Reusable impedimetric aptasensor.

Abstract

A novel impedimetric aptasensor using a mixed self-assembled monolayer composed of thiol-modified thrombin binding aptamer and 2-mercaptoethanol on a gold electrode is reported. The changes of interfacial features of the electrode were probed in the presence of the reversible redox couple, Fe(CN)6(3-/4-), using impedance measurements. The electrode surface was partially blocked due to the self-assembly of aptamer or the formation of the aptamer-thrombin complex, resulting in an increase of the interfacial electron-transfer resistance detected by electrochemical impedance spectroscopy or cyclic voltammetry. The aptasensor was regenerated by breaking the complex formed between the aptamer and thrombin using 2.0 M NaCl solution, and the immobilized aptamer subsequently was used for repeated detection of thrombin. The aptamer-functionalized electrode showed a linear response of the charge-transfer resistance to the increase of thrombin concentration in the range of 5.0-35.0 nM and the thrombin was easily detectable to a concentration of 2.0 nM.

Hybrid systems composed of a glucose oxidase (GOx)/peroxidase-mimicking DNAzyme, and of microperoxidase-11 (MP-11)/anti-thrombin aptamer were synthesized. The hybrid systems were employed as amplifying labels for the colorimetric or chemiluminescence detection of an enzyme functions, and thrombin analysis, respectively. In the GOx/DNAzyme system, the GOx-mediated oxidation of glucose led to the formation of H(2)O(2), and this activated the oxidation of ABTS to a colored product, or to the generation of chemiluminescence in the presence of luminol. The MP-11/anti-thrombin aptamer enabled the amplified analysis of thrombin by the MP-11-mediated generation of chemiluminescence in the presence of luminol/H(2)O(2).

Methods developed for SELEX.

SELEX (systematic evolution of ligands by exponential enrichment) is a process that involves the progressive purification from a combinatorial library of nucleic acid ligands with a high affinity for a particular target by repeated rounds of partitioning and amplification. With the development of aptamer technology over the last decade, various modified SELEX processes have arisen that allow various aptamers to be developed against a wide variety of molecules, irrespective of the target size. In the present review, the separation methods used in such SELEX processes are reviewed.
