Third-harmonic generation from single gold nanoparticles.

Abstract

We report the first observation of third-harmonic signals from individual gold colloids down to 40 nm diameter. Excited with 1-ps pulses at 1500 nm, the colloids generate 500-nm light, close to the plasmon resonance. The third-harmonic intensity varies as the square of the colloid surface area. Although weak, the third-harmonic signals of gold labels as small as 15 nm in diameter are expected to be accessible with 100-fs pulses. They could be used in microscopy for single-biomolecule tracking.

Formation of spherical nanostructures by the controlled aggregation of gold colloids.

Stable suspensions of size-uniform spherical assemblies of 5-8 nm gold colloids in toluene are readily obtained by cross linking the colloidal particles using alkanedithiols within a defined range of gold-dithiol molar ratios. The assemblies are very stable and remain suspended in toluene for several months without significant aggregation. These porous gold spheres can be further organized into hierarchically assembled relatively linear chains by the addition of ethanol.

Stability of thiol-passivated gold particles at elevated temperatures studied by X-ray photoelectron spectroscopy.

The thermal stability of thiol-passivated gold colloids has been studied by means of X-ray photoelectron spectroscopy. Different colloids were chemically synthesized with thiol lengths from 3 to 8, and 16 carbon atoms. Depending on the synthesis parameters, the mean gold particle size varied from 1.6 to 4.9 nm for the particular colloid. Temperature-dependent measurements revealed a general tendency of shells with longer thiol chains (>5 carbon atoms) to be more stable in terms of desorption but to show a X-ray radiation-induced damage, which was absent with shorter thiols. Additionally, the self-assembling properties of the colloids were characterized by TEM, showing differences as a function of the chain lengths.

[Dynamic atomic force microscopic observation of the three-dimensional conformation of protein A-gold single molecule].

OBJECTIVE: To determine the three-dimensional (3D) conformation of staphylococcal protein A-gold (SPA-G) single molecule using atomic force microscope (AFM) so as to evaluate the feasibility of using this molecule for in situ labeling of the neuronal membrane protein. METHODS: AFM was used to acquire the images of SPA-G binding to the surface of mica under physiological condition for determining the 3D conformation of the molecule. RESULTS: SPA-G single molecule was shown to contain a characteristic structure with a chain in the shape of mirror image of the letter C, which had the dimension of 48.80 nm x 42.13 nm x 20.53 nm. CONCLUSION: AFM provide a new means for morphological investigation of the biomacromolecule at the nanometer scale in physiological conditions, and SPA-G can be utilized for in situ labeling of the neuronal membrane receptors.
