[Bonding to dentin].

Abstract

The development of adhesive systems is mainly concentrated on the possibility of chemical bond formation. Although glass-ionomer bonds chemically both to enamel and dentin and proved to be a successful restorative in class V restorations, the ultimate goal is to accomplish a strong and durable bond with the esthetic composite materials. The mechanism of chemical bonding to dentin apatite and collagen is discussed for a number of recently developed dentin bonding agents. Bond strengths up to 15 MPa can be obtained, but these are still not able to resist the forces of polymerization shrinkage of the composite materials.

Tissue factor and thrombomodulin expression on endothelial cell-seeded collagen modules for tissue engineering.

The creation of functional tissue engineering constructs to repair or replace diseased tissues requires a well-formed vasculature network within the construct and the endothelial cells lining that vascular bed must display a nonthrombogenic phenotype. A new approach to tissue engineering involves the assembly of smaller components (modules fabricated at the hundred micron scale) into larger constructs. The modules, collagen gel containing the particular tissue cell of interest, are covered with endothelial cells prior to assembly so that the interconnected channels that are formed are lined with endothelial cells, creating a mimic of a vascular network. Here, we confirmed (using confocal microscopy primarily) that the human umbilical vein endothelial cells, seeded on collagen gel modules without a second embedded cell and without flow, bore the molecular markers of low thrombogenicity. Two days, after seeding on the modules, endothelial cells displayed the typical cobblestone morphology, formed tight cell-cell junctions and covered the whole module surface. Immunofluorescence staining showed that at both 2 days and 7 days after seeding, only a few cells expressed tissue factor while this number was dramatically increased after TNFalpha stimulation. On the other hand, thrombomodulin was expressed by the majority of seeded cells and expression was reduced after TNFalpha stimulation.

Application of crosslinkers to dentin collagen enhances the ultimate tensile strength.

The stabilization of dentin collagen with biocompatible crosslinking agents may be of clinical importance to improve dentin bond strength. The present study aimed to evaluate the effect of three collagen crosslinking agents on the ultimate tensile strength (UTS) of undemineralized and demineralized dentin. Ten freshly extracted sound molars were sectioned into 0.5 x 0.5 mm2 thick beams. The beams were either demineralized or kept undemineralized. Then, specimens were subdivided into four groups according to treatments--PBS solution (control), 5% glutaraldehyde (GD), 0.5% proanthocyanidin PBS solution (PA), and 0.625% genipin PBS solution (GE). Specimens were kept in their respective solutions for either 4 or 40 h. To assess UTS, specimens were subjected to tensile forces at a crosshead speed of 1 mm/min. Statistical analysis was performed using two-way ANOVA and Fisher's PLSD test (p < 0.05). Statistically significant increases in UTS were observed for demineralized dentin after PA and GE dentin treatment, when compared with those of the control group. Dentin treated with GD showed no statistically significant differences in UTS when compared with that the control. Undemineralized dentin revealed no significant differences as compared to that of the control, regardless of the collagen crosslinkers. The application of two naturally occurring crosslinkers, i.e., PA and GE, to dentin collagen significantly improves UTS, indicating its potential value in restorative dentistry.

Using gold nanorods to probe cell-induced collagen deformation.

In biological tissue, complex mechanisms of cellular response are closely linked to the mechanical environment that cells experience. The key to understanding these mechanisms may lie in measurement of local mechanical fields near living cells and between cells. We have developed a novel optical measurement technique which combines the light elastically scattered from gold nanorods with digital image analysis to track local deformations that occur in vitro between cells, in real time, under darkfield optical microscopy. We find that measurable tension and compression exist in the intercellular matrix at the length scale of micrometers, as the cells assess, adapt, and rearrange their environment.
