Latex antigens.

Abstract

Previous studies have demonstrated that some children with spina bifida have IgE to proteins in natural rubber. In this study we compare different sources of latex antigen and identify possible antigenic peptides by radioimmunoblotting technique. Sera were collected from 26 children with spina bifida, tested by RAST with ammoniated latex extract (AL), and frozen until use. Extracts were prepared from ammoniated and nonammoniated latex, and the proteins were separated by electrophoresis on a 15% sodium dodecyl suflate-polyacrylamide gel and transferred to polyvinylene difluoride (PVDF). Strips of PVDF were then incubated with individual sera and 125I-labeled rabbit antihuman IgE before development by autoradiography; 18/26 sera were AL RAST positive; 0/8 AL RAST-negative patients had any binding to the latex proteins on PVDF. Sera from all patients were tested in a RAST with a nonammoniated latex extract (NAL), and the results were comparable to the AL RAST. Liquid-phase AL and NAL were comparable in their ability to inhibit the binding of patient's IgE to solid-phase AL and NAL. Sera from 14 RAST-positive patients were tested by immunoblotting with separated, reduced, and nonreduced AL and NAL. All 14 sera demonstrated IgE binding to a 14 kd peptide, which was more pronounced in reduced NAL. These results suggest that the 14 kd peptide in NAL is a major antigen in rubber allergy but that AL is an acceptable antigen source for in vitro diagnostic studies.

Isolation and characterization of phage-displayed single chain antibodies recognizing nonreducing terminal mannose residues. 2. Expression, purification, and characterization of recombinant single chain antibodies.

Since phage-display technology is probably the best available strategy to produce antibodies directed against various carbohydrate moieties, we employed phage-display technology to generate human single chain antibodies (scFvs) using neoglycolipids as carbohydrate antigens. An accompanying paper in this issue describes how phage-displayed antibodies (phage Abs) that recognized nonreducing terminal mannose residues were isolated and characterized. In this study, four independent scFv genes, isolated by a mannotriose (Man3)-bearing lipid as an antigen as previously described, were used to construct expression vectors to produce soluble scFv proteins in quantity. Both bacterial and mammalian expression systems were used to produce glutathione S-transferase-scFv fusion proteins and scFv-human IgG1 Fc conjugates, respectively. The expressed scFv fusion proteins were purified to apparent homogeneity with yields of approximately 1 and 48 mg, from 1 L of bacterial culture and myeloma cell media, respectively. Surface plasmon resonance and ELISA analyses confirmed that purified scFv proteins showed Man3 specificity. The humanized antibody in scFv-Fc form, derived from clone 5A3, was a disulfide-liked dimer with a molecular mass of 108 kDa. According to a bivalent model, the kinetics parameters of its binding to Man3 were determined to be ka = 4.03 x 104 M-1 s-1, kd = 5.77 x 10-4 s-1, KA = 6.98 x 107 M-1, and KD = 1.43 x 10-8 M. This study thus established the foundation for isolation of carbohydrate-specific scFv genes and eventual production of humanized scFv-Fc type antibodies.

Generation of plant protein microarrays and investigation of antigen-antibody interactions.

The application of proteomics methods, such as the protein microarray technology, in plant science has been strongly supported by the completion of genome sequencing projects of Arabidopsis thaliana and rice. In this chapter we describe a method to generate plant protein microarrays and to use them for characterizing monoclonal antibodies or polyclonal sera with regard to their specificity and cross-reactivity. The method starts with characterized E. coli cDNA expression clones encoding His-tagged plant proteins. After expression and purification of these recombinant proteins in high throughput, protein microarrays are generated utilizing a contact printer. For the detection of the recombinant proteins on the microarrays, an anti-RGS-His6 antibody is used. To characterize specific antibodies, the microarrays are incubated with the respective antibody solutions followed by fluorescently labeled secondary antibodies. Signal detection is performed by means of an arrayscanner system. Protein microarrays containing the whole proteome of a plant will represent the ideal format to test antibody specificity and cross-reactivity in the future.

Standardization of FSH, LH and hCG--current position and future prospects.

Gonadotropin measurements contribute significantly to patient management in both endocrinology and oncology. Differences in calibration, antibody specificities and assay design mean that gonadotropin results obtained in different methods are still not comparable. Comparing patient results obtained in different methods therefore remains problematic, whether for individual patient care, when assessing the results of multicentre clinical trials, or when formulating national and international guidelines and recommendations. Achieving improved comparability of results for these important analytes will require clear descriptive nomenclature, accurate calibration with highly purified standards, careful characterization of what gonadotropin isoforms methods are measuring, broad recommendations about the most clinically appropriate antibody combinations, and increased awareness of clinically relevant interferences and the action required to minimise their effect. Encouraging manufacturers to standardize and carefully describe the evaluation methods they use, such that data from different manufacturers can readily be compared, is also a pre-requisite for future progress.
