Does streptomycin cause an error catastrophe?

Abstract

We have examined the interpretation that streptomycin kills a bacterial culture by initiating the so-called error catastrophe. In particular, we asked whether the increased translational error rate induced by the antibiotic gives rise to an autocatalytic loss of functional fidelity of the devices responsible for gene expression, which ultimately causes the death of the culture. We have analyzed the performance characteristics of one of these devices, namely the ribosome in streptomycin-treated bacteria. We find that, although the treated ribosomes are constructed from error-containing proteins, they are not significantly different in elongation rate and fidelity from those ribosomes taken from untreated bacteria. We conclude that the bacteriocidal effect of streptomycin is not due to the initiation of an error catastrophe.

Dihydrostreptomycin analogues with structural variations in their guanidino groups were prepared. The analogue with a methyl group on the guanidine at C-1 was nearly as active as dihydrostreptomycin against bacteria. However, the 2-imidazolin-2-ylamino substituent at C-1 eliminated activity. No analogue with a substituent on the C-3 guanidino group was active.

The spectrophotometric determination of streptomycins.

The spectra produced by the acid treatment and alkaline treatment of streptomycin and dihydrostreptomycin have been investigated to obtain suitable methods for their determination. Mixtures of the two antibiotics have been successfully determined in the UV region. Thus, using the orthogonal function method, dihydrostreptomycin has been determined after acid treatment whereas streptomycin has been determined after alkaline treatment followed by immediate neutralization.
