Molecular design of artificial ribonucleases using electrostatic interaction.

Abstract

A number of small organic ribonucleases have been synthesized with rigid polycationic structures containing an aromatic framework with two residues of bis-quaternary salts of 1,4-diazabicyclo[2.2.2]octane (DABCO) bearing various substituents. The compounds carrying positively charged groups connected via rigid linker are expected to bend the sugar-phosphate backbone and can stimulate the intramolecular phosphoester transfer reaction.

From supramolecular porphyrin tweezers to dynamic A(n)B(m)C(l)D(k) multiporphyrin arrangements through orthogonal coordination.

A dynamic, supramolecular, three-component A(n)B(m)C(l) bis(zinc porphyrin) tweezer has been prepared quantitatively using the heteroleptic bisphenanthroline (HETPHEN) concept. Upon addition of nitrogenous spacers of different length, namely, the extended bipyridine 3 a, 4,4'-bipyridine (3 b), and 1,4-diazabicyclo[2.2.2]octane (DABCO; 3 c), to set up an additional orthogonal binding motif (Zn(Por)-N(spacer)), three structurally different, still dynamic, four-component A(n)B(m)C(l)D(k) assemblies were cleanly formed, as indicated by UV/Vis and NMR titrations as well as by DOSY investigations. The structures were identified as a bridged monotweezer A(2)BC(2)D, a doubly bridged double tweezer A(4)B(2)C(4)D(2), and a triply bridged double tweezer A(4)B(2)C(4)D(3), the latter resembling a porphyrin stack. Notably, the same structures were equally formed directly from a mixture of the constituents A, B, C, and D put together in any sequence if the correct stoichiometry was applied.

Study of the electrospray ionization tandem mass spectrometry of sildenafil derivatives.

A detailed analysis of the product ion spectrum generated from the protonated molecule of sildenafil (Viagra(R)) under multiple tandem mass spectrometry (ESI-MS(n)) conditions using an ion-trap mass spectrometer is reported. Molecular composition data of the fragment ions were obtained with the aid of comparisons of the multiple tandem mass spectra of eight sildenafil derivatives in two series, the structures of which are identical except for some substituted alkyl groups. Attempts have been made to provide rational pathways for the formation of the fragment ions from these protonated sildenafil derivatives. The structure-fragmentation relationships will facilitate the characterization of the structures of other sildenafil analogs.

Structure and serotonin 5-HT2C receptor activity of ortho- and meta-substituted phenylpiperazines.

The structural characteristics of ortho- and meta-substituted phenylpiperazines have been investigated in order to understand their actions at the serotonin 5-HT2C receptor. The crystal structures of the 4-methylated analogues of two phenylpiperazines that are already known as 5-HT2C ligands, 1-(1-naphthyl)-4-methylpiperazine (1NMP) and 1-[(3-trifluoromethyl)phenyl]-4-methylpiperazine (TFMPMP), and those of two novel 5-HT2C ligands, 1-(2-methoxyphenyl)piperazine (oMPP) and 1-(3-methoxyphenyl)piperazine (mMPP), are determined. Molecular mechanics calculations are performed to calculate the energy profiles of six phenylpiperazines for rotation about the central phenyl-nitrogen bond. The activities of several phenylpiperazines, in combination with their crystal structures and conformational characteristics, lead to the hypothesis that the conformation for which the piperazine ring and the phenyl ring are approximately co-planar should be the 5-HT2C receptor 'activating' conformation. This hypothesis is then used to predict the activities of the two novel 5-HT2C ligands oMPP and mMPP. oMPP is predicted to be an antagonist at this receptor, whereas mMPP is predicted to be an agonist. As this prediction was confirmed by in vitro and in vivo tests, the proposed conformation is very likely to be responsible for the activation of the 5-HT2C receptor.
