Study of pharmaceutical solid dispersions by microthermal analysis.

Abstract

PURPOSE: The main objective of the study was to determine the ability of microthermal analysis (microTA) to assess the crystallinity of the drug in two noncommercial pharmaceutical solid dispersions. METHODS: Pure substances, physical mixes, and solid dispersions were analyzed by microTA. The thermal values obtained by microTA were compared with data obtained by more conventional techniques like differential scanning calorimetry. RESULTS: microTA was able to detect the drug in the waxy matrix. The technique was capable of showing that relatively large amorphous drug domains (up to 70 microm) are formed during the solidification of the solid dispersion. These amorphous domains are thermodynamically unstable and can crystallize upon aging. CONCLUSIONS: microTA was successfully applied to the physical characterization of solid dispersions. Local thermal analysis (LTA) offers a unique way to probe a selected area on the surface of the sample. It is capable of melting the drug locally without melting the lower melting point excipient. These results gave an understanding of the poor dissolution performance of these solid dispersions upon aging.

Thermal analysis in the evaluation of sediment pollution.

Characterization of organic matter in four sediments in the infuence area of wastewater discharges was carried out by both chemical and thermal analysis in order to assess their pollution level. Oxidisable organic carbon and organic matter were calculated by the standard methodology in laboratory. Thermogravimetry (TG), between 50 and 900 degrees C, was simultaneously performed in oxidizing conditions on ground sediments samples after three different pretreatments. Linear regression adjust of thermal analysis results versus chemical parameters provides the better Pearson's coefficients, leading to the best coefficients for weight loss in 250-400 degree C temperature range versus oxidisable organic carbon and organic matter at 360 degrees C, respectively. These results demonstrated the utility of thermoanalysis technique for the evaluation of the organic matter content of fresh sediments.

An experimental study of sewage sludge incineration.

Sludge incinerated ash has been fired at different temperatures. This material was fired at 1050 degrees C for 3 h and until a peak of 1010 degrees C. After thermal treatment the ash was screened at 200 mesh. The ash was characterized by X-ray fluorescence and trace elements like Cr, Pb, Zn, Cu, and some oxides like quartz(SiO2), Al2O3, P2O5, Fe2O3 were found. Scanning electron microscopy(SEM) has shown one change of particle between 2 microm at 90 microm and apparent porosity, thermal analysis has shown loss of mass and residual decomposition in the TG, DTG and DTA curves.

Synthesis and fluorescence properties of rare earth (Eu3+ and Gd3+) complexes with alpha-naphthylacetic acid and 1,10-phenanthroline.

Rare earth binary complex Eu(NNA)3 (NNA, alpha-naphthylacetic acid), ternary complex Eu(NNA)3.phen (phen, 1,10-phenanthroline) and a series of dinuclear complexes with different mole ratios of Eu3+ to Gd3+ were synthesized. Many advanced approaches, such as element analysis, FTIR spectra, TG and DTA analysis, were used to determine the composition and structure of binary and ternary complex. Moreover, their fluorescence properties were studied by fluorescent spectra and lifetimes. The fluorescence spectra and decay curves of dinuclear complexes indicated that the fluorescence emission intensity was enhanced and the fluorescence lifetime was prolonged by Gd3+. The dinuclear complexes show the best properties when the mole ratio of Eu3+ to Gd3+ is 6:4. A new parameter Y, which was used to evaluate the effect of Gd3+, was introduced. In addition, the relationship of Y value and mole fraction of Gd3+ was analyzed by mathematical software. The results showed that Y value decreased by single exponential mode when the content of Gd3+ decreased.
