Substrate monolayers as electrochemical sensing elements for alpha-chymotrypsin.

Abstract

A disulfide, which carried two L-phenylalanyl p-nitroanilide (Phe-pNA) moieties at both ends, was prepared by the coupling of 11,11'-dithiodiundecanoic acid (DTUA) with Phe-pNA. The compound obtained (DTUA-Phe-pNA) formed a self-assembled monolayer (SAM) on a gold electrode and vacuum-evaporated gold thin film as proven by cyclic voltammetry, and reflection absorption spectroscopy, respectively. Incubation of alpha-chymotrypsin with the SAM-modified electrode induced both a decrease in anodic and cathodic peak currents (-DeltaIa and -DeltaIc) and an increase in potential difference (DeltaEp) in the cyclic voltammogram of potassium ferricyanide as a probe, which suggested the attack of the enzyme at the amide group between Phe and pNA groups of the SAM, resulting in the formation of an intermediate. The linear relationship between the initial rate of increase in the amount of enzyme bound to the SAM and in both DeltaIa (absolute value of the decrease in anodic peak current) and DeltaEp values was confirmed by the quartz crystal microbalance method. The binding rate of the enzyme to the Phe-pNA SAM was dependent on the surface density of the Phe-pNA group in the SAM. The alpha-chymotrypsin-induced increases in the DeltaIa and DeltaEp values were inhibited by the addition of N-acetyl-D-phenylalanine methyl ester (N-Ac-D-Phe-OMe). In contrast with alpha-chymotrypsin, trypsin did not show a significant increase in the DeltaIa and DeltaEp values upon incubation with Phe-pNA-carrying SAM. These results could be attributed to the specific attack of alpha-chymotrypsin to the amide group in the SAM. The inhibition constant for N-Ac-D-Phe-OMe in the SAM system was quite similar to that in the free substrate system, showing that the enzymatic reaction above the SAM proceeds in a similar way to that in the homogeneous solution system.

Convenient synthetic approach to 2,4-disubstituted quinazolines.

[reaction: see text] 2,4-Dialkyl or aryl quinazolines have been prepared in three steps starting from easily available anilides. A photochemically induced Fries rearrangement of the anilides gave several ortho-aminoacylbenzene derivatives that were acylated at the NH2. These acylamides underwent rapid cyclization to 2,4-disubstituted quinazolines (and benzoquinazolines) in the presence of ammonium formate under microwave activation. This procedure is compatible with different functional groups and allowed also the preparation of new quinazolines derived from naturally occurring amino acids.

Vanilloid receptor agonists and antagonists are mitochondrial inhibitors: how vanilloids cause non-vanilloid receptor mediated cell death.

Time-lapse photomicroscopy of human H460 lung cancer cells demonstrated of the transient receptor potential V1 (TRPV1) channel agonists, (E)-capsaicin and resiniferatoxin, and the TRPV1 antagonists, capsazepine, and SB366791, were able to bring about morphological changes characteristic of apoptosis and/or necrosis. Immunoblot analysis identified immunoreactivity for the transient receptor potential V1 (TRPV1) channel in rat brain samples, but not in rat heart mitochondria or in H460 cells. In isolated rat heart mitochondria, all four ligands caused concentration-dependent decreases in oxygen consumption and mitochondrial membrane potential. (E)-Capsaicin and capsazepine evoked concentration-dependent increases and decreases, respectively, in mitochondrial hydrogen peroxide production, whilst resiniferatoxin and SB366791 were without significant effect. These data support the hypothesis that (E)-capsaicin, resiniferatoxin, capsazepine, and SB366791 are all mitochondrial inhibitors, able to activate apoptosis and/or necrosis via non-receptor mediated mechanisms, and also support the use of TRPV1 ligands as anti-cancer agents.

Thiazolidinedione ameliorates renal injury in experimental diabetic rats through anti-inflammatory effects mediated by inhibition of NF-kappaB activation.

Thiazolidinedione (TZD), a ligand for peroxisome proliferator-activated receptor-gamma (PPAR-gamma), exerts anti-inflammatory effects independently of the insulin-sensitizing effect. In the present study, we tested the hypothesis that TZD prevents the progression of diabetic nephropathy by modulating the inflammatory process. Five-week-old Sprague-Dawley rats were divided into three groups: 1) nondiabetic control rats (non-DM), 2) diabetic rats (DM), and 3) diabetic rats treated with pioglitazone (DM+pio). Diabetes was induced by injection with streptozotocin (STZ). The DM+pio group received 0.0002% pioglitazone mixed in chow for 8 wk after induction of diabetes. Blood glucose and HbA1c were elevated in diabetic rats but did not change by treatment with pioglitazone. Pioglitazone reduced urinary albumin excretion and glomerular hypertrophy, suppressed the expression of transforming growth factor (TGF)-beta, type IV collagen, and ICAM-1, and infiltration of macrophages in the kidneys of diabetic rats. Furthermore, renal NF-kappaB activity was increased in diabetic rats and reduced by pioglitazone. PPAR-gamma was expressed in glomerular endothelial cells in the diabetic kidney and in cultured glomerular endothelial cells. High-glucose conditions increased the expression of ICAM-1 and the activation of NF-kappaB in cultured glomerular endothelial cells. These changes were reduced by pioglitazone, ciglitazone, and pyrrolidine dithiocarbamate, an inhibitor of NF-kappaB. However, pioglitazone did not show the changes in the presence of PPAR-gamma antagonist GW9662. Our results suggest that the preventive effects of pioglitazone may be mediated by its anti-inflammatory actions, including inhibition of NF-kappaB activation, ICAM-1 expression, and macrophage infiltration in the diabetic kidney.
