A stability indicating LC method for zolmitriptan.

Abstract

A gradient, reversed-phase liquid chromatographic (RP-LC) assay method was developed for the quantitative determination of zolmitriptan, used to treat severe migraine headaches. The developed method is also applicable for the related substances determination in bulk drugs. The chromatographic separation was achieved on a Waters X Terra RP18, 250 mm x 4.6 mm, 5 microm column. The gradient LC method employs solutions A and B as mobile phase. The solution A contains a mixture of phosphate buffer pH 9.85:methanol:acetonitrile (70:20:10, v/v/v) and solution B contains a mixture of phosphate buffer, pH 9.85:acetonitrile (30:70). The flow rate was 1.0 ml/min and the detection wavelength was 225 nm. In the developed HPLC method, the resolution between zolmitriptan and its potential impurities, namely Imp-1, Imp-2 and Imp-3 was found to be greater than 3. The drug was subjected to stress conditions of hydrolysis, oxidation, photolysis and thermal degradation. Considerable degradation was found to occur in alkaline medium and oxidative stress conditions. Degradation product formed during base hydrolysis was found to be Imp-3. The stress samples were assayed against a qualified reference standard and the mass balance was found close to 99.5%. The developed RP-LC method was validated with respect to linearity, accuracy, precision and robustness.

Aminolysis of 2,4-dinitrophenyl X-substituted benzoates and Y-substituted phenyl benzoates in MeCN: effect of the reaction medium on rate and mechanism.

Second-order rate constants (k(N)) have been determined spectrophotometrically for the reactions of 2,4-dinitrophenyl X-substituted benzoates (1 a-f) and Y-substituted phenyl benzoates (2 a-h) with a series of alicyclic secondary amines in MeCN at 25.0 +/- 0.1 degrees C. The k(N) values are only slightly larger in MeCN than in H2O, although the amines studied are approximately 8 pK(a) units more basic in the aprotic solvent than in H2O. The Yukawa-Tsuno plot for the aminolysis of 1 a-f is linear, indicating that the electronic nature of the substituent X in the nonleaving group does not affect the rate-determining step (RDS) or reaction mechanism. The Hammett correlation with sigma- constants also exhibits good linearity with a large slope (rho(Y) = 3.54) for the reactions of 2 a-h with piperidine, implying that the leaving-group departure occurs at the rate-determining step. Aminolysis of 2,4-dinitrophenyl benzoate (1 c) results in a linear Brønsted-type plot with a beta(nuc) value of 0.40, suggesting that bond formation between the attacking amine and the carbonyl carbon atom of 1 c is little advanced in the transition state (TS). A concerted mechanism is proposed for the aminolysis of 1 a-f in MeCN. The medium change from H2O to MeCN appears to force the reaction to proceed concertedly by decreasing the stability of the zwitterionic tetrahedral intermediate (T+/-) in aprotic solvent.

HPLC method for determination of SN-38 content and SN-38 entrapment efficiency in a novel liposome-based formulation, LE-SN38.

A simple HPLC method was developed for quantification of SN-38, 7-ethyl-10-hydroxycamptothecin, in a novel liposome-based formulation (LE-SN38). The chromatographic separation was achieved on an Agilent Zorbax SB-C18 (4.6 mmx250 mm, 5 microm) analytical column using a mobile phase consisting of a mixture of NaH2PO4 (pH 3.1, 25 mM) and acetonitrile (50:50, v/v). SN-38 was detected at UV wavelength of 265 nm and quantitatively determined using an external calibration method. The limit of detection (LOD) and limit of quantitation (LOQ) were found to be 0.05 and 0.25 microg/mL, respectively. The individual spike recovery of SN-38 ranged from 100 to 101%. The percent of relative standard deviation (%R.S.D.) of intra-day and inter-day analyses were less than 1.6%. The method validation results confirmed that the method is specific, linear, accurate, precise, robust and sensitive for its intended use. The current method was successfully applied to the determination of SN-38 content and drug entrapment efficiency in liposome-based formulation, LE-SN38 during early stage formulation development.

Kinetics and mechanism of the oxidation of hydroquinones by a trans-dioxoruthenium(VI) complex.

The kinetics of the oxidation of hydroquinone (H(2)Q) and its derivatives (H(2)Q-X) by trans-[Ru(VI)(tmc)(O)(2)](2+) (tmc = 1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetradecane) have been studied in aqueous acidic solutions and in acetonitrile. In H(2)O, the oxidation of H(2)Q has the following stoichiometry: trans-[Ru(VI)(tmc)(O)(2)](2+) + H(2)Q --> trans-[Ru(IV)(tmc)(O)(OH(2))](2+) + Q. The reaction is first order in both Ru(VI) and H(2)Q, and parallel pathways involving the oxidation of H(2)Q and HQ(-) are involved. The kinetic isotope effects are k(H(2)O)/k(D(2)O) = 4.9 and 1.2 at pH = 1.79 and 4.60, respectively. In CH(3)CN, the reaction occurs in two steps, the reduction of trans-[Ru(VI)(tmc)(O)(2)](2+) by 1 equiv of H(2)Q to trans-[Ru(IV)(tmc)(O)(CH(3)CN)](2+), followed by further reduction by another 1 equiv of H(2)Q to trans-[Ru(II)(tmc)(CH(3)CN)(2)](2+). Linear correlations between log(rate constant) at 298.0 K and the O-H bond dissociation energy of H(2)Q-X were obtained for reactions in both H(2)O and CH(3)CN, consistent with a H-atom transfer (HAT) mechanism. Plots of log(rate constant) against log(equilibrium constant) were also linear for these HAT reactions.
