Assembly of type C oncornaviruses: a model.

Abstract

The salient features of this model for oncornavirus assembly are that uncleaved precursor molecules to the internal virus polypeptides possess specific recognition sites both for viral envelope constituents already inserted in the cell membrane and for the viral RNA. After orderly alignment of these components at the budding site, virus maturation proceeds through specific proteolytic cleavage of the precursor components and association of the resultant molecules into the characteristic type C virion substructures revealed by electron microscopy.

Time-resolved transcriptomics of S. cerevisiae and S. pastorianus in response to plasma membrane stresses.

Yeasts are beneficial microorganisms for human society and are utilized for academic and industrial purposes. For academic purposes, S. cerevisiae is a well-investigated model for studying eukaryotic cellular processes. For industrial purposes, S. pastorianus, which has a hybrid genome of S. cerevisiae and S. eubayanus, has been served for lager beer production. During fermentation, S. pastorianus produces ~7% of EtOH, which induces plasma membrane (PM)/cell wall stress in yeast. Therefore, S. pastorianus may experience PM stress and adapt to the self-forming environment during fermentation. However, how yeast adapts to PM stress remains unclear. Here, we investigated the temporal cellular responses of S. cerevisiae and S. pastorianus during adaptation to PM stresses by time-resolved mRNA-seq analysis. Our data showed different transcriptional phenotypes between S. cerevisiae and S. pastorianus during adaptation. The results may reflect the distinct nature of the two yeasts that have evolved in different nutritional environments. The dataset presented here would provide a promising resource for studying the characteristic nature of these differentially domesticated yeasts upon PM stresses.

Spore ultrastructure in Sporothrix schenckii.

Pathogenic strains of Sporothrix schenkii may show triangular spores, whose angular shape is maintained by a tie-beam effect in the inner cell wall structure. This difference in wall structure lies adjacent to a folded and possibly more active part of the spore cytoplasm. The supposed generation of asci in old cultures was simulated by the death of hyphae which are reinvaded by intrahyphal growth with intrahyphal spore production, while true asci were not seen.

Physical studies of cell surface and cell membrane structure. Determination of phospholipid head group organization by deuterium and phosphorus nuclear magnetic resonance spectroscopy.

Phospholipid head group conformations in 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), DPPC-cholesterol, and DPPE-cholesterol dispersions, in excess water above the pure lipid gel to liquid-crystal phase transition temperature, have been calculated by using comparisons between experimental 2H and 31 P NMR spectral parameters and theoretical results obtained from a plausible model of head group motions. The new calculations are compared with results obtained in previous studies [Seelig, J., Gally, H. U., & Wohlgemuth, R. (1977) Biochem, Biophys. Acta 467, 109--117; Brown, M. F. & Seelig, J. (1978) Biochemistry 17, 381--384; Seelig, J., & Gally, H. U. (1976) Biochemistry 15, 5199--5204] and are shown to agree qualitatively under certain highly restrictive conditions. Under more general conditions, it is shown that many possible solutions are generated but that these may often be separated into a small number of likely conformations in which the head group torsion angles are restricted to specific ranges rather than to a discrete set of values. There is no NMR evidence, however, to support the notion that there are only single conformational solutions to the NMR measurements for the above phospholipid systems.
