Parallel, solution phase synthesis of dihydropyridine miticides via a versatile multicomponent reaction.

Abstract

A novel class of highly active dihydropyridine miticides was prepared using a multicomponent reaction process. The initial lead was rapidly optimized using solution phase parallel synthesis techniques and a positional scanning approach. Detailed structure-activity relationships were developed for the amino and carbonyl components of the molecule and used to select the best candidates for broad field testing. The chemistry, biology and toxicology of these compounds will be presented along with numerous structural variants of the reaction products.

Effects of Phos-Chek G75-F and Silv-Ex on red-winged blackbird (Agelaius phoeniceus) embryos.

Effects of field application levels of wildfire control chemicals, Phos-Chek G75-F (PC) and Silv-Ex (SE), were examined on red-winged blackbird (Agelaius phoeniceus) embryos. Embryos were more sensitive to PC and SE when eggs were immersed for 10s at an early developmental stage (days 3-5 of incubation) than at a later stage (days 6-9 of incubation). The LC(50) (concentration causing 50% mortality) for early stage embryos exposed to PC was 213.3g/L (slope=1.6; 95% confidence interval [CI]=129.1-326.1). The no observed effect concentration (NOEC) was below 135g PC/L, which caused a significant increase in embryonic mortality and represents the lowest field coverage level of 1gal/100feet(2). The LC(50) for early stage embryos exposed to SE was 19.8g/L (slope=1.5; 95% CI=11.7-52.2). Significant mortality was observed at 10g SE/L and marginal at 7.5g SE/L with an apparent NOEC around 5g SE/L. Neither chemical resulted in apparent developmental malformations.

Influence of agricultural adjuvants on droplet spectra.

The formation of a spray is the result of interaction between the nozzle type and the spray liquid. When adjuvants lower the surface tension, a shift to a finer spray quality might be expected. Increases in viscosity might cause coarser sprays. The spray pressure, nozzle type and spray liquid determine spray performance. Adjuvants that cause droplets of oil in the spray mixture, for example crop oils, petroleum oils and even some water-insoluble emulsifiers and surfactants, may unexpectedly increase the spray droplet size. In cases where a finer spray is obtained, the volume fraction of drops smaller than 100 microm diameter, V(100), is expected to increase, but for some adjuvants a decrease in V(100) is observed. Finally, spray droplets may also differ when the concentration of the applied adjuvants changes. An overview based on reports in the literature is given of the effects of different classes of adjuvants used for agricultural cropping on spray droplet spectra. The effects of these adjuvants on spray formation depend on the type of nozzle in combination with the applied pressure.

Community factors in the development of antibiotic resistance.

The global impact of antibiotic resistance is potentially devastating, threatening to set back progress against certain infectious diseases to the pre-antibiotic era. Although most antibiotic-resistant bacteria originally emerged in hospitals, drug-resistant strains are becoming more common in the community. Factors that facilitate the development of resistance within the community can be categorized as behavioral or environmental/policy. Behavioral factors include inappropriate use of antibiotics and ineffective infection control and hygiene practices. Environmental/policy factors include the continued use of antibiotics in agriculture and the lack of new drug development. A multifaceted approach that includes behavioral strategies in the community and the political will to make difficult regulatory decisions will help to minimize the problem of antimicrobial resistance globally.
