vesicles (GVs) and high-frequency focused ultrasound (FUS, 550 kHz), enabling spatially localized and minimally invasive stimulation. Low-frequency ultrasound (25 kHz) triggered activation but caused extensive tissue damage, while high-frequency FUS alone was biocompatible yet insufficient to activate FMNPs. Incorporation of GVs as a cavitation amplifier significantly boosted activation efficiency under mild acoustic conditions without observable tissue disruption. In planta fluorescence imaging confirmed that FMNPs retained their functionality after injection into leaf vasculature, and only the combination of GV and FUS produced a statistically significant fluorescence increase, indicating successful mechanochemical activation. This represents a demonstration of noninvasive and biocompatible ultrasound-induced mechanophore activation in live plants. This modular and noninvasive strategy opens possibilities for programmable release of regulatory and metabolic chemicals, biosensing, and synthetic molecular control in plant systems.
