Spectroscopic studies on IgG aggregate formation.

Abstract

Spectroscopic techniques have been used to examine the effects of IgG of heating, irradiation by ultraviolet light and exposure to glutaraldehyde. Relatively few changes were observed in treated IgG which remained unaggregated but several significant size-dependent changes were observed in aggregated IgG. These results suggest that IgG aggregate formation by cross-linking with glutaraldehyde involves the least perturbation of the basis IgG structure, whereas aggregate formation by heating involves the most, with ultraviolet irradiation occupying an intermediate position.

Covalent coupling of asparagus pea and tomato lectins to poly(lactide) microspheres.

Lectin-poly(lactide) microsphere conjugates specifically designed for oral administration were prepared and their activity and specificity in presence of mucus were characterized. The presence of hydroxyl or amino groups suitable for covalent coupling of lectins by the glutaraldehyde method at the surface of the microspheres have been ensured by preparing the particles in presence either of poly(vinyl alcohol) (PVA) or bovine serum albumin (BSA). Tomato and asparagus pea lectins could be covalently attached to these particles (1.0-1.3 mg/m(2) of particles). The conjugates demonstrated a 4-10 fold increase in their interactions with mucus compared to control particles. Moreover, the sugar specificity of the lectins was maintained.

Aneurysmal dilation of a pericardial patch prepared with glutharaldehyde and used for closure of a ventricular septal defect.

Pericardium patches are commonly used for the repair of congenital heart diseases. Aneurysmal dilation is a complication specific to the use of pericardial patches. Preparation of the pericardium with glutharaldehyde is considered to avoid this risk. In the present case report, we describe the development of a giant aneurysm of a heterologous patch used for closure of a ventricular septal defect in a child aged 14 days.
