A method for multiple superposition of structures.

Abstract

The study of families of protein structures is important in analysing the results of NMR structure determinations and in investigating mechanisms of molecular evolution at the level of conformation. A method is discussed for finding the transformations that mutually superpose an arbitrary number of structures in the least-squares sense given specified atom-to-atom correspondence.

Some different strategies of least-squares refinement of a molecule.

Refinements of a macromolecule (ribonuclease-A) based on structure amplitudes, magnitude of F, and structure amplitude squares, magnitude of F2, were carried out and the results compared. Although the conventional R values are higher for the magnitude of F2 refinement, positional parameters from both types of refinement were not significantly different. However, the mean-square displacements from magnitude of F2 refinements were systematically higher than for those using magnitude F. Various resolution windows and weighting schemes were employed during the work. Electron density maps were examined for magnitude of F2 refinements and were very similar to those using magnitude of F in spite of a conventional R factor of 0.29 using all 1.4 A data. While magnitude of F2 refinements may be formally more correct than magnitude of F refinements, there is little evidence that magnitude of F2 refinement is superior provided that a reasonable weighting strategy is adopted.

Least-Squares Analysis

Experimental design-based quantitative structure-toxicity relationship of some local anaesthetics using the PLS method.

A quantitative structure-toxicology (LD50) relationship for some N-alkylsuccinimides is presented. The relationship is based on a small and carefully selected training set using experimental design methodology. The good predictability, an overall r2 value of 0.75, with such an approach is demonstrated. Factors such as large N-alkyl groups, substitution in the aromatic ring and a long side chain between the two nitrogens are favourable for low toxicity.

Least-Squares Analysis

[Simultaneous analysis of vitamin B complex by least square UV-spectrophotometry].

Simultaneous analysis of VitB1, VitB2 and VitB3 in Vitamin B Complex tablets was made by least square UV-spectrophotometry. The results showed that the contents of VitB1, VitB2 and VitB3 could be calculated by computer with least square method using 12 absorbances of 239, 241, 257, 259, 261, 263, 266, 269, 270, 279 and 281 nm wavelengths. The reclamation of VitB1 was 101.4% (CV% = 0.88, n = 3), of VitB2 98.5% (CV = 0.43, n = 3), and of VitB3 101.3% (CV% = 0.15, n = 3).
